β-Lactoglobulin-carboxymethyl cyclodextrin (β-LG-CMCyD) conjugates were prepared by using water soluble carbodiimide. Three kinds of CMCyDs differing in molecular mass were used to investigate the effects of different CMCyD contents, net charge and hydrophobicity on the structural changes in β-lactoglobulin. The effect of CMCyDs on the structure of β-lactoglobulin was utilized to investigate the contribution of hydrophobic interactions to the stability of the protein. Spectroscopic studies suggested that the conformation around had not changed in either conjugate but the α-helix content of β-LG-CMCyD conjugates had markedly increased as compared with that of β-lactoglobulin. The differential scanning calorimetry technique confirmed that the addition of one glucose unit in β-LG-CMCyD conjugates, enthalpy change of calorimetry decreased and the denaturation temperature of each conjugate was higher than that of native β-lactoglobulin. The heat contents agreed well with the conformational transition measured by molar ellipticity at 222 nm ([θ]222) and Stoke's radius (RS) values. Therefore, hydrophobic forces play an important role in stabilizing and shielding of the β-LG-CMCyD conjugates.  相似文献

In this work, the suitability of a new polymer family has been investigated as capillary coatings for the analysis of peptides and basic proteins by CE. This polymer family has been designed to minimize or completely prevent protein–capillary wall interactions and to modify the EOF. These coating materials are linear polymeric chains bearing as side cationizable moiety a dentronic triamine derived from N,N,N’,N’‐tetraethyldiethylenetriamine (TEDETA), which is linked to the backbone through a spacer (unit labeled as TEDETAMA). Four different polymers have been prepared and evaluated: a homopolymer which comprised only of those cationizable repetitive units of TEDETAMA, and three copolymers that randomly incorporate TEDETAMA together with neutral hydrosoluble units of N‐(2‐hydroxypropyl) methacrylamide (HPMA) at different molar percentages (25:75, 50:50 and 75:25). It has been demonstrated that the composition of the copolymers influences the EOF and therefore the separation of the investigated biopolymers. Among the novel polymers studied, poly‐(TEDETAMA‐co‐HPMA) 50:50 copolymer was successfully applied as coating material of the inner capillary surface in CE‐UV and CE‐MS, providing EOF reversing together with fast and efficient baseline separation of peptides and basic proteins. Finally, the feasibility of the polymer‐coated capillary was shown through the analysis of lysozyme in a cheese sample.  相似文献

以1,3-双[3-(1-甲氧基-2-羟基丙氧基)丙基]四甲基二硅氧烷和八甲基环四硅氧烷(D4)为原料,通过阳离子催化开环聚合制备了1,3-双(3-(1-甲氧基-2-羟基丙氧基)丙基)封端聚硅氧烷,研究了反应温度,反应时间,催化剂种类及加入量对于聚合反应的影响,结果表明,最佳反应条件为:反应温度65℃,反应时间24h,浓硫酸作为催化剂加入量为反应物质量的0.3%,此时反应拥有最高的转化率。通过红外光谱与核磁共振光谱对产物进行了表征。  相似文献

Through a well-established enzymatic transformation glucosinolates release reactive isothiocyanates that can undergo further metabolic pathways affording a plethora of reactive metabolites. This study explores in detail the reaction of benzyl isothiocyanate, which possesses antitumor activity as alkylating agent, with d-glucosamine, commonly employed in oral treatments against osteoarthritis and inflammation. Structures of the resulting products and their evolution have been assessed and compared with those involving d-glucose, reported previously. Chemical results suggest that clinical treatments with d-glucosamine could reduce the beneficial effects associated with diets based on glucosinolate-rich foods.  相似文献

This work presents a structural and vibrational theoretical study of n‐propyl cyanide as a function of the nitrile and methyl torsional modes. A potential energy hypersurface is built at the MP4(SDQ)/aug‐cc‐pVTZ//MP2/aug‐cc‐pVTZ theory level. The equilibrium structure is found in a gauche conformation. Another minimum is found for the trans form. The maximum appears in a cis conformation. For the first time, the interconversion barriers between the different forms are calculated. A two‐dimensional anharmonic vibrational Hamiltonian is built for the nitrile and methyl torsional modes. We find the vibrational energy levels to organize in two stacks associated to the gauche and trans forms. Fundamental frequencies of 113.12 and 220.54 cm?1 are predicted for the nitrile and methyl torsions in the equilibrium, gauche, conformer. In addition, we find symmetry allowed transitions between the gauche and trans energy levels stacks. The lowest transition is predicted to appear at 24.49 cm?1. © 2010 Wiley Periodicals, Inc. Phenethyl isothiocyanate (PEITC) is a promising chemopreventive agent present in cruciferous vegetables. This paper describes the development of a method for the determination of PEITC in human plasma by liquid chromatography/tandem mass spectrometry (LC‐MS/MS). Atmospheric‐pressure chemical ionization was found more suitable for ionization of PEITC than electrospray ionization. Because of the lability of PEITC, a combination of low temperature and acidification was applied to minimize the degradation during the sample collection and preparation procedure. A simple protein precipitation with acetonitrile was used for the preparation of plasma samples. The analyte and 1‐phenylpropyl isothiocyanate as internal standard (IS) were subjected to chromatographic analysis on a C18 column (50 × 2.1 mm, 5 µm) using 85% methanol as mobile phase at a flow rate of 0.3 mL/min. The total analysis time for each chromatograph was 3 min and the results were linear over the studied range (5.00–250 ng/mL). The intra‐ and inter‐day precision values were acceptable as per US Food and Drug Administration guidelines. We have disclosed an efficient, one pot, novel multi-component approach for the synthesis of 5-aryl-[1,2,4]triazolidine-3-thiones from aldehyde, hydrazine hydrate, and trimethylsilyl isothiocyanate (TMSNCS) in the presence of catalytic amount of sulfamic acid. High yields, easy work-up procedure, no chromatographic separation, and novelty in multi-component strategy are the main merits of the present strategy.  相似文献

An efficient and facile protocol for the synthesis of azine and azole ring systems was reported. Whereas, reaction of cinnamoyl isothiocyanate with N-nucleophile containing compounds (namely, p-aminophenol (2), N1-phenylbenzene-1,4-diamine (5) and p-aminoacetophenone (8)) tolerated thiourea derivatives 3, 6, and 9, respectively. The later compounds underwent intramolecular cyclization upon treatment with EtONa to give pyrimidinethiones 4, 7, and 10, respectively, in moderate yield (74–79%). Compound 9 underwent intramolecular cyclization and condensation upon reaction with NaOH and benzaldehyde to give pyrimidinethione 12. Thiosemicarbazides 14 and 19 were obtained through reaction of heteroallen 1 with 2,4-dinitrophenylhydrazine 13 and hydrazone 18, respectively. Compound 14 was cyclized to pyrimidinethione 15 and triazine derivatives 17 through its reaction with EtONa at room temperature and refluxing temperature, respectively. Finally, base mediated and oxidative cyclization of thiourea derivative 19 with EtONa, Br2/AcOH, and Pb(OAc)2 afforded thiadiazole 20, benzothiazolotriazole 21, and triazolethione 22 derivatives, respectively.  相似文献
