Triazolo‐fused 2′,3′‐cyclic nucleoside analogs were synthesized by an intramolecular 1,3‐dipolar cycloaddition of nucleoside‐derived azido alkynes in a regio‐ and stereospecific manner. The uracil base in these target compounds was successfully transformed to the corresponding cytosine. The synthesized compounds were examined in a MAGI assay for their anti‐HIV activities, and in a H9 T lymphocytes assay for their cell toxicities. 相似文献

Synthesis of eight nucleoside analogues 4–11 with a methylenecyclobutane unit is described. Wittig reaction with 2‐hydroxymethylcyclobutanone 12 gave a mixture of Z (13) and E (14) derivatives, which was separated before functional modifications. The heterocyclic moieties were introduced via a Mitsunobu reaction either on the saturated chain or on the unsaturated chain. When adenine was used in this reaction, only the N‐9 substitution products were obtained. Removal of the protecting groups provided the target products. 相似文献

Two porous metal–organic frameworks (MOFs), [Zn3(L)(H2O)2] ? 3 DMF ? 7 H2O ( MOF‐1 ) and [(CH3)2NH2]6[Ni3(L)2(H2O)6] ? 3 DMF ? 15 H2O ( MOF‐2 ), were synthesized solvothermally (H6L=1,2,3,4,5,6‐hexakis(3‐carboxyphenyloxymethylene)benzene). In MOF ‐ 1 , neighboring ZnII trimers are linked by the backbones of L ligands to form a fascinating 3D six‐connected framework with the point symbol (412.63) (412.63). In MOF‐2 , eight L ligands bridge six NiII atoms to generate a rhombic‐dodecahedral [Ni6L8] cage. Each cage is surrounded by eight adjacent ones through sharing of carboxylate groups to yield an unusual 3D porous framework. Benzofurazane has been attached to nucleosides and dNTPs, either directly or through an acetylene linker, as a new redox label for electrochemical analysis of nucleotide sequences. Primer extension incorporation of the benzofurazane‐modified dNTPs by polymerases has been developed for the construction of labeled oligonucleotide probes. In combination with nitrophenyl and aminophenyl labels, we have successfully developed a three‐potential coding of DNA bases and have explored the relevant electrochemical potentials. The combination of benzofurazane and nitrophenyl reducible labels has proved to be excellent for ratiometric analysis of nucleotide sequences and is suitable for bioanalytical applications. 相似文献

An efficient synthesis of 2‐substituted benzoxazoles through RuCl3·3H2O catalyzed, air oxidized tandem reactions of 2‐aminophenols and aldehydes in [bmim]BF4 was developed. This synthetic strategy has such advantages as mild reaction conditions, cost‐free oxidant, readily available starting materials, and recyclable catalyst and solvent. As an application, it was successfully used in the synthesis of the unreported 5‐(benzoxazol‐2‐yl)‐2′‐deoxyuridines with potential biological activities. 相似文献

[Perfluoro-organic]/[silicon oxide] hybrids were formed by conducting sol-gel reactions of tetraethylorthosilicate within a perfluoro(carboxylate/sulfonate) bilayer membrane in the Co+2 form. FTIR and 29Si solid-state NMR spectroscopies were used to probe general aspects of molecular structure within the silicon oxide phase as a function of its relative content. The internal gel structure is considerably unconnected in terms of the population of Si O Si groups in cyclic vs. linear substructures and degree of Si atom coordination about bonded SiO4 units. In situ (HO)xSiO2[1-1/4x] intrastructure become increasingly less connected and more strained with regard to bonding geometry with increasing percent silicon oxide. Structural differences are seen between the silicon oxide component incorporated in carboxylate and sulfonate layers. These inorganically modified perfluorinated ionomers have potential as fast-proton conducting membranes for fuel cells and as permselective membranes in liquid pervaporation cells. © 1998 John Wiley & Sons, Inc. Abstract Pyrroline 5-carboxylate, a potent effector of redox dependent pathways, is a nutritionally responsive plasma constituent and is present in tissue culture medium conditioned by a variety of cells. Studies of P5C production and release both in vivo and in vitro have become increasingly important. To date, the assay used is a specific, sensitive radioenzymatic method. Unfortunately, the necessary materials (purified P5C reductase and NADP[3H], labeled in the transferable position) are not readily available. We now describe a method using commercially available reactants. This new method, based on the interaction of P5C with cysteine, is sensitive and specific; the presence of a wide variety of compounds only minimally affected the assay. [S35] Cysteine at a defined specific activity is reacted with P5C and the P5C-CYS adduct can be separated from the reactants by cation exchange column chromatography. To optimize the assay conditions, we characterized the time course, pH dependence and cysteine concentrations for adduct formation. Studies showed that the sulfhydryl group of cysteine is critical in the formation of the P5C-CYS adduct. Comparison of this new method to the previously published radioenzymatic method showed close correlation over a wide range of P5C concentrations in conditioned media. 相似文献
