Summary Reversed (RP-HPLC) and normal phase chromatographic (NP-HPLC) separations have been developed for diastereomers ofN-acyl-1-methyl-1,2,3,4-tetrahydo-β-carbolines which are acylated derivatives of simple natural β-carboline alkaloids. Separations of derivatives having different acyl moieties in theO,O-diacyl-tartaric acid ester subtituent differed remarkably. Little or no resolution in either NP-HPLC or RP-HPLC could be achieved with the diacetyl-tartrate derivative. Base-line separation by RP-HPLC but no separation by NP-HPLC was possible with the bulkier and more apolar dipivaloyl derivative. Retention order of the bis-benzoylated diastereomers was reversed and separation time increased dramatically by RP-HPLC. Good separation of the medium polarity, but rigid,N-camphanyl derivative by NP-HPLC has been achieved, whereas RP-HPLC could not be used for separation of these diastereomers. Separability of different diastereomers was highly dependent on polarity and rigidity of the derivatizingN-acyl moieties. Conformational analysis by molecular mechanics and comparison of the lowest energy conformational states of the diastereomers was applied to rationalise separation-retention behaviour of stereoisomers by RP-HPLC. A rapid, simple and rugged procedure without requiring any prior sample treatment was developed for the determination of acetylsalicylic acid (ASA) in tablets formulations by solid-phase fluorescence spectroscopy. The method was carried out on powdered samples, consisting of an active substance dispersed in lactose, maize starch, talc and magnesium stearate. Previous knowledge of the sample bulk composition is needed for proper application of the method. Wavelengths for maximum excitation and emission were 288 and 318 nm, respectively, and the fluorescence intensity was linear with ASA concentration within the 50-170 mg g−1 range. Detection and quantification limits were 2.2 and 7.3 mg g−1, and the analytical frequency was 200 h−1. For a typical sample, the relative standard deviation of results was estimated as 2.3% (n = 10). Accuracy was assessed by comparing the analytical results obtained with the proposed method with those related to a reference method recommended by British Pharmacopoeia: no differences between the methods were found at the 95% confidence level.  相似文献

Two spectrophotometric methods for the determination of Ethinylestradiol (ETE) and Levonorgestrel (LEV) by using the multivariate calibration technique of partial least square (PLS) and principal component regression (PCR) are presented. In this study the PLS and PCR are successfully applied to quantify both hormones using the information contained in the absorption spectra of appropriate solutions. In order to do this, a calibration set of standard samples composed of different mixtures of both compounds has been designed. The results found by application of the PLS and PCR methods to the simultaneous determination of mixtures, containing 4–11 μg ml−1 of ETE and 2–23 μg ml−1 of LEV, are reported. Five different oral contraceptives were analyzed and the results were very similar to that obtained by a reference liquid Chromatographic method.  相似文献

An efficient method was developed for the asymmetric synthesis of (R)-S-(1,2,4-triazol-3-yl)cysteines by the addition of 3,4-disubstituted 1,2,4-triazole-5-thiols at the electrophilic C=C bond in a NiII complex of a Schiff base of dehydroalanine with (S)-N-(N-benzylprolyl)aminobenzophenone. The stereoselectivity of the formation of diastereomeric complexes with the (S,R) configuration under conditions of thermodynamic control of the nucleophilic addition exceeds 94%. Acid treatment of the reaction mixtures afforded enantiomerically pure (R)-S-hetarylcysteines (ee >98%).  相似文献

Using a mass spectrometric sampling method, we have observed the decomposition of CH4 in an rf plasma usedfor diamond deposition. The gas samples were extracted through an orifice located downstream of the plasma zone and analyzed online. For the experiments a dilute mixture of H2 and CH4 containing 0.1–3% CH4 has been used. CH4 is converted to C2H2 and C2H4 quantitatively. Small amounts of heavier hydrocarbons are formed. A comparison of the experimental results with a recent kinetic model treating a purely thermal environment is made and the differences between our experiment and the model are explained. The role of acetylene as a species formed in an atmosphere rich in atomic hydrogen is proposed. The electron impact dissociation process is suggested as the rare-determining step in the plasma-chemical decomposition of methane.  相似文献

Biocatalysis and, to a lesser extent, biorecognition in non-aqueous media (including organic solvents as well as supercritical fluids and gases) constitute at present an exciting research area which has already demonstrated its biotechnological potential in numerous, varied applications. Less attention, however, has been paid to its analytical possibilities, even though many advantages have been postulated and a wide range of poorly water-soluble analytes are present in samples (or waste materials) from food and drink, petrochemical, pharmaceutical, military and other industries. The main approaches, developed in recent years to exploit the use of enzymes, antibodies or antibody mimics in water-restricted environments for analytical purposes, as well as possible future directions are briefly discussed.  相似文献

A sequential injection analysis (SIA) system was developed with the aim of obtaining an automatic and versatile way to prepare standards needed in the study of systems with higher dimensional sensor signals. To illustrate this, different analytical techniques were used in determinations of several analytes. Automated potentiometric calibrations of different potentiometric sensors, with and without interference, were carried out. Useful determinations of selectivity coefficients with two degrees of freedom were obtained. Simultaneous voltammetric determinations have also been done. Firstly, simultaneous determinations of lead and cadmium, using epoxy-graphite composite as the working electrode, have enabled a separate calibration for each metal to be obtained. Next, a voltammetric electronic tongue was designed and applied to the determination of oxidizable species. The use of artificial neural networks has solved the overlapped signal of ascorbic acid, 4-aminophenol and 4-acetamidophenol (paracetamol). A set of 63 data points was prepared automatically and has facilitated the training of an electronic tongue for these three analytes. Accurate predictions of test solutions, in the range of 12–410 μM for ascorbic acid, 17–530 μM for 4-aminophenol and 10–420 μM for paracetamol, have been achieved with RMSEs lower than 0.10 μM.  相似文献

Several non-equilibrium solid solutions belonging to the platinum-osmium systems Os0.9Pt0.1, Os0.8Pt0.2, Os0.5Pt0.5, Os0.7Pt0.3, Os0.75Pt0.25 are prepared and studied. The thermal decomposition of [Pt(NH3)4][OsCl6] in the hydrogen and helium atmosphere is investigated. It is found that the Pt0.5Os0.5 solid solution develops through the formation of (NH4)2[OsCl6] and a metallic phase based on Pt. The crystal structure of a double complex salt [Pt(NH3)4][OsCl6] is studied (X8-APEX Bruker, 1508 independent reflections, R = 2.04%). Crystal data for PtOsN4Cl6H12 are: a = 11.6216(5) Å, b = 11.0016(5) Å, c = 10.3819(5) Å, V = 1327.4(1) Å3, space group Cmca, Z = 4, d x = 3.333 g/cm3. The coordination polyhedron around Os is octahedral: 〈Os-Cl〉 2.357 Å, ∠Cl-Os-Cl 89.5–90.5°, while around Pt it is square-planar: Pt-N 2.046 Å, ∠ N-Pt-N 89.59° and 90.41°.  相似文献
