Inhibition of bacterial virulence is believed to be a new treatment option for bacterial infections. In the present study, we tested dipicolylamine (DPA), tripicolylamine (TPA), tris pyridine ethylene diamine (TPED), pyridine and thiophene derivatives as putative inhibitors of the bacterial virulence factors thermolysin (TLN), pseudolysin (PLN) and aureolysin (ALN) and the human zinc metalloproteases, matrix metalloprotease-9 (MMP-9) and matrix metalloprotease-14 (MMP-14). These compounds have nitrogen or sulfur as putative donor atoms for zinc chelation. In general, the compounds showed stronger inhibition of MMP-14 and PLN than of the other enzymes, with Ki values in the lower μM range. Except for DPA, none of the compounds showed significantly stronger inhibition of the virulence factors than of the human zinc metalloproteases. TPA and Zn230 were the only compounds that inhibited all five zinc metalloproteinases with a Ki value in the lower μM range. The thiophene compounds gave weak or no inhibition. Docking indicated that some of the compounds coordinated zinc by one oxygen atom from a hydroxyl or carbonyl group, or by oxygen atoms both from a hydroxyl group and a carbonyl group, and not by pyridine nitrogen as in DPA and TPA.  相似文献

Electrochemical sensors to quantify concentrations of emerging pollutants have attracted great attention from the industry and scientific community. Nanomaterials such as carbon black have been applied in sensors to identify substances that are toxic to the environment and human health due to their excellent electroanalytical properties. The aim of the study was to develop a novel electrochemical sensor for the endocrine disruptor hormone determination. To our knowledge, for the first time the synthesis of material based on carbon black containing immobilized palladium nanoparticles, with the application for the hormone ethinylestradiol, is reported in the literature. The material was synthesized, characterized, and applied to the determination in tap water and human urine of the synthetic hormone 17α-ethinylestradiol (EE2), which is currently considered an emerging pollutant. The morphology, structure and electrochemical performance of the sensors were characterized by scanning electron microscopy (SEM) and cyclic voltammetry (CV). Differential pulse voltammetry (DPV) in sodium phosphate buffer solution at pH 5.0 allowed the generation of a method to quantify the concentration of 17α-ethinylestradiol in a linear range of 0.5–119.0 μmol L−1, obtaining 81.0 nmol L−1 of calculated limit of detection (LOD). The system was efficient in detecting 17α-ethinylestradiol in real urine samples and showed no interferences for ascorbic acid, uric acid, progesterone, and dopamine. It is noteworthy that the results obtained showed good recovery values, considering that the urine samples were not previously treated or pre-concentrated, which suggests the development of an electrochemical sensor that works in situ and in real time to monitor relevant substances in the control clinical and environmental, with the possibility of point-of-care analyses.  相似文献

目的探讨不同激光美容治疗技术的临床应用效果。方法选取2014年1月—2016年9月期间温州医科大学附属眼视光医院收治的48例色素暗沉病症和良性皮肤增生相关病症患者作为研究对象,随机分为对照组和观察组,各24例。两组患者均根据美容需求制定YAG激光治疗方案,对照组单纯采用YAG激光(Q一Switehing Nd:YAGLaser Deviee)技术治疗,观察组在对照组基础上增加强脉冲光治疗,观察两组患者治疗1个月后临床疗效、皮肤损伤程度及损伤愈合时间。结果观察组患者激光美容治疗,治疗总有效率为95.83%(23/24),显著高于对照组(79.17%,19/24),组间差异有统计学意义(P0.05)。观察组皮肤损伤愈合时间(11.36±5.69)d显著低于对照组(15.23±6.58)d,组间差异有统计学意义(P0.05)。观察组和对照组患者两组皮肤损伤程度比较,无显著差异P0.05,无统计学意义。结论 YAG激光联合强脉冲光技术应用于临床医学美容治疗效果较好,且缩短了患者恢复时间,该激光美容治疗技术临床应用可靠。  相似文献

目的探讨分析PPH(吻合器痔上黏膜环切术)术治疗痔疮合并黏膜内脱垂的临床效果。方法实验时间2014年11月至2016年11月,实验对象为在广东省茂名市茂南区金塘卫生院接受治疗的痔疮合并黏膜内脱垂患者110例,随机分为观察组以及对照组,每组55例,对照组给予混合痔外剥内扎术治疗,观察组给予PPH术治疗,实验结束之后,对两组患者的手术时间、术后恢复时间、住院时间以及并发症的发生率进行对比分析。结果通过比较发现,观察组患者的上述指标与对照组相比差异具有统计学意义(P0.05)。结论 PPH术治疗痔疮合并黏膜内脱垂的疗效显著,可有效的降低并发症的发生,提高患者的生活质量,值得临床推广应用。  相似文献

以绿色廉价的天然椰果细菌纤维素(BC)为造孔剂,采用溶胶凝胶法制备了CuCe_(0.75)Zr_(0.25)O_x复合氧化物催化剂,通过TG/DTG、N2低温物理吸脱附、XRD、H_2-TPR、O_2-TPD和Raman等手段对催化剂进行了表征,并对其在固定床上挥发性有机物(VOCs)降解的催化性能进行了研究。结果表明,利用BC精细的纤维网状结构和亲水性能与活性金属盐溶液形成凝胶,可有效制备介孔结构的复合氧化物催化剂。制备过程中,凝胶形式和成胶温度对催化剂降解甲苯的活性有较大影响;采用醇凝胶形式在70℃时制备的ACCZ-70催化剂完全降解甲苯的温度为205℃,明显低于已有文献报道的催化剂,这主要归因于该催化剂具有良好的低温还原性和高达0.81的氧空穴浓度。而采用水凝胶制备的催化剂降解甲苯时,在120-140℃存在吸附现象。  相似文献

Current treatments for brain aneurysms are invasive, traumatic, and not suitable in most patients with increased risks. A new alternative method is using scaffold stents to create a local and focal attraction force of cells for an in situ reconstruction of the tunica media. For this purpose, a nanostructured bioactive coating is designed to render an asymmetric region of the stent scaffold magnetic and biomimetic, which utilizes bacterial nanocellulose (BNC) as a platform for both magnetic and cell attraction as well as proliferation. The magnetization of the BNC is realized through the reaction of Fe III and II, precipitating superparamagnetic iron oxide nanoparticles (SPION). Subsequently, magnetic bacterial nanocellulose (MBNC) is coated with polyethylene glycol to improve its biocompatibility. Cytotoxicity and biocompatibility are evaluated using porcine aortic smooth muscle cells. Preliminary cellular migration assays demonstrate the behavior between MBNC and cells labeled with SPION. In this work, (1) synthesis of BNC impregnated with magnetic nanoparticles is successfully demonstrated; (2) a viable, resilient, and biocompatible hydrogel membrane is tested for neuroendovascular application using a stent scaffold; (3) cell viability and minimal cytotoxicity is achieved; (4) cell migration tests and examination of cellular magnetic attraction confirm the viability of MBNC as a multifunctional coating.

考察了酪蛋白酸钠(sodium caseinate,SC)和乳清分离蛋白(whey protein isolate,WPI)在表面性质不同的3种羟基磷灰石(hydroxyapatite,HA)颗粒上的界面吸附,分析了蛋白质的分子构型和HA颗粒的表面性质等因素对蛋白质在HA界面吸附的影响,重点讨论了SC和WPI肽链上磷酸化丝氨酸基团(phosphorylated serine residues,Ser-P)的数量和分布对吸附差异的影响.通过傅里叶变换红外光谱和表面电位分析发现SC和WPI无法被比表面积较小的HA颗粒有效吸附,但是在有效吸附面积较高的球状纳米HA和棒状微米HA上能够被吸附.Ser-P的存在使得SC在HA界面的吸附量更高、吸附能力更强.Ser-P数量和分布的不同则导致了SC中不同的蛋白组分在HA界面的竞争性吸附:β-酪蛋白在2μmHA界面始终存在优先吸附性;当纳米HA的浓度低于15 mg/mL时,纳米HA界面会优先吸附αs-酪蛋白.  相似文献

组蛋白翻译后修饰(HPTMs)参与基因转录调控,其异常与肿瘤等重大疾病的发生发展密切相关。石蜡包埋组织是当前疾病研究的重要样本资源,对肿瘤机制和标志物研究具有重要意义。目前基于质谱的蛋白质组学技术已成为HPTMs分析的有力工具,而针对福尔马林固定石蜡包埋(FFPE)组织样品的HPTMs分析还十分有限。该研究发展了一种基于高效液相色谱-串联质谱的FFPE组织样本HPTMs分离分析新方法。通过研究并优化组蛋白的提取策略,建立了FFPE组织样本脱蜡水化处理、蛋白质提取与聚丙烯酰胺凝胶电泳分离相结合的组蛋白提取和分离方法。通过研究FFPE切片数量、组蛋白化学衍生化方法等对组蛋白鉴定的影响,确定了组蛋白处理的具体步骤。通过HPLC分离结合非依赖性采集模式的质谱分析,鉴定了组蛋白修饰的类型、位点和丰度。最后,将优化的实验方法应用于FFPE临床样本的HPTMs分析,鉴定了2例人乳腺浸润性癌和癌旁正常组织的HPTMs图谱,均获得了100种以上的不同组蛋白修饰形式的多肽。定量分析了他们的差异性水平,通过主成分降维分析,发现浸润性癌和癌旁正常组织之间组蛋白修饰丰度存在明显的差异,且差异性具有一定的规律,特别是涉及转录调控的组蛋白修饰与乳腺癌的预后和治疗靶点具有相关性,进而探讨了乳腺癌中异常HPTMs的生物学意义。该研究对临床石蜡样本中组蛋白修饰的分离分析以及肿瘤表观遗传标志物的检测进行了有益的探索。  相似文献

The main goal of this work was to study the structural transformation of humic acids (HAs) under the influence of selected strains of fungi (Aspergillus niger and Paecilomyces lilacinus) and bacteria (Bacillus sp., Paenibacillus polymyxa and Bacillus amyloliquefaciens) with/without the presence of NPK fertilizers. Two-year experiments were conducted on two different soils and HAs isolated from these soils were examined for structure, humification degree, and quantity using fluorescence and UV-Vis spectroscopy, elemental analysis, and extraction methods. Results showed that the applied additives contributed to the beneficial transformation of HAs, but effects differed for various soils. HAs from silty soil with higher organic carbon content showed simplification of their structure, and decreases in humification, molecular weight, and aromaticity under the influence of fungi and bacteria without NPK, and with NPK alone. With both fungi and NPK, increases in O/H and O/C atomic ratios indicated an increase in the number of O-containing functional groups. HAs from sandy soil did not show as many significant changes as did those from silty soil. Sandy soil exhibited a strong decline in HA content in the second year that was reduced/neutralized by the presence of fungi, bacteria, and NPK. Periodically observed fluorescence at ~300 nm/450 nm reflected formation of low-molecular HAs originating from the activity of microorganisms.  相似文献
