Synthetic studies toward the immunosuppressant FR901483. Facile construction of the azatricyclic skeleton

A concise synthesis of the azatricyclic core structure of FR901483, a potent immunosuppressant, has been accomplished. The key elements of the approach involve a nucleophilic addition to an acyl iminium ion, a ring closing metathesis and a lactone-lactam rearrangement to provide the tricyclic structure.     

Total synthesis of malayamycin A and analogues

The total synthesis of the bicyclic C-nucleoside malayamycin A is described starting with d-ribonolactone. A new method was developed to obtain preparatively important quantities of β-pseudouridine, which was used as an intermediate. The synthesis of a carba N-nucleoside analogue of malayamycin A is also described.     

Asymmetric synthesis of 6-(2′,3′,4′,5′,6′-pentafluorophenyl)-δ-lactones via “allyl”boranes: application for the synthesis of fluorinated analog of key pharmacophore of statin drugs

Asymmetric “allyl”boration of pentafluorobenzaldehyde with various α-pinene based “allyl”boranes provides homoallylic alcohols in high de and ee; the alcohols have been converted into δ-lactones via acryloylation, ring-closing metathesis and hydrogenation. Pentafluorophenyl analog of key pharmacophore of statin drugs has been synthesized using diastereoselective epoxidation and regioselective reduction as key steps.     

Concise asymmetric synthesis of (−)-halosaline and (2R,9aR)-(+)-2-hydroxy-quinolizidine by ruthenium-catalyzed ring-rearrangement metathesis

A ruthenium-catalyzed ring opening-ring closing metathesis reaction serves as the key step in the stereoselective synthesis of a new enantiopure 2-substituted-4,5-dehydropiperidine skeleton, a valuable intermediate for the synthesis of piperidine alkaloids (such as (−)-halosaline) and of hydroxylated quinolizidines (such as (2R,9aR)-(+)-2-hydroxy-quinolizidine).     

A high-resolution MAS NMR study on the potential catalysts Mo/HBeta for olefin metathesis: The interaction of Mo species with HBeta zeolite

The interfacial interaction of Mo species with the HBeta zeolite was studied by multinuclear MAS NMR, XRD and N₂ adsorption. As proved by the quantitative ²⁷Al MAS NMR, this interaction is so strong as to dealuminate the framework of HBeta, and leads to a new peak appearing at −14 ppm, which indicates the formation of crystalline Al₂(MoO₄)₃. This can also be detected by XRD measurements when the Mo loading is as high as 9.0 wt.%. The corresponding quantitative ²⁹Si and ¹H MAS NMR spectra show that the amount of silanols and Brønsted acidic sites decrease obviously with increasing Mo loading. This also reveals an interaction between Mo species and HBeta support through an oxygen bridge resulting from condensation with the hydroxyls on the support. At higher Mo loadings, the interaction is so strong that it results in an extraction of aluminum from the zeolite framework, and subsequently appearance of Al₂(MoO₄)₃ and loss of Brønsted acidic sites. These can be correlated to the low catalytic activity of Mo/HBeta in metathesis of ethylene and 2-butylene to propylene.

从单核及双核二甲胺钼络合物出发,制备了SiO_2负载的钼催化剂。程序升温分解实验(TPDE)及红外测试表明,双核钼络合物与SiO_2作用生成双钼中心物种,此物种可催化乙烯的二聚反应。280℃活化后此双钼中心物种变成单钼中心物种,单钼物种只催化乙烯的同系化及歧化反应。     

Preparation of solutions of amidomagnesium chlorides in poly(ethylene oxide) and their characterization by conductivity measurements

Polymer electrolyte systems were prepared for the first time by dissolution of amidomagnesium chlorides in poly(ethylene oxide), (PEO). For the preparation, solutions of (hexamethyldisilylamido)magnesium chloride, (dimethylpyrrolyl)magnesium chloride, (diisopropylamido)magnesium chloride, piperidinomagnesium chloride and morpholinomagnesium chloride were chosen. The composition of these polymer electrolyte systems corresponds to the general formula R₂NMgCl·P(EO)_n·THF. Most work has been done with the system (hexamethyldisilylamido)magnesium chloride in PEO, (Me₃Si)₂NMgCl·P(EO)_n·THF, with n= 3, 4, 5, or 7. The electrolytes have a soft rubber-like consistency. At 30 °C, electrical conductivities of 10⁻⁶–10⁻⁵ S/cm were found. The conductivities were measured in the temperature range 20–60 °C. Within this temperature range a linear dependence of the logarithms of the conductivity on the inverse temperature was found and activation energies for the conducting process of 30–60 kJ/mol were calculated. Using those polymer electrolytes with a high content of the amidomagnesium compound, a reversible magnesium deposition takes place by cathodic reduction at potentials below −1.9 V vs. a Ag/AgCl reference electrode. These polymer electrolytes were found to be stable against oxidation up to about −0.3 V vs. Ag/AgCl. Electronic Publication     

LIGHT-SCATTERING STUDY OF POLYELECTROLYTE HOLLOW CAPSULES

Silver halide (AgX) microcrystal was used as template to synthesize hollow polyelectrolyte capsules. These hollow capsules were characterized by laser light scattering (LLS) used to measure the size of the capsules in solution. The ratio of hydrodynamic radius (Rh) from dynamic LLS to the radius of gyration (Rg) from static LLS is almost unity, revealing that the entities are hollow in solution. The results suggest that the LLS method can be regarded as a good complement to the confocal laser scanning microscopy (CLSM) method for the characterization of small hollow capsules, and it possesses the advantage of not needing fluorescence labeling.
