Abstract

Chemically modified electrodes (CMEs) prepared by mixing cobalt phthalocyanine into carbon paste were shown to be able to carry out the electrocatalytic oxidation of nucleic acid species. the oxidation, which occurred at +0.4-0.5 V vs. Ag/AgC1 in 0.15 M NaOH, was observed only for ribose-containing compounds such as the ribonucleosides cytidine, uridine, adenosine, and guanine but not for deoxyribonucleosides such as thymidine or for the purine and pyrimidine bases by themselves. When used for electrochemical detection of the ribonucleosides following liquid chromatography, limits of detection were at the nmol level, with linear response extending at least two orders of magnitude higher. the stability of the CME response was such that 20-30 injections over one hour showed virtually no decrease in peak current and a relative standard deviation of only 2.5%. the detection limits seen with the CMEs were rougnly comparable to those seen for absorption detection at 280 nm, but the selectivity of the CMEs was much narrower in practice.  相似文献

Abstract

Poly (acridine orange) (PAO) film–modified electrode was prepared by the electrooxidation of Acridine orange on a glassy carbon electrode (GCE) for the detection of hydroquinone in the presence of o‐hydroquinone and m‐hydroquinone. The electrochemical behavior of hydroquinone on the modified electrode was investigated with respect to different solution acidity, scan rate, and accumulation time. A pair of sharp and well‐defined peaks was obtained at 0.45 and 0.42 V [vs. a saturated calomel electrode (SCE)] at the PAO film–modified electrode. The potential difference between this pair of cathodic and anodic peaks was decreased to only 30 mV as compared to the 241 mV that was obtained on the bare glassy carbon electrode (GCE). As to o‐hydroquinone and m‐hydroquinone, their corresponding oxidation peaks appeared at 0.55 V and 0.89 V (vs. SCE), respectively. The oxidation potential differences between these three isomers enabled the separate detection of hydroquinone. Under the optimum experimental situation, the oxidation peak current of hydroquinone was proportional to the concentration at the range of 6.8×10?7–9.6×10?5 M. The detection limit was been estimated as 3×10?7 M with 130 s accumulation. This method was applied to the hydroquinone detection in tap water samples.  相似文献

采用表面滴涂结合循环伏安法制备了碳纳米管负载氢氧化镍修饰电极(Ni(OH)2/MWNT/CCE)。研究了该修饰电极对葡萄糖的电催化氧化性能。结果表明,该修饰电极对葡萄糖具有良好的电催化氧化活性。在优化条件下,安培法检测葡萄糖的线性范围为2.0×10-7~5.7×10-4 mol.L-1(r=0.999 9,s=2 786.5μA.(mmol.L-1)-1.cm-2)和5.7×10-4~2.7×10-3 mol.L-1(r=0.999 1,s=2 005.2μA.(mmol.L-1)-1.cm-2),检出限(3sb)为8.0×10-8 mol.L-1。该法已成功用于血清中葡萄糖含量的测定。  相似文献

通过将吡咯单体在低温下与氧化石墨烯进行原位聚合,获得聚吡咯/石墨烯(Ppy/CRGO)复合材料.采用场发射电子显微镜(FESEM)、红外(FT-IR)和热重分析(TGA)对复合物的表面形貌、结构进行表征.FESEM结果表明,通过控制氧化石墨烯(GO)和吡咯单体的质量比例,可以对复合物的层状和厚度进行调控.FT-IR和TGA结果表明,聚吡咯(Ppy)是通过化学键合的方式与氧化石墨烯复合在一起.通过机械冷压法将粉末状Ppy/CRGO复合物压成圆片电极,并探讨了石墨烯和聚吡咯复合比例、反应时间、烘干温度和孔隙率等因素对Ppy/CRGO复合物电极的电学和电化学性能的影响.结果表明,Ppy与CRGO质量比为10∶1所制得的Ppy/CRGO复合物的电容量为421 F·g-1,通过在电极中引入孔隙,电容量能进一步提升为509 F·g-1.  相似文献

Assessment of xenoestrogenic activity in human serum samples requires the removal of endogenous sex hormones to assure that the activity measured originates from xenobiotic compounds only. Serum samples representing high, medium and lower accumulation of persistent organic pollutants (POPs) were extracted using solid-phase extraction (SPE) followed by normal-phase high-performance liquid chromatography (NP-HPLC) for separation of POPs from endogenous hormones. The recovery of polychlorinated biphenyl (PCB) congeners in spiked serum samples was up to 86 %, making the extraction method suitable for the study. MVLN cells, stably transfected with an estrogen receptor (ER) luciferase reporter vector (estrogen response element chemically activated luciferase expression, ERE-CALUX), were exposed to the reconstituted SPE-HPLC extracts for determination of the integrated estrogenic activity. The effects of PCBs were analyzed by direct in vitro exposure of PCBs (nos. 138, 153, 180) and by ex vivo analysis of SPE-HPLC extracts from serum spiked with the PCBs. Similar effects on ER transactivation were observed for the direct in vitro and the ex vivo analysis experiments. The ER transactivation responses determined for actual serum samples were in the linear range of the dose-response curve. 17β-Estradiol titrations showed that the xenoestrogenic effects were mediated via ER. Moreover, our SPE-HPLC-ERE-CALUX assay was demonstrated to elicit high interlaboratory correlation. In the present study the combination of SPE-HPLC purification and the ex vivo estrogenic responses measured by ERE-CALUX was validated and considered to be a valuable tool to assess the combined ER effect of lipophilic serum POPs where additive/synergistic and agonistic/antagonistic effects are integrated giving an overall estimate of exposure and bioactivity.   相似文献

将耐尔兰(Nile Blue, NB)分子修饰到碳纳米管(CNT)表面形成NB-CNT纳米复合体, 谱学结果表明, NB不仅能快速、高效地修饰到CNT表面, 而且还能有效地改善CNT在水溶液中的分散性能. 将NB-CNT修饰到玻碳(GC)电极表面制备了NB-CNT/GC电极, 循环伏安结果显示, 其伏安曲线上不仅表现出一对良好的、几乎对称的NB单体的氧化还原峰, 式量电位E0'几乎不随扫速而变化[其平均值为(-0.422±0.002) V (vs. SCE, 0.1 mol/L PBS, pH 7.0)]; 而且还显示出NB聚合体分子的氧化还原峰, E0'为-0.191 V (100 mV/s时). 进一步的实验结果表明, NB和CNT对NADH(即还原型β-烟酰胺腺嘌呤二核苷酸, 又称还原型辅酶I)的电化学氧化具有协同催化作用, 能使其氧化过电位降低多于560 mV; NB-CNT/GC电极还能较好地响应脱氢酶催化底物氧化过程中体系内NADH浓度的变化. 本文对碳纳米管功能化方法具有简单快速、电极制作容易以及催化效率高等优点, NB-CNT/GC电极有望在制作脱氢酶传感器方面得到应用.  相似文献

制备了多壁碳纳米管修饰碳糊电极并研究了锆-茜素红络合物在该电极上的阳极吸附伏安行为,采用二阶导数线性扫描伏安法进行分析,实验结果表明,在0.128 mol/L氨基乙酸-0.048 mol/L邻苯二甲酸氢钾缓冲液(pH 4.0)中,在200 mV富集60 s,在200~1200 mV范围内以200 mV/s的速率线性扫描,络合物吸附在该修饰电极表面,于832 mV(vs.SCE)处产生一个灵敏的氧化峰,为络合物中配体茜素红的氧化所产生。络合物的峰电流与锆的浓度在1.0×10-11~2.0×10-7mol/L范围内呈良好的线性关系;检出限(S/N=3)为6.0×10-12mol/L(富集时间180 s)。方法用于岩矿样品中痕量锆的测定,结果满意。  相似文献

Recently a novel chronopotentiometric method has been introduced that was found useful for assessing antioxidant activity. In our experiments, the performance of chemically modified working electrodes made of different pencil materials were tested in the novel method. Redox mediator film of Meldola Blue was formed on electrode surfaces. The stability of the film, the number of molecules structuring it has been studied. In antioxidant measurements, controlled potential oxidation of the mediator was made followed by recording the open cell potential in time. The initial slope of this was used as analytical signal. Results proved applicability of the pencil probes.  相似文献
