研究了新试剂对甲苯偶氮水杨基荧光酮(MBASF)与钼(Ⅵ)显色反应,以建立水解液中钼的形态分布测定方法。在盐酸介质中,对MBASF与钼(Ⅵ)发生灵敏的显色反应,形成稳定的红色配合物。配合物表观摩尔吸光系数达1.54 L·mol-1·cm-1,在0~12 μg·(25 mL)-1范围内钼(Ⅵ)服从比耳定律。除常见金属离子外,蛋白、多肽和氨基酸均有高的允许量。大豆酶解液A经等电点沉淀法除去未水解蛋白后得溶液B,溶液B经D301R树脂吸附后,分别采用pH 3.6和9.0的缓冲液洗脱分离得溶液C和D;A,B,C和D溶液分别用于光度法测定总钼、水解钼、结合态钼(多肽和氨基酸螯合态钼)和游离钼的含量。  相似文献

Soybean protein hydrolysates were prepared using two proteolytic enzymes (Alcalase and Protamex) and the degree of hydrolysis (DH) and their functional and antioxidant properties were evaluated. The highest DH value was 20%, with a yield of 19.77% and protein content of 51.64%. The total amino acid content was more than 41% for all protein hydrolysates. The protein hydrolysates from Protamex at pH 2.0 had excellent solubility, emulsifying activity, and foaming capacity, at 83.83%, 95.03 m2/g, and 93.84%, respectively. The water-holding capacity was 4.52 g/g for Alcalase, and the oil-holding capacity was 4.91 g/g for Protamex. The antioxidant activity (62.07%), as measured by the samples’ reaction with DPPH (2,2-diphenyl-1-picrylhydrazyl) and the reducing power (0.27) were the strongest for Protamex. An ABTS activity rate of 70.21% was recorded for Alcalase. These findings indicated a strong potential for the utilization of soybean protein hydrolysates to improve the functional properties and antioxidant activity of soybeans as well as their nutritional values.  相似文献

The interaction of soy protein isolate (SPI) and its hydrolysates (SPIHs) with cyanidin-3-O-glucoside (C3G) at pH 7.0 were investigated to clarify the changes in the antioxidant capacity of their complexes. The results of intrinsic fluorescence revealed that C3G binds to SPI/SPIHs mainly through hydrophobic interaction, and the binding affinity of SPI was stronger than that of SPIHs. Circular dichroism and Fourier-transform infrared spectroscopy analyses revealed that the interaction with C3G did not significantly change the secondary structures of SPI/SPIHs, while the surface hydrophobicity and average particle size of proteins decreased. Furthermore, the SPI/SPIHs-C3G interaction induced an antagonistic effect on the antioxidant capacity (ABTS and DPPH) of the complex system, with the masking effect on the ABTS scavenging capacity of the SPIHs-C3G complexes being lower than that of the SPI-C3G complexes. This study contributes to the design and development of functional beverages that are rich in hydrolysates and anthocyanins.  相似文献

Two new high-through put techniques, computer tomography X-ray densitometry (CT scan) and pyrolysis molecular beam mass spectrometry (pyMBMS), coupled with quantitative trait loci (QTL) analysis, were tested as a means to overcome the time and cost associated with conventional characterization of biomass feedstock components. Applications of these two techniques were evaluated using hybrid poplar for the CT scan and loblolly pine for the pyMBMS. Segregating progeny from hybrid poplar varied in specific gravity, with individual mean estimates ranging from 0.21–0.41. Progeny from loblolly pine varied in lignin, α cellulose, and mannan contents, with individual mean estimates of lignin content ranging from 28.7–33.1%, α cellulose content from 28.8–43.5% and mannan content from 4.2–10.1%. QTL analysis of the loblolly pine data suggested that eleven QTLs were associated with individual feedstock characteristics and that two QTLs for several feedstock components were linked to the same position on the loblolly pine genetic map. Each QTL individually accounted for 7–13% of the total phenotypic variation in associated loblolly pine feedstock components.  相似文献

Gels and pastes are used in many areas including application in the cosmetic, pharmaceutical and medical industries. A xylan derivative with pasty consistency was prepared in addition to the synthesis of xylan ethers and esters. The water content of the gel can be widely varied. The gel is formed although the xylan contains no cross-linking group. The hydrophobic and hydrophilic nature of the xylan gel can be adjusted by modifying the xylan. All of the gels can be dried using different methods (air drying, solvent exchange as well as freeze drying) and the dried gels can again disperse in water. The gels were characterised by porosity measurements (mercury intrusion), dynamic vapour sorption (DVS), scanning electron microscopy (SEM) and dispersion stability investigation respectively.  相似文献

V–VI (Bi2Se3, Bi2Te3, Sb2Se3, and Sb2Te3) semiconductors nanorods with the diameters of 40–90 nm have been synthesized starting from BiCl3 (or SbCl3) and Na2SeO3 (or Na2TeO3) by using NaH2PO2·H2O as the reduced agent at 140°C for 24 h. The samples were characterized by XRD, SEM and TEM. XPS spectra of the products show the obtained samples are close to stoichiometry. The hydrolysate of the starting materials could be served as the precursor and sharply decreases the growth of the samples; a possible hydrolyzation–reduction–crystallization process was also proposed to explore the formation mechanism of these nanorods.  相似文献

Chicken stomachs can be processed into collagen hydrolysate usable in cosmetic products. The aim of the study was to verify the effects of a carbopol gel formulation enriched with 1.0% (w/w) chicken hydrolysate on the properties of the skin in the periorbital area after regular application twice a day for eight weeks in volunteers ageed 50 ± 9 years. Skin hydration, transepidermal water loss (TEWL), skin elasticity and skin relief were evaluated. Overall, skin hydration increased by 11.82% and 9.45%, TEWL decreased by 25.70% and 17.80% (always reported for the right and left area). Generally, there was an increase in skin elasticity, a decrease in skin roughness, as the resonance times decreased by 85%. The average reduction of wrinkles was 35.40% on the right and 41.20% on the left. For all results, it can be seen that the longer the cosmetic gel formulation is applied, the better the results. Due to the positive effect on the quality and functionality of the skin, it is possible to apply the cosmetic gel formulation in the periorbital area. The advantage of the product with chicken collagen hydrolysate is also the biocompatibility with the skin and the biodegradability of the formulation.  相似文献

Efficient utilization of the pentosan fraction of hemicellulose from lignocellulosic feedstocks offers an opportunity to increase the yield and to reduce the cost of producing fuel ethanol. During prehydrolysis (acid hydrolysis or autohydrolysis of hemicellulose), acetic acid is formed as a consequence of the deacetylation of the acetylated moiety of hemicellulose. Recombinant Escherichia coli B (ATCC 11303), carrying the plasmid pLO1297 with pyruvate decarboxylase and alcohol dehydrogenase II genes from Zymomonas mobilis (CP4), converts xylose to ethanol with a product yield that approaches theoretical maximum. Although other pentose-utilizing microorganisms are inhibited by acetic acid, the recombinant E. coli displays a high tolerance for acetic acid. In xylose fermentations with a synthetic medium (Luria broth), where the pH was controlled at 7, neither yield nor productivity was affected by the addition of 10.7 g/L acetic acid. Nutrient-supplemented, hardwood (aspen) hemicellulose hydrolysate (40.7 g/L xylose) was completely fermented to ethanol (16.3 g/L) in 98 h. When the acetic acid concentration was reduced from 5.6 to 0.8 g/L, the fermentation time decreased to 58 h. Overliming, with Ca(OH)2 to pH 10, followed by neutralization to pH 7 with sulfuric acid and removal of insolubles, resulted in a twofold increase in volumetric productivity. The maximum productivity was 0.93 g/L/h. The xylose-to-ethanol conversion efficiency and productivity in Ca(OH)2-treated hardwood prehydrolysate, fortified with only mineral salts, were 94% and 0.26 g/L/h, respectively. The recombinant E. coli exhibits a xylose-to-ethanol conversion efficiency that is superior to that of other pentose-utilizing yeasts currently being investigated for the production of fuel ethanol from lignocellulosic materials.  相似文献

The sugarcane bagasse hydrolysate, which is rich in xylose, can be used as culture medium for Candida guilliermondii in xylitol production. However, the hydrolysate obtained from bagasse by acid hydrolysis at 120°C for 20 min has by-products (acetic acid and furfural, among others), which are toxic to the yeast over certain concentrations. So, the hydrolysate must be pretreated before using in fermentation. The pretreatment variables considered were: adsorption time (15,37.5, and 60 min), type of acid used (H2So4 and H3Po4), hydrolysate concentration (original, twofold, and fourfold. concentrated), and active charcoal (0.5, 1.75 and 3.0%). The suitability of the pretreatment was followed by measuring the xylose reductase (XR) and xylitol dehydrogenase (XD) activity of yeast grown in each treated hydrolysate. The response surface methodology (24 full factorial design with a centered face) indicated that the hydrolysate might be concentrated fourfold and the pH adjusted to 7.0 with CaO, followed by reduction to 5.5 with H3PO4. After that it was treated with active charcoal (3.0%) by 60 min. This pretreated hydrolysate attained the high XR/XD ratio of 4.5.  相似文献
