Carotenogenesis of the lactose-negative yeast Rhodotorula rubra GED5 was studied by cocultivation with Kluyveromyces lactis MP11 in whey ultrafiltrate (WU) (35, 50, and 70 g of lactose/L). Maximum yields of cell mass (24.3 g/L) and carotenoids (10.2 mg/L of culture fluid or 0.421 μ g/g of dry cells) were obtained by growing the microbial association in WU (50 g of lactose/L) in a fermentor with an airflow rate of 0.8 L/(L·min), agitation of 220 rpm, and temperature of 30°C. The identified carotenoid pigments—β-carotene, torulene, and torularhodin—reached maximum concentrations (133, 26.9, and 222.3 μg/g of dry cells, respectively) on d 5 for torulene and d 6 for β-carotene and torularhodin.  相似文献

The effect of ammonium nitrate concentration in the citric acid biosynthesis by Aspergillus niger NC-12 in single-stage continuous cultures with biomass retention was investigated. Experiments were carried out in a BIOMER laboratory fermenter with 5 dm3 working volume. At the initial stage of each cultivation, the substrate in the bioreactor contained 1.5 g NH4NO3 dm−3. After 120 h onwards, the bioreactor was fed continuously at a constant dilution rate of 0.009 h−1. NH4NO3 concentration in the feed was varied from one culture to another, ranging between 0.5 g dm−3 and 2.5 g dm−3. Promising results were obtained when NH4NO3 concentration of 1.5 g dm−3 was used. The observed concentration of citric acid (c P) and yield of citric acid with respect to the introduced sucrose (Y P/S) were 117.88 g dm−3 and 78.59 %, respectively. The efficiency coefficient of citric acid biosynthesis (K ef) was very high, amounting to 83.38. 构建微生物细胞工厂是化学品、生物能源以及药物分子可持续生产的可行性策略。然而,微生物的代谢复杂、调控严谨,制约着目标产物高效合成。蛋白质组学和代谢组学可以从系统生物学角度分析酶和代谢物组分,从而理解复杂的生物系统,为微生物代谢工程改造提供重要线索。该文介绍了蛋白质组学和代谢组学在微生物代谢工程中的应用,包括基因组尺度代谢模型构建、菌株生物合成优化、指导菌株耐受性改造、限速步骤预测、植物次级代谢途径挖掘,从而为微生物合成天然产物提供新的基因或途径。在此基础上,该文还展望了生物大数据未来的发展方向。  相似文献

Polyamide composed of furan dimer, which is prepared from biomass‐derived organic molecule 2‐furfural, is synthesized. The reaction of 2,2′‐furan dimer 5,5′‐dicarbonyl chloride with several 1,ω‐diamines was carried out with a solution or interfacial polycondensation leading to the corresponding polyamide. Measurement of the melting point was performed resulting to exhibit a higher temperature compared with the related polyamide bearing a single furan ring composed of furan‐2,5‐dicarboxylic acid (FDCA). The main objective of this study was to evaluate the sound absorption properties of rigid polyurethane foams (PUFs) produced from crude glycerol (CG) and/or liquefied coffee grounds derived polyol (POL). The lignin content of POL proved to have a major influence on the structure and mechanical properties of the foams. Indeed, the POL content increased the cell size of the foams and their stiffness, which subsequently influenced the sound absorption coefficients. The POL derived foam has slightly higher sound absorption coefficient values at lower frequencies, while the CG foam has higher sound absorption coefficient values at higher frequencies. In turn, the foam prepared using a 50/50 mixture of polyols presents slightly higher sound absorption coefficient values in the medium frequencies range due to a balance between the cell structure and the mechanical properties. The results obtained seem to suggest that the mechanisms involved in sound wave absorption depend on the formulation used to prepare the foams. Additionally higher POL contents improved the thermal stability of PUFs as well as their mechanical properties. From this work the suitability of CG and/or POL derived PUFs as sound absorbing materials has been proven.  相似文献

A simple, cost-effective strategy was developed to effectively improve the electron transfer efficiency as well as the power output of microbial fuel cells (MFCs) by decorating the commercial carbon paper (CP) anode with an advanced Mo2C/reduced graphene oxide (Mo2C/RGO) composite. Benefiting from the synergistic effects of the superior electrocatalytic activity of Mo2C, the high surface area, and prominent conductivity of RGO, the MFC equipped with this Mo2C/RGO composite yielded a remarkable output power density of 1747±37.6 mW m−2, which was considerably higher than that of CP-MFC (926.8±6.3 mW m−2). Importantly, the composite also facilitated the formation of 3D hybrid biofilm and could effectively improve the bacteria–electrode interaction. These features resulted in an enhanced coulombic efficiency up 13.2 %, nearly one order of magnitude higher than that of the CP (1.2 %).  相似文献

Lignin is an important source of synthetic materials because of its abundance in nature, low cost, stable supply, and no competition to the human food supply. Lignin, a cross‐linked phenolic polymer, contains a large number of aromatic groups that can be used as a substitute for petroleum‐based aromatic fine chemicals. However, modification of lignin is necessary for its application in advanced materials due to its chemically inert nature and structural complexity. Polymeric modification of lignin via graft copolymerization represents an important avenue for modification because this method forms stable covalent bond linkages between lignin and synthetic functional polymers. In this review, we discuss recent synthetic strategies toward polymeric modification of lignin using graft copolymerization and the special properties and applications of the produced lignin copolymers. © 2017 Wiley Periodicals, Inc. 以废弃汽车外轮胎热解后的副产物轮胎热解焦(Tyre pyrolysis char,TPC)为原料,利用均匀沉淀法制备以轮胎焦为载体的负载型Ni/TPC催化剂,采用EDX、SEM、XRD、TG、BET手段对催化剂进行了表征与分析,同时使用管式炉测试了Ni/TPC催化剂在秸秆热解燃气重整中的催化性能,并考察了热解温度、保温时间、镍负载量及催化时间对秸秆热解燃气重整效果的影响。研究结果表明,TPC富含焦和金属,Ni/TPC催化剂分散均匀,热稳定性好,比表面积为62 m2/g。催化剂活性测试显示,Ni/TPC催化剂用于作物秸秆热解燃气重整具有很强的催化活性,可显著提高燃气中可燃气体含量;热解温度在750℃、保温时间10 min、30%的Ni负载量时Ni/TPC催化剂的催化效率最高,连续使用850 min后,燃气中的H2含量仍相对提高到50%以上,长时间使用后活性结构由Ni3ZnC0.7转变成FeNi3,催化活性依然较强且趋于稳定,TPC可以作为良好的新型镍基催化剂载体。  相似文献

Abstract

A microbial electrode has been developed that uses im -mobilized Nocardia erythropolis for the determination of cholesterol. The relationship between the measuring signal and the cholesterol concentration was linear up from T5-130 um, with a reproducibility of 2 – 7 %. To shorten the measuring time the derivative of the current – time curve was monitored. The resulting response time was 35 – 70 sec. The sensor was usuable for more than 4 weeks.  相似文献
