用UV-Vis吸收光谱、荧光光谱、圆二色谱以及核磁共振光谱等手段研究了硫堇(TH)与两个不同序列寡核苷酸的作用。TH与寡核苷酸作用后的吸收光谱和荧光光谱产生了明显的减色红移和荧光猝灭效应。分别计算了TH与[oligo d(GC)]2和[oligo d(AT)]2作用的荧光猝灭常数和结合常数,结果表明TH与GC序列的结合能力比与AT序列更强。通过TH与[oligo d(GC)]2作用后双螺旋链构象变化以及TH质子的1HNMR谱峰明显变宽,进一步说明TH与寡核苷酸结合的序列选择性。  相似文献

Twenty pure elemental metal samples have been studied with a coincidence Doppler broadening system (CDB). The results show the relationship between the CDB spectra and the electronic structure of these samples. The experimental results are compared to simple theoretical predictions, which show that the high-momentum part of the Doppler-broadening spectra can be used to distinguish different elements.  相似文献

The relationship between the redox reaction of camphor‐induced cytochrome P‐450 (P‐450cam) and its activity was measured by using cyclic voltammetry. The redox potential of P‐450cam solution shifted to the lower side of the potential by binding of substrate, and the change was proportional to the amount of the substrate binding to the protein. The substrate binding was inhibited at the low concentration of oxygen in the reaction solution. The reaction product, hydroxycamphor, was observed in the reaction mixture by gas chromatography/mass spectroscopy. However, hydroxycamphor was not observed at an oxygen concentration of about a tenth part of the saturated one. The shift of redox potential of P‐450cam solution corresponded to the substrate specificity of the activity. These results suggest that the redox reaction of P‐450cam related to the substrate‐binding to the protein and its activity. Furthermore, the present system was very simple and speedy for the measurement of the activity. Copyright © 1999 John Wiley & Sons, Ltd.  相似文献

The substrate specificity of a camphor-induced cytochrome P-450 (P-450cam) was measured by using a new assay system: electrochemical control of P-450cam activity by protein immobilization on an electrode. Immobilized P-450cam showed the obvious substrate specificity for hydroxylation of the substrate, suggesting that the simple assay system is applicable for the study of the effect of the other components of the electron transfer system on activity. Recently, G-quadruplex/hemin (G4/hemin) complexes have been found to exhibit peroxidase activity, and this feature has been extensively exploited for colorimetric detection of various targets. To further understand and characterize this important DNAzyme, its substrate specificity, inactivation mechanism, and kinetics have been examined by comparison with horseradish peroxidase (HRP). G4/hemin DNAzyme exhibits broader substrate specificity and much higher inactivation rate than HRP because of the exposure of the catalytic hemin center. The inactivation of G4/hemin DNAzyme is mainly attributed to the degradation of hemin by H(2)O(2) rather than the destruction of G4. Both the inactivation rate and catalytic oxidation rate of G4/hemin DNAzyme depend on the concentration of H(2)O(2), which suggests that active intermediates formed by G4/hemin and H(2)O(2) are the branch point of catalysis and inactivation. Reducing substrates greatly inhibit the inactivation of G4/hemin DNAzyme by rapidly reacting with the active intermediates. A possible catalytic and inactivation process of G4/hemin has been proposed. These results imply a potential cause for the hemin-mediated cellular injury and provide insightful information for the future application of G4/hemin DNAzyme.  相似文献

(1) Sesame oil aroma has stress-relieving properties, but there is little information on its effective use and active ingredients. (2) Methods: ICR male mice were housed under water-immersion stress for 24 h. Then, the scent of sesame oil or a typical ingredient was inhaled to the stress groups for 30, 60, or 90 min. We investigated the effects of sesame oil aroma on mice behavior and the expression of the dual specificity phosphatase 1 (DUSP1) gene, a candidate stress marker gene in the brain. (3) Results: In an elevated plus-maze test, the rate of entering into the open arm of a maze and the staying time were increased to a maximum after 60 min of inhalation, but these effects decreased 90 min after inhalation. As for the single component, anxiolytic effects were observed in the 2,5-dimethylpyrazine and 2-methoxy phenol group, but the effect was weakened in the furfuryl mercaptan group. The expression levels of DUSP1 in the hippocampus and striatum were significantly decreased in 2,5-dimethylpyrazine and 2-methoxy phenol groups. (4) Conclusions: We clarified the active ingredients and optimal concentrations of sesame oil for its sedative effect. In particular, 2,5-dimethylpyrazine and 2-methoxy phenol significantly suppressed the stress-induced changes in the expression of DUSP1, which are strong anti-stress agents. Our results suggest that these molecules may be powerful anti-stress agents.  相似文献

The discovery and characterization of molecular interactions is crucial towards a better understanding of complex biological processes. Particularly protein-protein interactions (i.e., PPIs), which are responsible for a variety of cellular functions from intracellular signaling to enzyme-substrate specificity, have been studied broadly over the past decades. Position-specific scoring matrices (PSSM) in particular are used extensively to help determine interaction specificity or candidate interaction motifs at the residue level. However, not all studies successfully report their results as a candidate interaction motif. In many cases, this may be due to a lack of suitable tools for simple analysis and motif generation. Peptide Specificity Analyst (PeSA) was developed with the goal of filling this information gap and providing an easy to use software to aid peptide array analysis and motif generation. PeSA utilizes two models of motif creation: (1) frequency-based using a user-defined peptide list, and (2) weight-based using experimental binding results. The ability to produce motifs effortlessly will make studying, interpreting and disseminating peptide specificity results in an effortless and straightforward process.  相似文献

研究氢抽取反应OH+H2S对于理解酸雨形成、空气污染和气候变化的原因具有重要意义. 本文在降维模型下使用量子含时波包方法研究了OH+H2S→H2O+SH反应的动力学行为. 研究表明,该反应在低碰撞能时表现出无垒反应的特征,而在高碰撞能下表现出具有显著势垒的激活反应的特征. 激发反应物H2S分子的对称或反对称伸缩模式比激发弯曲模式更有效地促进了反应,该动力学特征可以通过各正则模式与反应坐标的耦合强度来解释. 此外,模式指定的反应速率常数表现出明显的非阿伦尼乌斯温度依赖性.  相似文献

An HPLC-based method has been developed for composition analysis of six positional isomers of phosphatidylinositol (PI), of which the phosphatidyl group was connected to different positions of the myo-inositol moiety. The method employed a combination of two types of HPLC analyses. One was direct separation of the six PI isomers into four peaks of 1(3)-PI, 2-PI, 4(6)-PI and 5-PI on a normal-phase silica gel column. The second method was for the separations of 1-PI from 3-PI and 4-PI from 6-PI, which were not separable on the normal-phase column. This method involved conversion of PI isomers into pentakis-(R)-1-phenylethylcarbamate (PEC) derivatives, which were separated on a reversed-phase column. Using the established method, positional specificity of several engineered phospholipases D in enzymatic synthesis of PI from myo-inositol and phosphatidylcholine was investigated. This was performed by analyzing the isomeric composition of PIs synthesized by the mutant enzymes. Among five mutant enzymes tested, two showed strong specificity to 1-OH, one showed moderate preference to 1-OH, one preferred 3-OH, and one showed broad specificity towards 1-, 3-, 4- and 6-OH.  相似文献
