Natural herbal medicines are an important source of enzyme inhibitors for the discovery of new drugs. A number of natural extracts such as green tea have been used in prevention and treatment of diseases due to their low‐cost, low toxicity and good performance. The present study reports an online assay of the activity and inhibition of the green tea extract of the Glucose 6‐phosphate dehydrogenase (G6PDH) enzyme using multilayer capillary electrophoresis based immobilized enzyme microreactors (CE‐IMERs). The multilayer CE‐IMERs were produced with layer‐by‐layer electrostatic assembly, which can easily enhance the enzyme loading capacity of the microreactor. The activity of the G6PDH enzyme was determined and the enzyme inhibition by the inhibitors from green tea extract was investigated using online assay of the multilayer CE‐IMERs. The Michaelis constant (Km) of the enzyme, the IC50 and Ki values of the inhibitors were achieved and found to agree with those obtained using offline assays. The results show a competitive inhibition of green tea extract on the G6PDH enzyme. The present study provides an efficient and easy‐to‐operate approach for determining G6PDH enzyme reaction and the inhibition of green tea extract, which may be beneficial in research and the development of natural herbal medicines. Many properties of Brownian motion on spaces with varying dimension (BMVD in abbreviation) have been explored in Chen and Lou (2018). In this paper, we study Brownian motion with drift on spaces with varying dimension (BMVD with drift in abbreviation). Such a process can be conveniently defined by a regular Dirichlet form that is not necessarily symmetric. Through the method of Duhamel’s principle, it is established in this paper that the transition density of BMVD with drift has the same type of two-sided Gaussian bounds as that for BMVD (without drift). As a corollary, we derive Green function estimate for BMVD with drift.  相似文献

We present an existence result for Lévy‐type processes which requires only weak regularity assumptions on the symbol with respect to the space variable x. Applications range from existence and uniqueness results for Lévy‐driven SDEs with Hölder continuous coefficients to existence results for stable‐like processes and Lévy‐type processes with symbols of variable order. Moreover, we obtain heat kernel estimates for a class of Lévy and Lévy‐type processes. The paper includes an extensive list of Lévy(‐type) processes satisfying the assumptions of our results.  相似文献

Motivated by applications to machine learning, we construct a reversible and irreducible Markov chain whose state space is a certain collection of measurable sets of a chosen l.c.h. space X. We study the resulting network (connected undirected graph), including transience, Royden and Riesz decompositions, and kernel factorization. We describe a construction for Hilbert spaces of signed measures which comes equipped with a new notion of reproducing kernels and there is a unique solution to a regularized optimization problem involving the approximation of L2 functions by functions of finite energy. The latter has applications to machine learning (for Markov random fields, for example).  相似文献

To increase the profitability and sustainability of agricultural waste, a facile green approach was established to synthesize zinc oxide nanoparticles (ZnO NPs) using saffron leaf extract as a reducing and stabilizing agent. Structural characteristics of NPs were investigated by X-ray diffraction (XRD), Fourier-transform infrared (FTIR), field emission scanning electron microscopy (FESEM), and UV–Visible (UV–Vis) spectroscopy. Characterization results revealed that ZnO NPs is highly crystalline with a hexagonal wurtzite structure and spherical particles with diameter less than 50 nm, as confirmed by XRD and FESEM techniques. UV–Vis absorption spectra depicted an absorption peak at 370 nm, which confirms the formation of ZnO NPs. FTIR spectral analysis confirmed the presence of functional groups and metal oxygen groups. The biological activities of ZnO NPs were also investigated. The antibacterial effect of ZnO NPs was investigated against selected food pathogens (Salmonella Typhimurium, Listeria monocytogenes, and Enterococcus faecalis). The study results prove that the green synthesized ZnO NPs show enhanced antibacterial activity against S. Typhimurium when compared with other strains. A dose-dependent free radical scavenging activity was observed for ZnO NPs in both 2,2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH) and fluorescence recovery after photobleaching (FRAP) assays. The ZnO NPs were evaluated for their photocatalytic activity during the degradation of methylene blue (MB) dye in aqueous solutions. The maximum removal of MB achieved was 64% with an initial ZnO NP concentration of 12 mg/mL under UV light. The present study revealed that the agricultural waste (saffron leaf) provides a simple and eco-friendly option to sustainably synthesize ZnO NPs for use as a photocatalyst. In addition, this is the first report on saffron leaf-mediated synthesis of ZnO NPs.  相似文献

We report an ecofriendly synthetic approach for the fabrication of biogenic gold nanoparticles (AuNPs) using electron-rich sea cucumber extract as a bio-reductant and stabilizing agent in reducing gold cations into AuNPs at the optimal conditions. The produced AuNPs are spherical in shape with an average particle size of 11 ± 1.5 nm in transmission electron microscopy (TEM) and exhibited a crystal structure of face-centered cubic in X-ray diffraction (XRD) analyses. Our results indicated that bioinspired AuNPs demonstrate superior catalytic activity in the safe and facile one-pot synthesis of polyhydroquinoline derivatives under solvent-free reaction conditions. This green route encompasses multiple benefits including highly recyclable bioinspired catalyst (5 cycles), short reaction times, convenient workout, high to excellent product yields (82%–97%), and nonhazardous conditions.  相似文献

A rapid and sensitive liquid chromatography tandem mass spectrometry (LC/MS/MS) method was developed and validated using spinasterol as the internal standard (IS) for the simultaneous determination of shionone and epi‐friedelinol in rat plasma. Plasma samples were pretreated using liquid–liquid extraction with ethyl ether. Chromatographic separation was achieved on a C18 column (100 × 2.1 mm, 5 μm) with an isocratic elution consisting of acetonitrile–0.1% formic acid water (75:25, v/v) at a flow rate of 0.30 mL/min. Detection was performed under the selected reaction monitoring scan using an electrospray ionization in the positive ion mode. The mass transitions were as follows: m/z 427.4 → 95.1 for shionone, m/z 411.4 → 205.2 for epi‐friedelinol and m/z 395.3 → 105.2 for IS. All calibration curves exhibited good linearity (r > 0.995) over the concentration range for both components. The intra‐ and inter‐day precisions at three QC and lower limit of quantitation levels were both <10.21% in terms of relative standard deviation, and the accuracy ranged from ?7.13 to 8.02% in terms of relative error. The extraction recoveries of the compounds ranged from 82.07 to 89.81%. The developed method was successfully applied to the pharmacokinetic study of shionone and epi‐friedelinol after oral administration of Aster tataricus extract to rats. A rapid and sensitive liquid chromatography with tandem mass spectrometry (LC‐MS/MS) method was developed and validated for the simultaneous determination of luteolin, luteolin‐7‐O β ‐D‐glucopyranoside, physalin A, physalin D and physalin L in rat plasma. Scutellarein and dexamethasone were used as the internal standards (IS). Plasma samples were prepared by liquid‐liquid extraction with ethyl acetate. The five constituents were separated on an Acquity UPLC BEH C18 column (100 mm × 2.1 mm, 1.7 μm). A gradient elution procedure was used with acetonitrile (A)‐0.1% aqueous formic acid (B). Mass spectrometric detection was performed in negative ion multiple reaction monitoring mode with an electrospray ionization (ESI) source. This method showed good linearity (r 2 > 0.997) over a concentration range of 2.0–500 ng/mL with a lower limit of quantification of 2.0 ng/mL for all five compounds. The inter‐ and intra‐day accuracy ranged from 91.7 to 104%, and precisions (RSD) were <6.46% for all analytes. The extraction recoveries of all analytes were >85%. This validated method was successfully applied for the first time to the pharmacokinetic study of five ingredients after oral administration of 70% ethanol extract of Chinese lantern in rats.  相似文献
