Abstract The electrochemical behavior of 9‐phenanthrol in the presence of copper (II) at a static mercury drop electrode was investigated to provide the basis for development of an inexpensive, sensitive, and reliable method for determination of polycyclic aromatic hydrocarbon (PAH) metabolites in biological matrices. Optimum experimental conditions for analytical applications were obtained in 0.005 M NaOH solution using an accumulation potential of ?0.25 V, a scan rate of 5 mV. s?1, a pulse height of 25 mV, and a differential pulse scan mode. The response of 9‐phenanthrol is linear over the concentration range 1.0–12.0 ppb. For an accumulation time of 5 minutes, the detection limit was found to be 0.2 ppb (1.03×10?9 M). The more convenient relation to measure the 9‐phenanthrol in the presence of copper and other metals was also investigated. The utility of the method was demonstrated by the presence of 9‐phenanthrol in samples of sea water and human urine. Cyclic voltammetry was used to characterize the interfacial and redox behavior. 相似文献

lectins. Specific binding patterns to normal and transformed mammary tissues were evaluated by Isoform 1 fromCratylia mollis lectin (Cra Iso 1). Protocols using a direct method, incubation of Cra Iso I conjugated to peroxidase (Cra Iso 1-Per) with
mammary tissues, followed by diaminobenzidine and hydrogen peroxidase interaction, were performed. Neoplastic tissues, marked
by Cra Iso 1, showed a higher intensity of staining than normal ones, in comparison withCanavalia ensiformis lectin, Concanavalin A (Con A), conjugated to peroxidase (Con A-Per). The assay with Cra Iso 1 also indicated a possible

HIV infection leads to progressive alterations of humoral immune functions, including B-cell hyperplasia, hypergammaglobulinemia,
elevated autoantibody titers, a poor response to neoantigens and mitogens, polyclonal B-cell activation, monoclonal gammopathies,
and a significant deterioration of the antigen-specific humoral response. There is also an important isotypic imbalance of
the antibody (Ab) response in the systemic compartment and a profound modification of mucosal immune functions. These abnormalities
may contribute to disease progression and development of opportunistic infections, despite the presence of serum-neutralizing
anti-HIV Abs. Equally important are the abnormal selection mechanisms of the Ab repertoire that seem to be responsible for
B-cell clonal deletions. The VH3 gene family, which encodes for approx 50% of immunoglobulins expressed by peripheral B-cells from normal adults, is underrepresented
in human monoclonal antibodies to HIV-1 and in the peripheral B-cells of AIDS patients. These abnormalities, together with
features of germinal center alteration, could be responsible for the clonal elimination of a subset of B-cells, and could

Summary Using the X-ray crystal structure of the human topoisomerase I (top1) – DNA cleavable complex and the Sybyl software package, we have developed a general model for the ternary cleavable complex formed with four protoberberine alkaloids differing in the substitution on the terminal phenyl rings and covering a broad range of the top1-poisoning activities. This model has the drug intercalated with its planar chromophore between the −1 and +1 base pairs flanking the cleavage site, with the nonplanar portion pointing into the minor groove. The ternary complexes were geometry-optimized and relative interaction energies, computed by using the Tripos force field, were found to rank in correct order the biological potency of the compounds; in addition, the model is also consistent with the top1-poisoning inactivity of berberine, a major prototype of the protoberberine alkaloids. The model might serve as a rational basis for elaboration of the most active compound as a lead structure, in order to develop more potent top1 poisons as next generation anti-cancer drugs. 相似文献

A highly sensitive HPLC method was developed for the determination of xenoestrogenic compound, bisphenol A (BPA) in human breast milk samples. After a two-step liquid-liquid extraction, BPA was derivatized with fluorescent labeling reagent, 4-(4,5-diphenyl-1H-imidazol-2-yl)benzoyl chloride (DIB-Cl). The excess fluorescent reagent could be removed effectively using a column-switching system. The separation of DIB-BPA from endogenous materials in milk was carried out on two C(18) columns and fluorescence intensity was monitored at 475 nm with the excitation of 350 nm. A good linearity (r = 0.994) was observed of BPA in the concentration range of 0.2-5.0 ng mL(-1) in breast milk, and the detection limit was 0.11 ng mL(-1) at a signal-to-noise ratio of 3. Intra- and inter-day precision (RSD, %) were less than 8.7 and 10.4, respectively. Twenty-three breast milk samples of healthy lactating women were analyzed for the BPA concentration; the mean value was 0.61 +/- 0.20 ng mL(-1), with no correlation to the lipid content of milk samples. 相似文献

The multi-component isotherms for human insulin (HI) and desamido human insulin (dHI) over reversed phase packing (C18) and with 29.8% (w/w) ethanol-water as mobile phase have been determined experimentally. The isotherms of HI in ethanol-water differ from those obtained with the more commonly applied methanol-water and acetonitrile-water mobile phase, as described in this paper. The isotherm exhibits anti Langmuirian behavior and can be very well modeled by an anti Langmuir isotherm presented in this paper. 将18-甲基炔诺酮加到人血清白蛋白-酮洛芬平衡溶液中,室温孵育达平衡后应用毛细管电泳-迎头分析法研究了18-甲基炔诺酮和酮洛芬在人血清白蛋白分子上的置换相互作用。一大体积样品虹吸进样至未涂层毛细管(65 cm×50 μm i.d.,有效长度35 cm),进样时间80 s,毛细管两端高度差11 cm,工作电压10 kV,运行缓冲液为67 mmol/L磷酸盐缓冲液(pH 7.4),游离酮洛芬浓度由前沿峰的高度直接测定。当酮洛芬样品溶液中酮洛芬的总浓度分别为100 μmol/L和200 μmol/L时,随着18-甲基炔诺酮加入量的增加(18-甲基炔诺酮的浓度由0增至200 μmol/L),游离酮洛芬的浓度分别从22.4 增至26.4 μmol/L和从82.1增至106.2 μmol/L。由上面的结果可推论:高浓度的18-甲基炔诺酮可将酮洛芬从它的第二类结合位点上置换出来。 相似文献

根据中国营养学会推荐的热量标准,研制了中学生营养平衡米膳食最佳模式。按该模式,中学生每日应摄入的膳食品种及其数量如下:主食:营养平衡米,初中男生510~597g,女生468~510g;高中男生618~683g,女生532~575g;副食相同:青菜或水果500g或600g,鱼或肉100g或50g,蛋类40g,大豆制品30g,花生油12.5g,盐6g。调查结果表明,蛋白质、脂肪、碳水化合物,热量摄入量全部达标准要求。Q(蛋白质)∶Q(脂肪)∶Q(碳水化合物):高中男女生为16∶20∶64,初中男、女生分别为17∶20∶63。提示中学生营养平衡米膳食最佳模式使中学生达到营养平衡,建议推广食用。 相似文献
