Synthesis and Properties of 6-Amino-7-hetaryl-5-R-5H-pyrrolo[2,3-b]pyrazine-2,3-dicarbonitriles

We have established that when 5-chloro-6-[cyano(2,3-dihydro-1-R-benzo[d]azol-2-yl)methyl]-2,3-pyrazinedicarbonitriles are reacted with nucleophilic reagents (aliphatic and aromatic amines, hydrogen sulfide), annelation of the five-membered ring occurs on the [b] face of the pyrazine with formation of 6-amino-7-hetaryl-5-R-5H-pyrrolo[2,3-b]pyrazine-2,3-dicarbonitriles and 6-amino-7-(1H-benzo[d]imidazol-2-yl)thieno[2,3-b]pyrazine-2,3-dicarbonitrile respectively. Further heating with excess of acylating reagent leads to formation of a novel heterocyclic system 1H-benzo[4,5]imidazo[1,2-c]pyrazino[2',3':4,5]pyrrolo[3,2-e]pyrimidine. Reaction of vicinal dinitriles with hydrazine hydrate leads to the novel system 1H-pyrrolo[2',3':5,6]pyrazino[2,3-d]pyridazine.     

A general route for the synthesis of pyrimido[4′,5′:4,5]thieno[2,3-c]pyridazine derivatives

Summary A facile synthesis of 8-substituted pyrimido[4,5:4,5]thieno[2,3-c]pyridazines (6a–i) has been accomplished. The sequence involves the ring closure of a heterocyclic aminonitrile precursor (3) after reaction with (dichloromethylene)-dimethylammonium chloride.

Eine allgemeine Synthese von Pyrimido[4,5:4,5]thieno[2,3-c]pyridazin-Derivaten

Zusammenfassung Es wurde ein einfacher Syntheseweg für 8-substituierte Pyrimido[4,5:4,5]thieno-[2,3-c]pyridazine entwickelt. Die Reaktion verläuft über den Ringschluß eines heterocyclischen Aminonitrilvorläufers (3) nach Umsetzung mit Dichlormethylen-dimethylammoniumchlorid.

     

3-炔基咪唑并[1,2-b]哒嗪的简便合成

以咪唑并[1,2-b]哒嗪为原料,与N-碘代丁二酰亚胺在三氟乙酸/二氯甲烷中于室温进行碘代反应制得3-碘咪唑并[1,2-b]哒嗪(3);3与三甲基硅基乙炔在Pd(OAc)_2催化下经Sonogashira偶联反应制得3-三甲基硅基乙炔基咪唑并[1,2-b]哒嗪(4);4在碳酸钾作用下脱除三甲基硅基合成了3-炔基咪唑并[1,2-b]哒嗪,总收率61.2%,其结构经~1H NMR和HR-MS确证。     

Preparation of novel pyridine-fused tris-heterocycles; pyrido[4,3-e]pyrrolo-/pyrido[4,3-e]furano[2,3-c]pyridazines and pyrido[3,4-b]pyrrolo[3,2-d]pyrrole

Three novel pyrido-fused tris-heterocycles have been prepared based on a Suzuki coupling and subsequent cyclisation approach. Pyrido[4,3-e]pyrrolo[2,3-c]pyridazine (3b, 77%) and pyrido[4,3-e]furano[2,3-c]pyridazine (5b, 76%) were obtained by intramolecular diazocoupling. Successful diazocoupling of furan (5b) is thus reported for the first time by NOBF₄ generation of the diazonium intermediate. N-TIPS-pyrido[3,4-b]pyrrolo[3,2-d]pyrrole (TIPS-4b) was synthesised by thermal cyclisation of pyridyl nitrene in considerably higher yield (71%) than previously experienced from similar cyclisations, due to TIPS-activation.     

手性方酰化哒嗪胺及铜(II)配合物的荧光性研究

以3-甲基-6-氯哒嗪和α-氨基丙醇、方酸为原料合成了一种新的手性方酰化哒嗪胺衍生物和铜(II)配合物, 新化合物和铜(II)配合物结构用IR, ¹H NMR及元素分析对其表征. 并对新化合物的铜(II)配合物的荧光性进行了研究.     

X-ray crystal and molecular structure of [3,6-bis(2-pyridyl)pyridazine-N 1,N2]bis(triphenylphosphine) copper(I) hexafluorophosphate

[3,6-bis(2-pyridyl)pyridazine-N ¹,N²]bis(triphenylphosphine) copper(I) hexafluorophosphate crystallizes in space group $P\bar 1$ with cell dimensionaa=14.050(2),b=11.941(2),c=15.200(2) Å, α=71.28(1), β=67.02(1), and γ=84.78(2)^o. In the title compound, the mononuclear cation consists of a tetrahedral copper(I) center involving two nitrogen donors of the 3,6-bis(2-pyridyl)pyridazine (dppn) and two P atoms of the triphenylphosphine molecules. In the dppn ligand the two non-coordinating N atoms (one from pyridine and one from pyridazine) are mutallytrans.     

3-芳氧基-6-取代哒嗪的合成及其除草活性

合成了系列3-芳氧基-6-取代哒嗪类化合物.化合物结构经¹HNMR、元素分析、IR和MS确证.生物活性测试结果表明,该类化合物具有很好的除草活性,讨论了其结构与除草活性的关系.     

合成金鸡纳生物碱衍生物的简便方法

以CaH2为缚酸剂,DMF为溶剂,金鸡纳生物碱通过3,6-二氯哒嗪或1,4-二氯-2,3-二氮杂萘桥连,高产率地合成了用作手性配体的13个金鸡纳生物碱衍生物,其结构经NMR确证.     

Reaction of N‐Nonaflyl and N‐Cyano‐Benzotriazoles with Enamines

N‐Nonaflyl‐benzotriazole 1a reacts with enamines 2 in tetrahydrofurane (THF) at room temperature to afford o‐nonafluorobutansulfonamido‐phenylazo‐enamines 3 in 74–81% yield. Compound 1a reacts with 1‐diethylaminobutadien 2f twice, affording pyridazine derivative 3f in 20% yield. Ringopening of N‐cyano‐benzotriazole 1b with pyrrolidinocyclohexene 2a affords, under cleavage of pyrrolidine 1,2,3,4‐tetrahydro‐dibenzo[4,5:e]imidazo[1,2‐b][1,2,4]triazine 4 in 43% yield.     

含胆甾基的双取代哒嗪的合成和介晶性

合成了8个未见报道的中心桥连基为哒嗪环、酯基、CH＝N基,含有胆甾基、二个苯环、不同末端链长度的双取代哒嗪化合物,并通过示差扫描量热法(DSC)对其介晶性进行了表征.研究表明,末端链长度对相变温度和清亮点温度均有影响,但对相变温度范围影响较小.
