A novel nonenzymatic sandwich-type electrochemical immunosensor has been developed to detect squamous cell carcinoma antigen (SCCA). Nitrogen-doped graphene sheet (N-GS) was used to increase capacity of capturing primary antibodies (Ab1). Carbon-supported Pd–Au binary nanoparticles (Pd–Au/C) were synthesized and used to label secondary antibodies (Ab2). The specific binding of SCCA and antibodies enabled a quantitative attachment of Pd–Au/C on the electrode surface. Electrocatalytic analysis showed that the prepared Pd–Au/C exhibit excellent electrocatalytic activity towards hydrogen peroxide (H2O2). We use current response of electrocatalytic labels Pd–Au/C to detect the concentration of SCCA. The unique nonenzymatic immunosensor exhibits a relatively wide linear range from 0.005 to 2 ng mL−1 and high sensitivity with a low detection limit of 1.7 pg mL−1. The immunsensor also shows good reproducibility (4.2%) and stability (5.8%), which makes it an enormous application prospect in clinical research.  相似文献

Modified glass fibers with three different amino silanes—(1) 3-aminopropyltriethoxysilane (APTES), (2) 3-aminopropylmethyldiethoxysilane (APDES), and (3) 3-aminopropyldimethylethoxysilane (APMES)—were labeled with 5-dimethylaminonaphthalene-1-sulfonylchloride (DNS), and its fluorescence was followed as a function of curing time for two epoxide formulations (TRIEPOX-LM, Gairesa, Spain, and diglycidyl ester bisphenol A–ethylenediamine), as well as a function of water uptake. It was demonstrated that the fluorescence response from fluorophores directly attached to the coupling region in glass fibers/epoxy composites is a very sensible method for nondestructive and “in situ” monitoring of any changes that appear at the interphase (rigidity and solvent accessibility). In addition, fluorescence from a dansyl label shows more rigidity when APMES is used as the coupling agent. This suggests that the greater the cross-linking degree at the coupling region, the smaller the epoxy chain interdiffusion. Finally, the water uptake process in glass fiber/epoxy composites can be divided into two steps; the first step corresponds to the diffusion of water to the coupling region, and the second step corresponds to the hydrolysis of siloxane bonds in the interphase. Both steps are dependent on the surface treatment of the glass fibers.  相似文献

Increased competition from store brands is forcing manufacturers to re-evaluate their strategies in regard to pricing and contracting with trade intermediaries. We analyze a supply chain in which a retailer accepts (with the appropriate contractual agreements) a national brand for resale and then determines whether to introduce a store brand, how to price the store brand, and what quantities of the product(s) to order. We show that when the national brand’s cost per unit quality (CPUQ) is larger than the store brand’s CPUQ, then the retailer seeks to introduce the store brand (SB) and the national brand (NB) manufacturer/supplier is unable to deter him from doing so. We find that the efficiency loss in the decentralized supply chain becomes smaller when a store brand is introduced. Recognizing the inadequacy of standard contracts in coordinating this supply chain, we propose a simple minimum order quantity contract that can coordinate this supply chain.  相似文献

There are no easily obtainable EPR spectral parameters for lipid spin labels that describe profiles of membrane fluidity. The order parameter, which is most often used as a measure of membrane fluidity, describes the amplitude of wobbling motion of alkyl chains relative to the membrane normal and does not contain explicitly time or velocity. Thus, this parameter can be considered as nondynamic. The spin-lattice relaxation rate () obtained from saturation-recovery EPR measurements of lipid spin labels in deoxygenated samples depends primarily on the rotational correlation time of the nitroxide moiety within the lipid bilayer. Thus, can be used as a convenient quantitative measure of membrane fluidity that reflects local membrane dynamics. profiles obtained for 1-palmitoyl-2-(n-doxylstearoyl)phosphatidylcholine (n-PC) spin labels in dimyristoylphosphatidylcholine (DMPC) membranes with and without 50 mol% cholesterol are presented in parallel with profiles of the rotational diffusion coefficient, R, obtained from simulation of EPR spectra using Freed’s model. These profiles are compared with profiles of the order parameter obtained directly from EPR spectra and with profiles of the order parameter obtained from simulation of EPR spectra. It is shown that and R profiles reveal changes in membrane fluidity that depend on the motional properties of the lipid alkyl chain. We find that cholesterol has a rigidifying effect only to the depth occupied by the rigid steroid ring structure and a fluidizing effect at deeper locations. These effects cannot be differentiated by profiles of the order parameter. All profiles in this study were obtained at X-band (9.5 GHz).  相似文献

The resulting cutinase–pincer–metal hybrids crystallized under halide‐rich conditions to give monomeric crystal structures, but also crystallized under halide‐poor conditions to form a metal‐induced dimer. Pulsed electron paramagnetic resonance (EPR) dipolar spectroscopy (PDS) offers several methods for measuring dipolar coupling constants and thus the distance between electron spin centers. Up to now, PDS measurements have been mostly applied to spin centers whose g-anisotropies are moderate and therefore have a negligible effect on the dipolar coupling constants. In contrast, spin centers with large g-anisotropy yield dipolar coupling constants that depend on the g-values. In this case, the usual methods of extracting distances from the raw PDS data cannot be applied. Here, the effect of the g-anisotropy on PDS data is studied in detail on the example of the low-spin Fe3+ ion. First, this effect is described theoretically, using the work of Bedilo and Maryasov (Appl. Magn. Reson. 2006 , 30, 683–702) as a basis. Then, two known Fe3+/nitroxide compounds and one new Fe3+/trityl compound were synthesized and PDS measurements were carried out on them using a method called relaxation induced dipolar modulation enhancement (RIDME). Based on the theoretical results, a RIDME data analysis procedure was developed, which facilitated the extraction of the inter-spin distance and the orientation of the inter-spin vector relative to the Fe3+ g-tensor frame from the RIDME data. The accuracy of the determined distances and orientations was confirmed by comparison with MD simulations. This method can thus be applied to the highly relevant class of metalloproteins with, for example, low-spin Fe3+ ions.  相似文献

A rapid, homogenous, antibody-free assay for phosphatase enzymes was developed using the phosphorescent platinum (II)-coproporphyrin label (PtCP) and time-resolved fluorescent detection. An internally quenched decameric peptide substrate containing a phospho-tyrosine residue, labeled with PtCP-maleimide and dabcyl-NHS at its termini was designed. Phosphatase catalysed dephosphorylation of the substrate resulted in a minor increase in PtCP signal, while subsequent cleavage by chymotrypsin at the dephosphorylated Tyr-Leu site provided a 3.5 fold enhancement of PtCP phosphorescence. This phosphorescence phosphatase enhancement assay was optimized to a 96 well plate format with detection on a commercial TR-F plate reader, and applied to measure the activity and inhibition of alkaline phosphatase, recombinant human CD45, and tyrosine phosphatases in Jurkat cell lysates within 40 min. Parameters of these enzymatic reactions such as Km's, limits of detection (L.O.D's) and IC50 values for the non-specific inhibitor sodium orthovanadate were also determined.  相似文献

We present a new photoswitchable spin label for light-induced pulsed electron paramagnetic resonance dipolar spectroscopy (LiPDS), the photoexcited triplet state of erythrosin B (EB), which is ideal for biological applications. With this label, we perform an in-depth study of the orientational effects in dipolar traces acquired using the refocused laser-induced magnetic dipole technique to obtain information on the distance and relative orientation between the EB and nitroxide labels in a rigid model peptide, in good agreement with density functional theory predictions. Additionally, we show that these orientational effects can be averaged to enable an orientation-independent analysis to determine the distance distribution. Furthermore, we demonstrate the feasibility of these experiments above liquid nitrogen temperatures, removing the need for expensive liquid helium or cryogen-free cryostats. The variety of choices in photoswitchable spin labels and the affordability of the experiments are critical for LiPDS to become a widespread methodology in structural biology.  相似文献

The detection and quantification of nanoplastics in aquatic environments is one of the major challenges in environmental and analytical research nowadays. The use of common analytical techniques for this purpose is not only hampered by the size of nanoplastics, but also because they are mainly made of carbon. In addition, the expected concentrations in environmental samples are below the detection limit of the majority of analytical techniques. In this context, the great detection capabilities of Inductively Coupled Plasma Mass Spectrometry (ICP-MS) in its Single Particle mode (SP-ICP-MS) have made of this technique a good candidate for the analysis of nanoplastics. Since the monitoring of carbon by ICP-MS faces several difficulties, the use of metal tags, taking advantage of the great potential of nanoplastics to adsorb chemical compounds, has been proposed as an alternative. In this perspectives paper, three different strategies for the analysis of polystyrene (PS) nanoplastics by SP-ICP-MS based on the use of metals species (ions, hydrophobic organometallic compound, and nanoparticles) as tags are presented and discussed. Advantages and disadvantages of each strategy, which rely on the labelling process, are highlighted. The metal nanoparticles labelling strategy is shown as a promising tool for the detection and quantification of nanoplastics in aqueous matrices by SP-ICP-MS.  相似文献
