Constrained Dipeptide Surrogates: 5- and 7-Hydroxy Indolizidin-2-one Amino Acid Synthesis from Iodolactonization of Dehydro-2,8-diamino Azelates

The constrained dipeptide surrogates 5- and 7-hydroxy indolizidin-2-one N-(Boc)amino acids have been synthesized from L-serine as a chiral educt. A linear precursor ∆⁴-unsaturated (2S,8S)-2,8-bis[N-(Boc)amino]azelic acid was prepared in five steps from L-serine. Although epoxidation and dihydroxylation pathways gave mixtures of hydroxy indolizidin-2-one diastereomers, iodolactonization of the ∆⁴-azelate stereoselectively delivered a lactone iodide from which separable (5S)- and (7S)-hydroxy indolizidin-2-one N-(Boc)amino esters were synthesized by sequences featuring intramolecular iodide displacement and lactam formation. X-ray analysis of the (7S)-hydroxy indolizidin-2-one N-(Boc)amino ester indicated that the backbone dihedral angles embedded in the bicyclic ring system resembled those of the central residues of an ideal type II’ β-turn indicating the potential for peptide mimicry.     

THE SYNTHESIS OF 1,3,2-DIOXAPHOSPHOLENES. EVIDENCE FOR AN ARYLOXYPHOSPHINIDENE INTERMEDIATE

Abstract

The reaction of 2,6-di-t-butyl-4-methylphenylphosphorodichloridite with α-diketones in the presence of magnesium metal affords modest yields of 1,3,2-dioxaphospholenes. This may be the first evidence for the existence of the monocoordinate aryloxyphosphinidene intermediate, ROP.     

Reaction of dichloromaleic anhydride with 1,8-diaminonaphthalene: Synthesis and X-ray diffraction structure of 8,9-dichloropyrrolo[1,2-a]perimidin-10-one

The reaction of dichloromaleic anhydride with 1,8-diaminonaphthalene in refluxing toluene or 1,2-dichloroethane produces the new heterocyclic compound 8,9-dichloropyrrolo[1,2-a]perimidin-10-one in low yields. 8,9-Dichloropyrrolo[1,2-a]perimidin-10-one has been isolated by column chromatography and characterized in solution by IR, ¹H NMR, and UV/vis spectroscopies. The solid-state structure was unequivocally established by single-crystal X-ray diffraction analysis. 8,9-Dichloropyrrolo[1,2-a]perimidin-10-one crystallizes in the monoclinic space group P2 ₁/c, a = 7.475(1)Å, b = 10.650(2)Å, c = 14.468(2)Å, = 94.478(2)°, V = 1148.3(3) ų, Z = 4, and d _calc = 1.672 Mg/m³; R = 0.0289, R _w = 0.0762 for 1644 reflections with I > 2(I). The nature of the HOMO and LUMO in 8,9-dichloropyrrolo[1,2-a]perimidin-10-one has been determined by extended Hückel molecular orbital calculations, and these data are discussed relative to the cyclic voltammetric data obtained at a platinum electrode.     

Efficient Synthesis of 2-Arylamino-2-imidazolines and 2-Ami-nobenzimidazoles with Aminoiminomethanesulfonic Acid Derivatives

A highly efficient synthesis of 2‐arylamino‐2‐imidazolines and 2‐aminobenzimidazoles from aminoimino‐methanesulfonic acid derivatives is described. The method is simple and practical, generating imidazoline and benzimidazoline derivatives in excellent isolated yields.     

Syntheses, Optical Properties and Photoactivated Insecticidal Activities of Cyclic Arylethynylsilanes

Syntheses and optical properties of cyclic arylethynylsilanes containing one or two trialkyne pockets were described. Optical properties such as UV‐vis spectra, photoluminescence and quantum yield based on these conjugated structural features were compared with each other. The photoactivated insecticidal activities against the 4th‐instar larvae of Aedes albopictus (Skuse) were evaluated.     

Synthesis and domino metathesis of functionalized 7-oxanorbornene analogs toward cis-fused heterocycles

Here, we report our strategy to synthesize structurally diverse cis-fused heterocycles via 7-oxanorbornene analogs. By the intramolecular Diels-Alder reaction of 3-iodoacrylamide, followed by the domino metathesis reaction, the cis-fused heterobicyclic skeleton was efficiently constructed in 5-8 steps.     

One-pot methodology for conversion of o-halogen nitrobenzenes to benzofuroxans

Reaction of o-halonitrobenzenes with sodium azide under reflux of DMF/H₂O gives benzofuroxans in one step in moderate to good yields. This is a faster methodology compared to the conventional procedure involving the preparation and subsequent pyrolysis of o-nitrophenyl azides. For comparison, the reaction was also performed under phase-transfer catalysis.     

Efficient copper-catalyzed N-arylation of NH-containing heterocycles and sulfonamides with arenediazonium tetrafluoroborates

A practical copper-catalyzed N-arylation of NH-containing heterocycles with arenediazonium tetrafluoroborates has been developed using CuCl as catalyst and K₂CO₃ as additive under ligand-free conditions. This reaction system has wide substrate scope including imides, 1H-pyrazole, 1H-tetrazoles, 1,2-benzisothiazol-3(2H)-one, and related sulfonamides and gives moderate to excellent yields (up to 95%) of the desired products. This strategy is very general, simple, environmentally friendly, and tolerant of oxygen.     

A Density Functional Study on the Hydrolysis Process of Non-classical Transplatin（Ⅱ） with Two Same Planar Heterocycle Amines

In the present work, the hydrolysis process of non-classical transplatin（Ⅱ） with two same planar heterocycle amines has been studied using hybrid density functional theory （B3LYP） and IEF-PCM solvation models. Optimizations were performed at the B3LYP level using a combined basis set of （LanL2DZ＋6-31 ＋G（d,p）） with single-point energy evaluations using the B3LYP/6-31 ＋ ＋G（3df,2pd） approach in vacuo and in aqueous solution. For the obtained structures of reactants, intermediates, transition states, and products, both thermodynamic （reaction energies and Gibbs energies） and kinetic （reaction barriers） characteristics were estimated. In comparison with cisplatin, decreased activation energies were obtained. The result implies that the non-classical transplatin with two same planar heterocycle amines increases the equatorial steric effect and lowers reaction barriers, which may assist in designing novel Pt-based anticancer drugs.
