本文通过酶活性测定,荧光光谱和紫外光谱研究了外加Cu2+与烟草多酚氧化酶(简称PPO)的相互作用。结果表明,微量铜的加入能增加酶的活性,[Cu2+]/[PPO]为0.20左右时酶活性最大,[Cu2+]/[PPO]为0.91时,Cu2+开始表现出对PPO活性的抑制;Cu2+对PPO内源荧光的猝灭机制属于形成络合物所引起的静态猝灭,猝灭常数Ksv为8.0375×103L·mol-1;Cu2+的加入使PPO蛋白质分子构象发生变化,α-螺旋含量增加,多肽链及Trp和Tyr残基的芳杂环进一步向分子内收缩,疏水基团之间的疏水作用增强。  相似文献

The present short review deals with electroanalytical aspects of electrochemical response of ascorbic acid (Vitamin C) at conducting and electrogenerated polymer modified electrodes. Two main topics are considered: (i) electrocatalytic oxidation of ascorbate at conducting polymer modified electrodes, leading to electroanalytical techniques for ascorbate assay, and (ii) retardation of ascorbate penetration through a layer of electrogenerated polymers, leading to permselective coatings and their diverse uses, especially for biosensing devices.  相似文献

 利用交替三线性分解算法与反相高效液相 二极管阵列检测 (RP HPLC DAD)相结合 ,在洗脱时间为1 0 86min~ 1 399min(间隔 1 / 1 50min)、紫外吸收波长为 2 68nm~ 2 98nm(间隔 1nm)时对苯二酚位置异构体的重叠及光谱体系进行了分辨研究。分辨结果与实际结果一致。同时测定了水溶液中共存的邻苯二酚、间苯二酚和对苯二酚的含量 ,回收率分别为 (1 0 0 1± 1 0 ) % ,(99 4± 1 4) % ,(1 0 0 5± 1 7) %。研究结果表明 :该方法定量快速准确 ,实验操作步骤简单 ,解决了在干扰物存在条件下三者很难同时分辨的问题。  相似文献

报导了用乙基纤维素和乙炔黑获得的导电复合材料构成的葡萄糖氧化酶生物传感器的制备方法.讨论了多种因素对该生物传感器响应电流的影响.测得此电极酶催化反应的活化能为40.3 kJ•mol-1. AFM实验表明,用环己烷洗去石蜡的导电复合材料 葡萄糖氧化酶生物传感器具有粒状结构,这有利于酶催化反应的进行.  相似文献

A surface-renewable glucose biosensor is reported. The glucose biosensor is developed using glucose oxidase (GOD) encapsulated in organically modified solgel glass (ORMOSIL) network in the composite matetial. The organic group in the ORMOSIL network controls the hydrophobicity of the electrode surface and thus limits the wettability of the electrode surface. The graphite powder provides the conductivity for the electrode.Ferrocenecarboxylic acid in phosphate buffer solution (pH 7.0) transfers electron between enzyme and electrode. Cyclic volammetry and amperometric measurements have been used to exmine the electrochemical behavior of glucose biosensor as shown in Fig. 1 and Fig.2. The electrode gives a linear response range of 1 -20mM glucose with a sensitivity of 3.26 μA· mM-1. The electrode can be renewed easily in reproducible manner by a simple polishing step.  相似文献

Immobilization of invertase and glucose oxidase in conducting polypyrrole and copolymers of poly 2-methylbutyl-2-(3-thienyl) acetate with pyrrole were achieved via electrochemical method. Sodium dodecyl sulphate was found to be the most suitable supporting electrolyte. Maximum reaction rate, Michaelis-Menten constant and optimum temperatures were determined for native and immobilized enzymes. Storage and operational stabilities of enzyme electrodes were also investigated.  相似文献

Carbon nanotubes used for constructing biosensor was described for the first time. Single-wall carbon nanotubes (SWNTs) functionalized with carboxylic acid groups were used to immobilize glucose oxidase forming a glucose biosensor. A detection scheme for the simultaneous evaluation of two bioassays based on fluorescence spectroscopy is presented. For the determination of hydrogen peroxide-generating enzymes or peroxidases, the non-fluorescent 4-(N-methylhydrazino)-7-nitro-2,1,3-benzooxadiazole (MNBDH) is converted to the strongly fluorescent 4-(N-methylamino)-7-nitro-2,1,3-benzooxadiazole (MNBDA). Phosphatases are detected based on the cleavage of the non-fluorescent 5-fluorosalicyl phosphate (5-FSAP) under formation of the fluorescent 5-fluorosalicylic acid (5-FSA). While excitation of the fluorophores may be carried out at the same wavelength, their emission spectra differ significantly. This allows the read-out of both assays using commercially available microplate readers without additional chemometric tools. Compared with individual assays, limits of detection are similar, and linearity of the calibration functions for both enzymes is observed over 2-3 concentration decades starting at the limit of quantification. The simultaneous determination of glucose oxidase and acid phosphatase in honey is presented as example for the application of the detection scheme.  相似文献

Traditional Chinese medicine (TCM) is the key to unlock treasures of Chinese civilization. TCM and its compound play a beneficial role in medical activities to cure diseases, especially in major public health events such as novel coronavirus epidemics across the globe. The chemical composition in Chinese medicine formula is complex and diverse, but their effective substances resemble “mystery boxes”. Revealing their active ingredients and their mechanisms of action has become focal point and difficulty of research for herbalists. Although the existing research methods are numerous and constantly updated iteratively, there is remain a lack of prospective reviews. Hence, this paper provides a comprehensive account of existing new approaches and technologies based on previous studies with an in vitro to in vivo perspective. In addition, the bottlenecks of studies on Chinese medicine formula effective substances are also revealed. Especially, we look ahead to new perspectives, technologies and applications for its future development. This work reviews based on new perspectives to open horizons for the future research. Consequently, herbal compounding pharmaceutical substances study should carry on the essence of TCM while pursuing innovations in the field.  相似文献
