We investigated the incorporation efficiencies of different fluorescently labelled dNTPs with polymerases by complementary strand synthesis. For this reason single stranded DNA was immobilized on a coverslip and the increase of fluorescence due to the synthesis of the corresponding strand with tagged dNTPs was detected with a supercritical angle fluorescence biosensor in real-time. By comparison of the observed signal intensities it was possible to conclude that the system Cy5-dCTP-Klenow (exonuclease free) fragment gives the best incorporation yield of the investigated enzymes and dNTPs. 相似文献

The effect of annealing condition on the microstructure evolution, together with mechanical properties of Fe-28.5Ni steel processed by one and six cycles of accumulative roll bonding (ARB), was explored. The evolution of microstructure was studied by means of Electron Backscattered Diffraction method. The ARB-processed specimens were annealed for 30 minutes between 500°C and 600°C, and the effect of these annealing processes was elaborated. Results showed a significantly reduced martensite phase during ARB process cycles. Moreover, a refined and stabilised ultrafine structure was obtained in 6-cycle ARB-processed specimen. Results also show that by annealing the 1-cycle specimen at 550°C, no significant microstructural change and mechanical properties variation are identified. As such, no significant microstructural change and mechanical properties variation were notified for annealing of 6-cycle ARB-processed specimen at 500°C. However, annealing at 600°C in both conditions led to a complete recrystallisation of the deformed structure. Annealing at 550°C in 6-cycle ARB-processed specimen showed partial recrystallisation as well as an abnormal grain growth characteristic. The abnormal grain growth was seen by annealing of 1-cycle ARB-processed specimen at 600°C. The changing features of dislocations were discussed in these specimens in terms of changing the dynamics of low- and high-angle grain boundaries. Tensile test results also showed a significant increase in the yield/ultimate tensile strengths with the application of ARB cycles. Although ARB process led to a rapid reduction of total elongation, full restoration during high-temperature annealing returned this value back to the initial, non-deformed, condition. 相似文献

ABSTRACT Ausforming is being widely used to overcome the kinetics barrier in nanostructured bainitic steels and to enhance its practical industrial applications. It would also affect the microstructural characteristics and consequently the resultant mechanical performance. This article aims to investigate the tensile behaviour of nano bainite obtained from plastically deformed primary austenite at three different heat treatment stages. 10% of ausforming has been implemented and nanostructured bainite obtained after austempering at 300°C for 2, 4 and 6?h in salt bath furnaces. Results indicated that ausforming could successfully increase the volume fraction of bainite within the microstructure and refine the bainite packets sizes. Moreover, it has been demonstrated that higher mechanical stability of retained austenite and therefore more effective TRIP effect could be attained during the tensile test which consequently resulted in elongation improvement without deteriorating the strength properties. The elongation method with intermediate mechanical and electrostatic embedding (ELG-IMEE) is proposed. The electrostatic embedding uses atomic charges generated by a charge sensitivity analysis (CSA) method and parameterized for three different population analyses, namely, the Merz–Singh–Kollman scheme, the charge model 5, and the atomic polar tensor. The obtained CSA models were tested on two model systems. Test calculations show that the electrostatic embedding provides several times of decrease in the difference of energies of testing and reference calculations in comparison with the conventional elongation approach (ELG). The mechanical embedding is implemented in a combination of the conventional elongation method and the ONIOM approach. Moreover, it was demonstrated that the geometry optimization with the ELG-IMEE reduces the errors in the optimized structures by about one order in root-mean-square deviation, when compared to ELG. 相似文献

Allelopathic materials inside a tree can produce positive or negative change in the survival, growth, reproduction and behaviour of other organisms if they escape into the environment. To assess these effects, this work was carried out to evaluate the allelopathic impact of Eucalyptus erythrocorys L. on seed germination and seedling growth of two weeds: Sinapis arvensis L. and Phalaris canariensis L.; on one cultivated crop: Triticum durum L. Aqueous; and on ethanolic leaf extracts of E. erythrocorys L. The study was effected using four concentrations (10, 20, 25 and 30 μL/mL) while distilled water was used as a control. The results showed that the E. erythrocorys L. crude extracts had an inhibitory effect on seed germination and seedling growth of both studied weeds and wheat. The inhibition rate was increased by the increase in extract concentration. Only ethanolic extracts of E. erythrocorys L. induced a significant inhibition of seed germination of durum wheat. The effect of E. erythrocorys L. crude extracts was more severe on weeds than on durum wheat. These results indicate that the seedling growth, especially radicle elongation, was the more sensitive indicator to evaluate the effects of extracts than was the seed germination. 相似文献

The sequence-specific recognitions between DNA and proteins are playing important roles in many biological functions. The double-stranded DNA microarrays (dsDNA microarrays) can be used to study the sequence-specific recognitions between DNAs and proteins in highly parallel way. In this paper, two different elongation processes in forming dsDNA from the immobilized oligonucleotides have been compared in order to optimize the fabrication of dsDNA microarrays: (1) elongation from the hairpins formed by the self-hybridized oligonucleatides spotted on a glass; (2) elongation from the complementary primers hybridized on the spotted oligonucleatides. The results suggested that the dsDNA probes density produced by the hybridized-primer extension was about four times lower than those by the self-hybridized hairpins. Meanwhile, in order to reduce the cost of dsDNA microarrays, we have replaced the Klenow DNA polymerase with Taq DNA polymerase, and optimized the reaction conditions of on-chip elongation. Our experiements showed that the elongation temperature of 50 °C and the Mg2+ concentration of 2.5 mM are the optimized conditions in elongation with Taq DNA polymerase. A dsDNA microarray has been successfully constructed with the above method to detect NF-kB protein. 相似文献

The transition state of addition of free radicals and atoms to multiple bonds is considered as a result of intersecting of two parabolic potential curves. One of them characterizes the stretching vibration of the attacked multiple bond, and another curve characterizes the stretching vibration of the bond formed in the transition state. The force constant of the latter is calculated by an empirical equation that correlates the force constant with the bond dissociation energy. In the framework of this model, the thermally neutral activation energy (E
e0) and the elongation of the attacked and formed bonds (r
e) in the transition state were calculated from the experimental data (activation energy (E
e) and enthalpy of reaction (H

Summary: Recycled PET as well as ABS - organomodified montmorillonite nanocomposites were prepared via melt compounding in a counter-rotating twin screw extruder. The topological changes in polymer matrices as dependency on clay modification have been evaluated from dynamic experiments in the shear flow using low amplitude oscillatory measurements. Flow characteristics of all studied organoclay nanocomposites showed shear-thinning behavior at low frequencies. Filling of PET with some organoclays led to degradation reactions, which were reflected by lower magnitudes of viscosity and storage modulus in the range of higher frequencies as compared to unfilled polymer matrix. On the contrary, no degradation during the processing of different organoclays with recycled ABS has been observed. 相似文献

Cultivation of myxobacteria of the Nannocystis genus led to the isolation and structure elucidation of a class of novel cyclic lactone inhibitors of elongation factor 1. Whole genome sequence analysis and annotation enabled identification of the putative biosynthetic cluster and synthesis process. In biological assays the compounds displayed anti‐fungal and cytotoxic activity. Combined genetic and proteomic approaches identified the eukaryotic translation elongation factor 1α (EF‐1α) as the primary target for this compound class. Nannocystin A ( 1 ) displayed differential activity across various cancer cell lines and EEF1A1 expression levels appear to be the main differentiating factor. Biochemical and genetic evidence support an overlapping binding site of 1 with the anti‐cancer compound didemnin B on EF‐1α. This myxobacterial chemotype thus offers an interesting starting point for further investigations of the potential of therapeutics targeting elongation factor 1. 相似文献
