This work describes a simple, versatile solid-phase peptide-synthesis (SPPS) method for preparing micelle-forming poly(ethylene oxide)-block-peptide block copolymers for drug delivery. To demonstrate its utility, this SPPS method was used to construct two series of micelle-forming block copolymers (one of constant core-composition and variable length; the other of constant core length and variable composition). The block copolymers were then used to study in detail the effect of size and composition on micellization. The various block copolymers were prepared by a combination of SPPS for the peptide block, followed by solution–phase conjugation of the peptide block with a proprionic acid derivative of poly(ethylene oxide) (PEO) to form the PEO-b-peptide block copolymer. The composition of each block component was characterized by mass spectrometry (MALDI and ES-MS). Block copolymer compositions were characterized by 1H NMR. All the block copolymers were found to form micelles as judged by transmission electron microscopy (TEM) and light scattering analysis. To demonstrate their potential as drug delivery systems, micelles prepared from one member of the PEO-b-peptide block copolymer series were physically loaded with the anticancer drug doxorubicin (DOX). Micelle static and dynamic stability were found to correlate strongly with micelle core length. In contrast, these same micellization properties appear to be a complex function of core composition, and no clear trends could be identified from among the set of compositionally varying, fixed length block copolymer micelles. We conclude that SPPS can be used to construct biocompatible block copolymers with well-defined core lengths and compositions, which in turn can be used to study and to tailor the behavior of block copolymer micelles.  相似文献

The equilibrium in the systems containing a polymer based on dibenzo-18-crown-6 and binary or multicomponent aqueous solutions of NaNO3, KNO3, RbNO3, and AgNO3 was studied. The constants of the sorption of the individual electrolytes from solution into the polymer were determined. 本文研究了丙烯酸酯共聚物载体对驱蚊剂N,N-二乙基间甲苯酰胺(DETA)慢释放行为的控制,探讨了单体配比、共聚物分子量以及DETA含量对释放速率的影响。结果表明。增加亲水性单体丙烯酸(AA),可提高DETA的释放速率;降低共聚物数均分子量,会减少DETA的释放量;剂型中DETA含量与释放量存在线性正比关系,但DETA的含量增加并不导致释放有效时间的线性增加。本文还就共聚物膜的透湿性和碱溶性作了研究,亲水性单体AA的增加同样导致透湿性和碱溶性的增加。制备的聚合物驱蚊剂经现场应用试验,表明具有良好的长效驱蚊特性。  相似文献

The retention behavior of lymphocyte subpopulations, B cell, T cell and null cell, derived from rat spleen to polyamine-graft-poly(2-hydroxyethyl methacrylate) copolymer (HA) surface was investigated, focusing on the conformational transition of the polyamine side chain as well as the protonation of amino groups in the polyamine grafts. Furthermore, the availability of HA was discussed as a column adsorbent for separation of lymphocyte subpopulations derived from spleen. The conformational transition of polyamine grafts significantly influenced the mode of retention of lymphocyte subpopulations. When polyamine grafts existed in an aggregated conformation (protonatin degree α < 0.5), the retention of lymphocyte subpopulations was decreased in the order B cell> null cell> T cell. On the other hand, when polyamine existed in an extended conformation into the aqueous interior from the matrix interface (α > 0.5), T cell retention became greater than null cell retention, resulting in a decreased B cell> T cell> null cell order. These results indicate that the differential retention of spleen lymphocyte subpopulations is attributed to their differential responses to the change in matrix interface accompanied by the protonation of amino groups. Furthermore, spleen lymphocytes were compared with lymph node lymphocytes in terms of resolution efficacy by an HA copolymer column.  相似文献

Ethylene/ethane sorption characteristics were determined for dry Pebax™ (poly(amide 12-block-tetramethylenoxide) copolymer)/AgBF4 membranes by using an electronic microbalance. The membranes containing 0.7 and 22 wt.% AgBF4 showed a dual-mode sorption isotherm. The ethane isotherms for all the membranes were of the Henry-type, which is the normal sorption for gases in rubbery polymers. The abnormal presence of Langmuir sorption sites only for ethylene in the rubbery copolymer, never reported sofar, is attributed to the silver-based specific complexation sites. The silver salt which dissolved in limited amounts in the rubbery copolymer had a much smaller Langmuir sorption capacity than the salt that crystallized in the copolymer. The sorption kinetics indicate that the crystallized salt did adsorb slowly ethylene according to a zeroth-order kinetics, but not ethane. The gas uptake kinetics resulting from a step of the pressure surrounding the copolymer exhibited one stage for ethane but two stages for ethylene. For the latter, there was first a fast Fickian sorption stage, then a drift of the zeroth-order sorption of ethylene on salt crystals, which contributes for a large part to the total uptake. The zeroth-order sorption suggests that the sorbed ethylene amount in the second-stage is independent of the crystal-surface coverage. The value of the Fickian diffusion coefficient calculated by fitting the kinetics with a solution of the second Fick’s law was 5 × 10−12 m2/s for both ethylene (the first stage) and ethane, and is typical for small organic compounds in a rubbery material.  相似文献

The long-term environmental ageing of conductive composite films containing ethylene-co-vinyl-acetate (EVA) copolymer and a complex of polyaniline (PANI) and dodecylbenzenesulfonic acid (DBSA) was studied by using differential scanning calorimetry (DSC). We assume that both phase separation and crosslinking of PANI main chains occur in the systems. On the other hand, the competition between PANI–DBSA complex self-organization and crystallization of EVA matrix result in structural changes and formation of continuous conductive network, responsible for significantly increased (ca five orders of magnitude) electrical conductivity of the aged films. Ethylene and different amounts of 1,7-octadiene were copolymerized using the metallocene catalyst system ethylidene-bis(fluorenyl) zirconium dichloride and methylaluminoxane (MAO) at both 50 and 90 °C. The catalyst activity has slightly increased with the addition of low amounts of the diene in relation to the homopolymerization of ethylene. The obtained polymers were characterized according to their melting temperature (Tm) and crystallinity degree (xc) by differential scanning calorimetry (DSC). Weight-average molecular weight (Mw) and polydispersity were determined by gel permeation chromatography (GPC). Diene contents in the copolymer were obtained through the FTIR spectroscopy. The results indicated that at polymerization temperature of 90 °C, crosslinking bonds in the obtained copolymers were low, differently from what was observed at 50 °C. The diene content in the copolymer achieved more than 3 mol% and the comonomer conversion was around 15%. Moreover, the obtained copolymers have Mw around 100,000 and large polydispersity.  相似文献

The impact properties of two selected metallocene-catalyzed ethylene-butene copolymers and one conventionalcopolymer were evaluated using Izod impact test. It is found that the metallocene-catalyzed copolymer shows superior impactproperties. This result was explained on the basis of the more homogeneous inter-molecular composition distribution andnarrower molecular weight distribution, which leads to more homogeneous morphology with fewer defects. Stepwisecrystallization improves the impact properties, especially in the crack propagation process, to a large extent. This is due to thedecrease of entanglements by stepwise crystallization, which is advantageous for the chain slip and shear. The polymer withheterogeneous intra-molecular composition distribution exhibits a more evident improvement of impact properties understepwise crystallization.  相似文献

This paper mainly deals with the design and synthesis of a novel styrene-butadieneblock copolymer. When this copolymer is used in the tread portion of tyres, it can improvewet skid resistance and reduce rolling resistance without sacrificing its general physical-mechanical properties. The visco-elastic curve of tire tread using the novel copolymer asits rubber portion was showed. Reactivity ratios for two monomers in the polymerizingsystem were calculated. The diagrams of differelitial, integral and finite difference calculithroughout the whole molecular chain were presented.
