The extraction of Li+ with thenoyltrifluoroacetone (Htta) in the presence of 1,10-phenanthroline (phen) has been studied in various organic solvents. The remarkable enhancement of the extraction of Li+, that is a synergistic effect, was observed by the addition of phen, and the high extractability of Li+ was attained in toluene, benzene, chlorobenzene and o-dichlorobenzene. The extraction equilibrium of Li+, Na+ and K+ (denoted as M+) in the presence or absence of phen in chlorobenzene and the adduct formation reaction in the organic phase were studied in detail. The adduct of Li+ was Li(tta)(phen) in the wide concentration range of phen in the organic phase, while in Na+ and K+ M(tta)(phen)2 also exists in the high concentration region. The maximum value of the separation factor between Li+ and Na+ was observed in the present system and was larger than that in the Htta-trioctylphosphine oxide (TOPO)-benzene system reported previously. 相似文献

Summary A fully integrated chromatographic system was developed for the determination of leukotrienes in biological samples using
photodiode-array detection (PDAD), which eliminates time consuming manual sample handling steps. A special solid phase extraction,
(SPE) methodology for leucotriene metabolite stability was developed which increased the recoveries and eliminates the contamination
risk of biological samples. The inherent instability (autooxidation) of many of the leukotriene mediators, and the adsorption
effects onto exposed surfaces in vials and in the chromatographic system were found to be very important parameters to control
in order to circumvent high loss of sample analytes. By binding the cell supernatants to the functionalities of the SPE support
stabilised these mediators. Cell culture samples were eluted through a disposable C18 SPE column. The SPE columns were allowed to thaw and deposited in an automated sample handling unit (ASPEC XL). Desorption
of the analytes was followed by a second on-line SPE step, to eliminate remaining interfering matrix compounds. Typical recoveries
when stored at −70°C were in-between 55–97% except for (LTE4) which was found to be around 40% after 72 days of storage. Seven reversed-phase packings were studied. Selectivity factors,
as well as the separation efficiencies, were found to differ for the various C18 bonded silica stationary phases. This integrated on-line column liquid chromatographic system was applied to the determination
of leukotriene B4, leukotriene C4, leukotriene D4, leukotriene and E4 in human cell extracts using prostaglandin B2 as the internal standard. More than 1500 biological samples were analysed. Some validation data are presented for unattended

To determine the influence of various culture conditions, transformed cells of Escherichia coli expressing recombinant green fluorescent protein (GFPuv) were grown in nine cultures with four variable conditions (storage
of inoculated broth at 4°C prior to incubation, agitation speed, isopropyl-β-d-thiogalactopyranoside [IPTG] concentration, and induction time). The pelleted cells were resuspended in extraction buffer
and subjected to the three-phase partitioning (TPP) extraction method. To determine the most appropriate purification resin,
protein extracts were eluted through one of four types of HiTrap hydrophobic interaction chromatography (HIC) columns prepacked
with methyl, butyl, octyl, or phenyl resins and analyzed further on a 12% sodium dodecylsulfatepolyacrylamidegel. With Coomassie
staining, a single band between 27 (standard GFPuv) and 29 kDa (molecular weight standard) was visualized for every HIC column
sample. TPP extraction with HIC elution provided about 90% of the GFPuv recovered and eight-fold GFPuv enrichment related
to the specific mass. Rotary speed and IPTG concentration showed, respectively, greater negative and positive influences on
GFPuv expression at the beginning of the logarithmic phase for the set culture conditions (37°C, 24-h incubation). 相似文献

The potential of micellar electrokinetic capillary chromatography (MEKC) with UV-detection for the simultaneous determination of various selected pesticides in water samples was investigated. The developed method using solid-phase extraction showed high efficiency and good resolution with detection limits in the 0.2 to 0.5 ppm range. Comparison of several SPE cartridges demonstrates their suitability for the extraction of pesticides with different hydrophobicity achieving 5000-fold enrichment. The described method involving SPE procedure and MEKC separation enables the successful determination of a wide spectrum of pesticides in water in the range of maximum residue limits (MRLs). 相似文献

A simple and versatile system is described for the on-line coupling of SFE to capillary GC. The interfacing consists of a programmed temperature vaporizer (PTV) injector. With this injector it is possible to combine solute trapping, elimination of a high flow of extraction fluid, and quantitative transfer of solutes to the seperation column. The problems caused by impurities in the extraction fluid in on-line SFE-GC are discussed. Simple methods are described for the purification of commercially available carbon dioxide. The trapping efficiency of the PTV injector is studied. Applications of the SFE-PTV-GC system are given for the analysis of polymer anti-degradants, polar compounds, and samples with environmental relevance. 相似文献

A novel extraction method, namely dynamic ultrasound-assisted extraction, is investigated. This technique is efficient with respect to both time and solvent consumption because it utilizes ultrasonic energy in dynamic mode during extraction. Polyphenols (chlorogenic acid, esculetin, rutin, scopoletin, and quercitrin) are extracted from a tobacco (Nicotina tobaccum L.) sample for 10 min with 6 mL of solvent. Fresh solvent is continuously pumped through the sample, with which the analytes can be rapidly extracted, and the possibility of degradation efficiently avoided. Methanol involving 0.5% w/v ascorbic acid was used as extraction solvent; optimal flow rate and extraction time were investigated. The extract was cleaned up by C18 disposable cartridge. The spiked and nonspiked tobacco samples were used for the evaluation of the proposed method. Recoveries obtained were varied from 96 to 108% and RSDs from 2.0 to 4.6%. This extraction technique was revealed to recover larger amounts of polyphenols from tobacco, compared to the static ultrasound-assisted extraction method. 相似文献
