The activity of cellulase has traditionally been described by pH and temperature; however, the buffering medium is also an important factor, Taking plain water as a reference medium, three kinds of buffer including KH2PO4/K5HPO4, citric acid/sodium citrate, and acetic acid/sodium acetate were adopted to survey their effects on the activity of cellulase. Chromatographic assays indicated that xylose, glucose, and cellobiose were the major products and that minor products such as cellotriose and cellotetraose were present in some cases. The activities of cellulase based on glucose production showed that the phosphate buffer acted as a deactivator for cellulase and each of the two organic acid buffers acted as activators for cellulase. The concentration of activation buffer should be high to reach a high cellulase activity; however, this effect would be compensated for by the product inhibition of cellulase. The highest activity obtained was 4.16 ± 0.08 (× 10?3) IU mg?1 for the citric acid/sodium citrate buffer under pH 4.80, 40 °C and an agitation speed of 150 rpm. 相似文献
The effects of catalysts, pH and reaction conditions on the course of the hydrolysis and condensation of ETS40 (ethyl silicate 40), and on the composition of the reaction products were studied with the aid of gas and gel chromatography, potentiometry and gelation tests. Strong acids (HCl, HClO4, HNO3, H2SO4, p-toluenesulphonic acid), weak acids (Cl3, CCOOH, ClCH2COOH, (COOH)2, CH3COOH and HCOOH) and bases (LiOH, NH4,OH) were used as catalysts.
The hydrolysis rate increased with increasing temperature, catalyst concentration, initial water concentration and initial ethyl silicate concentration, whereas it decreased with increasing number of Si atoms in the ethyl silicate molecules. At pH 0–7 the hydrolysis was acid catalysed, but at pH above 7.0 it was base catalysed. Simultaneously with the hydrolysis, condensation occurred at a rate which increased with increasing temperature, catalyst concentration, ETS40 concentration and, above all, with increasing initial water concentration. The condensation rate depended on the pH. The condensation was at its slowest for pH around 2.0. For pH below 2.0, the condensation increased with increasing hydrogen ion concentration; for pH above 2.0 the condensation increased with decreasing hydrogen ion concentration. Phosphoric acid and hydrofluoric acid increased the rate of condensation considerably. The reaction of ETS40 with water at pH around 2.0 gave rise during the hydrolysis to solutions of ethoxyhydroxysiloxanes with an average of 14–20 Si atoms in a molecule, which displayed long-term stability. 相似文献
Orifluoroacetylation of the 25(OH)-group with subsequent dehydration of the 14(OH)-group takes place in the reaction of 20-hydroxyecdysone 20,22-acetonide and 2,3:20,22-diacetonide with trifluoroacetic anhydride in the presence of pyridine. A rapid peptide-bond hydrolysis by means of microwave irradiation is introduced for the facile preparation of protein hydrolysates used for amino acid analysis. The optimal hydrolysis condition has been determined using several enzymes with known amino acid compositions. The effects of hydrolysis time on the recovery of various labile and hydrophobic amino acids are also exemplified in the microwave heating of standard amino acids. The method has been applied to the complete amino acid analysis with a single nonvolatile solvent of methanesulfonic acid with good recovery of tryptophan and half-cystine. It provides a radical expedition of protein and peptide hydrolysis via commercial microwave ovens and specially-designed Teflon-Pyrex tubes, circumventing the tedious procedures using vacuum-sealed pyrex lubes heating at 110°C for more than 24 h. This novel type of microwave chemistry associated with rapid peptide-bond cleavage is of great potential in the automation of the complete process of amino acid analysis starting from the preparation of protein hydrolysates. 相似文献

Nano sized crystalline particles/polymer hybrids were synthesized form designed metal-organic precursors. The newly developed method is composed of the synthesis of organic matrix by polymerization and the in situ nucleation and growth of crystalline oxide particles in the organic matrix below 100°C. The design of metal-organic precursor modified with polymerizable ligand and the selection of reaction conditions does influence the size and crystallinity of ceramic particles in organic matrix. The nano-sized magnetic particle/polymer hybrid exhibits the interesting feature of superparamagnetism and quantum size effect. The crystalline particles of BaTiO3/, PbTiO3/, and KNbO3/polymer hybrids behave to be dielectric and show the typical electro-rheological behavior. 相似文献
Oxidative couplings of 2-naphthol, 6-bromo-2-naphthol and2-naphthylamine were achieved at room temperature in the presence of H2O2, horseradish peroxidase and a suitable cyclodextrin.2-Thionaphthol behaved differently, yielding the corresponding disulfide. Yields of binaphthyl derivatives were generally excellent, and a fairly good enantiomeric excess was observed. Under similar reaction conditions methyl 2-(6-methoxy-2-naphthyl) propanoate, when treated with esterase in the presence of cyclodextrin, yielded naproxen (a well-known anti-inflammatory drug) with a good enantiomeric excess. No reaction product was detected in the absence of cyclodextrin. Cyclodextrins do not act as simple transfer agents. 相似文献
Basic salts of Co(II) crystallize in the rhombohedral structure. Two different polytypes, 3R1 and 3R2, with distinct stacking sequences of the metal hydroxide slabs, are possible within the rhombohedral structure. These polytypes are generated by simple translation of successive layers by (2/3, 1/3, z) or (1/3, 2/3, z). The symmetry of the anion and the mode of coordination influences polytype selection. Cobalt hydroxynitrate crystallizes in the structure of the 3R2 polytype while the hydroxytartarate, hydroxychloride and α-cobalt hydroxide crystallize in the structure of the 3R1 polytype. Cobalt hydroxysulfate is turbostratically disordered. The turbostratic disorder is a direct consequence of the mismatch between the crystallographically defined interlayer sites generated within the crystal and the tetrahedral symmetry of the SO42− ions. 相似文献
