This study outlines antioxidant and anti-AChE activities of the polysaccharide (PSH) extract from the mushroom species Amanita strobiliformis. Both the presence of α and ß glucans within the aforementioned extract was recorded. PSH extract displayed a profound scavenging activity of OH radicals (IC50 value, 11.86 ± 0.59 μg/mL) and high potential for reduction of Fe3+ ions (174.11 ± 8.70 mg eq. AA/g d.w.) being almost 48- and 5-fold more effective than mannitol and butylated hydroxytoluene used as a positive control, respectively. Compared with galanthamine (0.001 μg), the same extract exhibited a moderate anti-AChE activity (10 μg) in solid. Since purified PSH extract exhibited higher bioactivity (IC50 value 7.27 ± 0.31 μg/mL, 197.68 ± 9.47 mg eq. AA/g d.w. and 0.1 μg, respectively), it can be predominantly ascribed to the polysaccharide compounds. A. strobiliformis PSH extract may be considered as a promising resource of potent bioactive polysaccharides of natural origin successfully addressing both oxidative stress and lack of acetylcholine.  相似文献

Corn silk is a well‐known traditional Chinese medicine that has been widely used for its antidiabetic, antioxidant, antihyperlipidemic, and other effects in China for thousands of years. Numerous studies have revealed that corn silk contains multiple bioactive constituents that are beneficial for human health. However, the constituents of corn silk in vivo remain ambiguous. In this study, high‐throughput ultra‐high‐performance liquid chromatography combined with quadrupole time‐of‐flight mass spectrometry technology using multivariate statistical analysis was established to systematically investigate the constituents migrating into blood from corn silk aqueous extract. As a result, 76 compounds were identified, including caffeic acid and ten of its derivatives, (E)‐p‐coumaric acid and two of its derivatives, ferulic acid and four of its derivatives, and five flavones. Among the identified constituents, 21 constituents, including nine prototype components and 12 metabolites derived from eight components, were characterized in sequence. Based on the significance of the results, the applied approach was powerful for the accurate determination and rapid screening of bioactive components from corn silk aqueous extract. The obtained results are valuable for the in‐depth understanding and further pharmacological study of corn silk aqueous extract.  相似文献

The extracts of Schisandra chinensis (Turcz.) Baill. (Schisandraceae) have various therapeutic effects, including inflammation and allergy. In this study, gomisin M2 (GM2) was isolated from S. chinensis and its beneficial effects were assessed against atopic dermatitis (AD). We evaluated the therapeutic effects of GM2 on 2,4-dinitrochlorobenzene (DNCB) and Dermatophagoides farinae extract (DFE)-induced AD-like skin lesions with BALB/c mice ears and within the tumor necrosis factor (TNF)-α and interferon (IFN)-γ-stimulated keratinocytes. The oral administration of GM2 resulted in reduced epidermal and dermal thickness, infiltration of tissue eosinophils, mast cells, and helper T cells in AD-like lesions. GM2 suppressed the expression of IL-1β, IL-4, IL-5, IL-6, IL-12a, and TSLP in ear tissue and the expression of IFN-γ, IL-4, and IL-17A in auricular lymph nodes. GM2 also inhibited STAT1 and NF-κB phosphorylation in DNCB/DFE-induced AD-like lesions. The oral administration of GM2 reduced levels of IgE (DFE-specific and total) and IgG2a in the mice sera, as well as protein levels of IL-4, IL-6, and TSLP in ear tissues. In TNF-α/IFN-γ-stimulated keratinocytes, GM2 significantly inhibited IL-1β, IL-6, CXCL8, and CCL22 through the suppression of STAT1 phosphorylation and the nuclear translocation of NF-κB. Taken together, these results indicate that GM2 is a biologically active compound that exhibits inhibitory effects on skin inflammation and suggests that GM2 might serve as a remedy in inflammatory skin diseases, specifically on AD.  相似文献

This study aimed to investigate the chemical composition of Tribulus terrestris L. fruit (TT) extract named TT15 and its protective effect against ischemic stroke (IS) as well as corresponding mechanisms. The chemical composition of TT15 was analyzed by liquid chromatography-mass spectrometry (LC-MS), and the compound identification was conducted via searching the in-house database. The LC-MS-based multi-omics approach was applied to search the differential metabolites and differential proteins in rat brain tissue and to explore the biomarker and molecular mechanism of TT15 against middle cerebral artery occlusion (MCAO). A total of 20 compounds were identified from TT15, mainly including alkaloids, flavonoids, phenols, quinones, and esters. These 20 compounds significantly affected the metabolism of 44 metabolites and the expression of 51 proteins. Joint pathway analysis showed that these metabolites and proteins were mainly involved in the response to elevated platelet cytosolic Ca2+ and platelet activation, which inferred that TT15 may exert a protective effect against cerebral ischemic injury via regulating platelet function. This study provides useful information for further exploration of the mechanisms of TT extract against IS.  相似文献

A rapid and useful approach for screening potential bioactive components in Ginkgo biloba extract (GBE) with preventive effect against diabetic nephropathy (DN) was developed using mesangial cells extraction coupled with high‐performance liquid chromatography tandem mass spectrometry (LC‐MS/MS) analysis. Mesangial cells were first divided into two groups according to their treatments with high glucose or high glucose plus GBE. After incubation for 4, 8, 12, 16, 24 and 48 h, the cells were harvested and extracted with 40% acetic acid in water before LC‐MS/MS analysis. Then, 19 compounds and five metabolites were found to selectively combine with mesangial cells. Notably, compounds including quercetin and rutin were identified or tentatively characterized according to the results of retention time and MS spectra, which is highly consistent with our previous reports that quercetin and rutin are potent protective agents against glomerulosclerosis in DN. Therefore, all these results indicate that target cell extraction coupled with LC‐MS/MS analysis can be successfully applied for predicting the bioactive components in GBE with preventive effect against DN. Blossoms of Inula nervosa Wall. (BINW) are traditionally used as an analgesic and antitussive in China. In this study, in vitro anticomplementary activities of crude extract from BINW in 21 batches and of extracts of four monomeric compounds were evaluated by the classical pathway. The effect of the region of origin on the quality of BINW was evaluated by fingerprint analysis for the first time. Furthermore, chemometric methods including similarity analysis and principal component analysis were employed to evaluate the quality of BINW. The nine major monomeric compounds were quantitated by ultra‐high‐performance liquid chromatography. All nine analytes demonstrated excellent linearity with recoveries ranging from 97.25% to 102.76%. The limits of detection and quantification were 0.07–12.20 μg/mL and 0.22–40.27 μg/mL, respectively. Results indicate that different regions of origin have a significant effect on the quality of BINW. Fingerprint analysis in combination with chemometrics and multi‐ingredient determination is an efficient and reliable approach for quality evaluation. The BINW samples from Yunnan had the highest ratio of 1,5‐dicaffeoylquinic acid and thymol; they also exhibited significantly higher anticomplementary activity than those from three other areas. This research aimed to study the Maillard reaction pathway in chicken meat. Owing to the complexity of real chicken meat, which is composed of many different types of amino acids and reducing sugars, the experiment was initiated with a glucose/lysine model system with the same concentration ratio of reactants as found in chicken meat. By considering glucose as the rate-limiting substrate, a kinetic model of the glucose/lysine model system was developed. Methylglyoxal (MG) was found to be the principal important α-dicarbonyl compound intermediates that further reacted to form melanoidins. Pyridine was a major volatile compound in this model system. The optimized kinetic model was then further validated in a chicken extract, for which the Maillard reaction mechanism has not been elucidated. However, the kinetic model of the glucose/lysine system could not explain the Maillard reaction in the chicken extract, presumably because both types of intermediates and reaction pathway depend on the reactants. Thus, a kinetic model of the Maillard reaction in the chicken extract was developed based on the main types of detected intermediates. Overall, MG was the central intermediate and acted as a substrate for the formation of furfural, volatile compounds, melanoidins, and unknown carbonyl compound(s) (Cn). Pyrazines and aldehydes were the major volatile compounds in the chicken extract.  相似文献

ABSTRACT

The asymmetric cross-aldol reaction of simple ketones (acetone, cyclohexanone) with isatin derivatives in the presence of crude extract from earthworms as green and effective biocatalyst proceeds easily in MeCN/H2O (1:1) as solvent to afford 3-hydroxy-2-oxindoles derivatives. Ten compounds were synthesized in high yields (62–88%) and moderate ee (29–42%). Structure of the synthesized compounds has been characterized on the basis of NMR spectra and CHN analysis. The ee of the obtained compounds was determined by chiral phase HPLC analysis.  相似文献

The aim of the study was the rapid green synthesis of titanium nanoparticles using the aqueous extract of Falcaria vulgaris leaves (TiNPs@FV) and exploring their antioxidant, cytotoxicity, antifungal, antibacterial, and cutaneous wound healing activities under in vitro and in vivo condition. These nanoparticles were characterized by UV-Vis, Fourier transform-infrared(FT-IR), X-ray diffraction XRD), field emission-scanning electron microscopy FE-SEM), and transmission electron microscopy TEM) analyses. The synthesized TiNPs@FV had great cell viability on human umbilical vein endothelial cells and indicted this method was nontoxic. DPPH (2,2-diphenyl-1-picrylhydrazyl) test revealed similar antioxidant potentials for F. vulgaris, TiNPs@FV, and butylated hydroxytoluene. All data of antibacterial, antifungal, and cutaneous wound healing tests were analyzed by SPSS 22 software. In the antimicrobial part of this study, TiNPs@FV indicated higher antifungal and antibacterial effects than all standard antibiotics (p ≤ 0.01). Minimal inhibitory concentration (MIC) and minimal fungicidal concentration of TiNPs@FV against all fungi were at 2–4 mg/mL and 2-8 mg/mL ranges, respectively. But, MIC and minimal bactericidal concentration of TiNPs@FV against all bacteria were at 2-8 mg/mL and 2-16 mg/mL ranges, respectively. In the part of cutaneous wound healing, use of TiNPs@FV ointment significantly (p ≤ 0.01) raised the wound contracture, vessel, hydroxyl proline, hexuronic acid, hexosamine, fibrocyte, and fibrocytes/fibroblast rate and significantly (p ≤ 0.01) decreased the wound area, total cells, neutrophil, and lymphocyte compared to other groups in rats. The results of FT-IR, UV-Vis, XRD, TEM, and FE-SEM confirm that the aqueous extract of F. vulgaris leaves can be used to yield titanium nanoparticles with a notable amount of remedial effects.  相似文献

Cellulose/silver nanoparticle composite films with in situ-generated silver nanoparticles (AgNPs) were prepared using Ocimum sanctum leaf extract as a reducing agent in the absence and presence of sunlight and were characterized by SEM, FTIR, XRD, and antibacterial tests. Sunlight hastened up the preparation of these composite films. The average size of the in situ-generated AgNPs was reduced by the sunlight. The antibacterial activity and other properties of the composites were enhanced by the sunlight. The cellulose/AgNP composite films with improved properties by sunlight can be considered for medical purpose as antibacterial dressing materials.  相似文献
