Arginase isolated from beef liver was covalently attached to a polyacrylamide bead support bearing carboxylic groups activated by a water-soluble carbodiimide. The most favorable carbodiimide wasN-cyclohexyl-Nt’-(methyl-2-p-nitrophenyl-2-oxoethyl) aminopropyl carbodiimide methyl bromide, but for practical purposes,N-cyclohexyl-Nt’-morpholinoethyl carbodiimide methyl tosylate was used. The optimal conditions for the coupling procedure were determined. The catalytic activity of the immobilized arginase was 290–340 U/g solid or 2.9–3.4 U/mL wet gel. The pH optimum for the catalytic activity was pH 9.5, the apparent temperature maximum was at 60°C and Kmapp was calculated to be 0.37M L-arginine. Immobilization markedly improved the conformational stability of arginase. At 60°C, the pH for maximal stability was found to be 8.0. The immobilized arginase was used for the production of L-ornithine and D-arginine.  相似文献

A review on developments of liquid membranes (LMs) in the field of gas and vapor separation of the last 16 years is presented. Liquid membrane configurations employing supports, i.e. immobilized, supported and contained liquid membranes are focussed and detailed information on the respective materials, i.e. supports (supplier, type, thickness, pore width, porosity, tortuosity), liquids and carriers, are presented together with their specific separation tasks. Performance of different LMs in terms of permeability and selectivity as well as stability (duration of testing, applied differential pressures) are compared and discussed. Finally, different preparation methods of LMs are illustrated.  相似文献

针对人神经母细胞瘤SH-SY5Y细胞系的磷酸化膜蛋白质组,发展了基于多酶酶解法结合杂化硅胶基质固定化钛离子亲和色谱(Ti4+-IMAC)整体柱富集的分析策略。该方法通过对细胞裂解液进行超速离心,以及1 mol/L NaCl和0.1 mol/L Na2CO3顺序清洗,获得膜蛋白质组分。所提取的蛋白质分别经胰蛋白酶、胰凝乳蛋白酶和胃蛋白酶平行酶解,产生的肽段经Ti4+-IMAC整体柱选择性富集磷酸肽后,采用纳升级反相液相色谱分离和质谱鉴定,成功鉴定到43个磷酸化蛋白质,其中有14个定位于膜上。研究结果表明,采用该策略开展SH-SY5Y细胞系磷酸化膜蛋白质组学分析有望加速对该肿瘤的研究和相关潜在标记物的筛选。  相似文献

The axial dispersion of a liquid fluidized bed of controlled pore silica (CPS) particles has been determined by the pulse tracer method. The CPS used was the same as for enzyme immobilization, having an average diameter of 0.436 mm and mean pore size of 37.5 nm. The fluidization liquid is α-amylase liquefied manioc starch, 30% w/v, 45°C pH=4.5. Nominal bed porosities tested were 0.7 and 0.8. The results show that the axial dispersion coefficient increases with greater superficial liquid velocities. Various available correlations tested disagree with each other to a large extent and are unable to represent collected experimental data.  相似文献

Summary: Synthesis, characteristics and medical diagnostic application of core-shell poly(styrene/α-tert-butoxy-ω-(vinylbenzyl)polyglycidol) (P(S/PGL)) microspheres are described. The particles were prepared by soap-free emulsion polymerization of styrene and α-tert-butoxy-ω-(vinylbenzyl)polyglycidol macromonomer (PGL) initiated with potassium persulfate. The polymerization in water yielded microspheres with diameters in the range from 220 to 650 nm, depending on concentration of the macromonomer. Polydispersity was usually below 1.06. The fraction of PGL in interfacial layer of microspheres ranged from 0 to 42 mol.% (estimated by XPS). Microspheres suspended in NaCl solutions manifested fully reversible swelling-deswelling properties of their interfacial layer with characteristic transition temperature (Tt) slightly dependent on NaCl concentration. The adsorption of human serum albumin on the surface of P(S/PGL) microspheres was highly reduced when the PGL surface fraction exceeded 40 mol.%. P(S/PGL) microspheres with immobilized antigens Helicobacter pylori were used for detection of antibodies against. The test was based on monitoring differences in electrophoretic mobility of P(S/PGL) microspheres covalently bound H. pylori antigens and antibodies against H. pylori in blood serum.  相似文献

Some metal‐chelating peptides have antioxidant properties, with potential nutrition, health, and cosmetics applications. This study aimed to simulate their separation on immobilized metal ion affinity chromatography from their affinity constant for immobilized metal ion determined in surface plasmon resonance, both technics are based on peptide‐metal ion interactions. In our approach, first, the affinity constant of synthetic peptides was determined by surface plasmon resonance and used as input data to numerically simulate the chromatographic separation with a transport‐dispersive model based on Langmuir adsorption isotherm. Then, chromatographic separation was applied on the same peptides to determine their retention time and compare this experimental tR with the simulated tR obtained from simulation from surface plasmon resonance data. For the investigated peptides, the relative values of tR were comparable. Hence, our study demonstrated the pertinence of such numerical simulation correlating immobilized metal ion affinity chromatography and surface plasmon resonance.  相似文献

The synthesis of formic acid from carbon dioxide and hydrogen using a silica immobilized ruthenium catalyst as precursor has been studied in different reaction conditions. The results revealed that the TOF (turn over frequency) of HCOOH achieved 1481.5 h^-1 on immobilized ruthenium catalyst near the critical pressure point of CO2 with H2 pressure of 4.0 MPa, reaction temperature of 80℃ and PPh3/Ru molar ratio of 6:1. The reaction activity of immobilized catalyst was higher than that of homogeneous catalyst, and the immobilized catalyst also offered the practical advantages such as easy separation and reuse.  相似文献

A porous, solid insoluble polysiloxane‐immobilized ligand system bearing pyrogallol active sites of the general formula P? (CH2)3? NH(CH2)3OC6H3(OH)2 (where P represents [Si? O]n siloxane network) has been prepared by the reaction of 3‐aminopropylpolysiloxane with 1,3‐dibromopropane followed by the reaction with pyrogallol. 13C CP‐MAS NMR and X‐ray photoelectron spectroscopy confirmed that the pyrogallol is chemically bonded to the siloxane backbone. Thermal analysis showed that the ligand system is stable under nitrogen at relatively high temperature. The polysiloxane–pyrogallol ligand system exhibits high potential for the uptake of the metal ions (Fe3+, Co2+, Ni2+ and Cu2+). Complexation of the pyrogallol ligand system for the metal ions at the optimum conditions was found to be in the order Fe3+ > Cu2+ > Ni2+ > Co2+. Copyright © 2005 John Wiley & Sons, Ltd.  相似文献

Cross‐polarization (CP) at fast magic angle spinning (MAS) frequencies leads to a splitting of the Hartmann–Hahn (HH) matching profile into a centerband and additional bands of higher orders. The matching profiles differ with the substance categories. Therefore, signal intensity is usually lost, when e.g. the routine standard NH4H2PO4 is used for optimizing the 1H–31P HH match prior to measuring phosphines and their metal complexes in polycrystalline or immobilized form. Here, a variety of model compounds, such as Ph2PCH2CH2PPh2 and (CO)2Ni(PPh3)2, which can be used as 31P CP standards for analogous substances or materials are presented. Investigating the influences of MAS frequency, contact time, 1H pulse power and sample volume on the matching profiles of the model compounds leads to general trends. Thereby, a new strategy for measuring difficult samples with CP at high MAS rates has been developed: their optimum CP parameters are derived from the most intense maxima in the HH matching profiles of the corresponding model compounds. This new strategy is compared with variations of a conventional ramp sequence. Although the latter generally provide smaller signal half‐widths, the new strategy leads to higher signal intensities. Staphylococcal nuclease has been insolubilized, directly through its amino groups, on CNBr-activated Sepharose 2B. For kinetic studies, a small substrate (thymidine 5′-(p-nitrophenyl phosphate) 3′-phosphate) has been used to measure the hydrolytic activity. With this system the absence of diffusional limitation has been proven. Eadie-Hofstee analysis of the data has been employed to determine the intrinsic kinetic constants of the insolubilized enzyme. Thek cat-pH andK M−pH profiles and the activation energies are similar for the soluble and for the insolubilized nuclease. At the same time conditions are established in which a stirred batch reactor containing particles of insolubilized nuclease behaves as an open system.  相似文献
