The protonation dynamics of the DNA base adenine (Ade) and its nucleoside 2'-deoxyadenosine (d-Ade) are investigated by monitoring the deprotonation kinetics of an N-heterocyclic DNA intercalator, acridine (Acr), in the confined environment of sodium dodecyl sulfate (SDS) micelles. Protonation of acridine (AcrH(+)) occurs at the hydrophilic interface and this species remains in dynamic equilibrium with its deprotonated counterpart (Acr) inside the hydrophobic core of SDS micelles. Quenching of the fluorescence of AcrH(+)* at 478 nm is observed after addition of Ade and d-Ade with Stern-Volmer constant (K(SV)) 298 and 75 M(-1), respectively, with a concomitant increment in Acr* at 425 nm. Time-resolved fluorescence studies reveal quenching in the lifetime of AcrH(+)*. The relative amplitude of AcrH(+)* decreases from 0.97 to 0.51 and 0.97 to 0.89 with equimolar addition of Ade and d-Ade, respectively. These observations are explained by excited-state proton transfer (ESPT) from AcrH(+)* to the bases. The reduced K(SV) value and negligible change in the relative amplitudes of AcrH(+)* with d-Ade infer that ESPT is hindered substantially by the presence of a 2'-deoxy sugar unit. Transient time-resolved absorption spectra of Acr reflect that Ade reduces the absorbance of (3)AcrH(+)*; however, d-Ade keeps it unaltered for more than a time delay of 2 μs. The optimized geometries calculated by quantum chemical methods reflect deprotonation of AcrH(+)* with protonation at the N1 position of Ade, while it remains protonated with d-Ade. The hindered ESPT between AcrH(+)* and d-Ade singles out the significance of the 2'-deoxy sugar moiety in controlling the deprotonation kinetics.  相似文献

研究了漆树酶在AOT/正辛烷/水反胶团中邻氨基苯酚的催化氧化性能。发现在40℃、pH=7.6、R=[H2O]/[AOT]≈18的条件下,漆树酶的活力最高,在含水量较低的反胶团中较稳定。当R=6.2,于30℃保存15h活力仍保持82%;"水池"内存在的1.0×10-3mol/L的Zn2+、Fe3+、Mg2+等离子对漆树酶有一定的抑制作用。漆树酶对邻氨基苯酚在反胶团和水溶液介质中,其主要氧化产物均为2-氨基-吩嗪-3-酮,但在反胶团中其反应产率比在水溶液中约高0.5倍。  相似文献

The temperature-dependent luminescence of tris (β-diketone) chelates of europium was used for in situ temperature measurements during cationic photopolymerizations of vinyl ethers. These molecular-level luminescent probes provided a real-time, noninvasive method for monitoring temperature during these high-speed polymerizations. Two specific probes, tris (benzoyl-1, 1, 1-trifluoroacetone) europium and tris (1,1,1,5,5,5,-hexafluoroacetylacetone) europium, met several stringent spectral and performance requirements for application in our system. The luminescence from these probes exhibits a reproducible temperature dependence over a wide temperature range and is not sensitive to changes in viscosity. In situ temperature profiles obtained using this novel technique verified the importance of thermal effects during these highly exothermic photopolymerizations. These studies have demonstrated the utility of the tris(β-diketone) europium chelates for characterizing the temperature during high-speed photopolymerizations that cannot be monitored using conventional techniques. © 1995 John Wiley & Sons, Inc.  相似文献

The rotational mobility of spin probes of different shape and size in low molecular and polymer micelles has been studied. Several probes having nitroxide fragment localized either in the vicinity of micelle interface or in the hydrocarbon core have been used. Upon increasing the number of carbon atoms in hydrocarbon chain of detergent from 7 to 13 (sodium alkyl sulfate micelles) or from 12 to 16 (alkyltrimethylammonium bromide micelles) the rotational mobility of spin probes is decreased by the factor 1.5–2.0. The spin probe rotational mobility in polymer micelles (the complexes of alkyltrimethylammonium bromides and polymethacrylic or polyacrylic acids) is less than mobility in free micelles of the same surfactants. The study of EPR-spectra of spin labeled polymethacrylic acid (PMA) indicated that formation of water soluble complexes of polymer and alkyltrimethylammonium bromides in alkaline solutions (pH 9) does not affect the polymer segmental mobility. On the other hand, the polymer complexes formation in slightly acidic water solution (pH 6) breaks down the compact PMA conformation, thus increasing the polymer segmental mobility. Possible structures of polymer micelles are discussed.  相似文献

在水溶液中制备了甲基环糊精(Me-β-CD)/疏水单体2-苯氧乙基丙烯酸酯(POEA)的包结物(1a)。利用紫外-可见光谱及核磁共振波谱等检测手段研究了Me-β-CD与POEA的包合作用,并对包合物的结构、性能进行了表征和分析。同时在水溶液中引发丙烯酰胺(AM)、阳离子单体丙烯酰氧乙基三甲基氯化铵(DAC)以及1a的均相共聚合得到疏水修饰的阳离子聚丙烯酰胺P(AM-DAC-POEA),实现了用绿色方法合成高效高分子絮凝剂的目的,并对该高分子对高岭土模拟污水的絮凝效果进行了考察。  相似文献

Whenp-toluidine is added to an aqueous solution of CTAB, a remarkable increase of viscosity is accompanied by a spectacular elasticity. We detected the existence of extremely elongated rod-like micelles in electron micrographs. SAXS measurements indicate a closely packed array of cylindrical rod-like micelles, brought about when solutions flow through a thin capillary. A scattering maximum ofd=160 Å almost corresponds to the distance between the nearest neighbours of the cylindrical rod-like micelles. This value agrees with the diameter measured on electron micrographs. The second broad peak (d=75 Å) is assigned to a subsidiary maximum of the shape function of the cylinder with infinite length.  相似文献

The influence of isopropyl alcohol (IPA) on the size and composition of the mixed micelles in mixtures of tetradecyltrimethylammonium bromide (TTAB) and chlorhexidine digluconate (CG) has been determined as a function of the composition of the systems. The addition of 0.5 M and 1.0 M IPA had little significant effect on the composition of the mixed micelles as determined both by analysis of critical micelle concentration (CMC) data using a theoretical treatment based on excess thermodynamic quantities and by an empirical treatment of conductivity data. Static and quasielastic light scattering measurements showed a progressive decrease of the aggregation number and hydrodynamic radius of TTAB micelles on addition of IPA, but minimal changes in the properties of the small CG aggregates. The results show that the micellar weight in the TTAB/CG/IPA solutions is determined by the ratio of the surfactants in the system and for each TTAB/CG ratio decreases on addition of IPA.  相似文献

The purpose of this study was to develop mixed polymeric micelles with high drug loading capacity to improve the oral bioavailability of icaritin with Soluplus® and Poloxamer 407 using a creative acid-base shift (ABS) method, which exhibits the advantages of exclusion of organic solvents, high drug loading and ease of scaling-up. The feasibility of the ABS method was successfully demonstrated by studies of icaritin-loaded polymeric micelles (IPMs). The prepared IPMs were characterized to have a spherical shape with a size of 72.74 ± 0.51 nm, and 13.18% drug loading content. In vitro release tests confirmed the faster release of icaritin from IPMs compared to an oil suspension. Furthermore, bioavailability of icaritin in IPMs in beagle dogs displayed a 14.9-fold increase when compared with the oil suspension. Transcellular transport studies of IPMs across Caco-2 cell monolayers confirmed that the IPMs were endocytosed in their intact forms through macropinocytosis, clathrin-, and caveolae-mediated pathways. In conclusion, the results suggested that the mixed micelles of Soluplus® and Poloxamer 407 could be a feasible drug delivery system to enhance oral bioavailability of icaritin, and the ABS method might be a promising technology for the preparation of polymeric micelles to encapsulate poorly water-soluble weakly acidic and alkaline drugs.  相似文献

In this preliminary report we present the mesomorphic properties and electro-optic investigations of three new 'banana-shaped' mesogens. The materials are structurally similar to Niori's the original bent-core materials but possess alkylthio substituents in place of alkoxy substituents. Microscopic investigations revealed an unusual 'spiral-domain texture', similar to that observed in the 'B7' phase of related materials. Further investigations support our earlier observations that the materials display antiferroelectric switching in this phase, in two of the new materials studied. The nature of this phase, in comparison with the 'switchable' (B2) and 'chiral-domain' (B4) phases of the original materials, is discussed.  相似文献
