Stereocontrolled syntheses of biotin-labeled oligosaccharide portions containing the carbohydrate moiety of glycoprotein from Echinococcus granulosus have been accomplished. Trisaccharide Galβ1-3Galβ1-3GalNAcα1-R (A), tetrasaccharide Galα1-4Galβ1-3Galβ1-3GalNAcα1-R (B), and pentasaccharide Galα1-4Galβ1-3Galβ1-3Galβ1-3GalNAcα1-R (C), (R = biotinylated probe) were synthesized by stepwise condensation and/or block synthesis by the use of 5-(methoxycarbonyl)pentyl 2-azido-4,6-O-benzylidene-2-deoxy-α-d-galactopyranoside as a common glycosyl acceptor. The synthesis of the tetrasaccharide and the pentasaccharide was improved from the viewpoint of reducing the number of synthetic steps and increasing the total yield by changing from stepwise condensation to block synthesis. Moreover, hexasaccharide E, which contains the oligosaccharide sequence which occurs in E. granulosus, was synthesized from trisaccharide D. We examined the antigenicity of these five oligosaccharides by an enzyme-linked immunosorbent assay (ELISA). Although compounds of C–E did not exhibit antigenicity against cystic echinococcosis (CE) patient sera, compounds B, D, and E showed good serodiagnostic potential for alveolar echinococcosis (AE). 相似文献

We describe herein, the creation of an organic phase enzyme electrode (OPEE) via avidin–biotin interactions built over an electrogenerated polymer. Multilayered polyphenol oxidase (PPO) assemblies were transferred into an organic solvent (chloroform) for the catechol detection at −0.2 V. In conjunction with an alginate gel, as a hydrophilic additive, the biosensor performance was widely enhanced. The effects of biotinylated polypyrrole film and alginate gel on the diffusion process through the biosensor coating are studied by rotating disk electrode experiments carried out in chloroform with hydroquinone as electroactive permeant. 相似文献

Thioi-terminated oligonucleotide was immobilized to gold sur-face by self-assembly method. A novel amplification strategy was introduced for improving the sensitivity of DNA hybridiza-tion using biotin labeled protein-streptavidin network complex. This complex can be formed in a cross-linking network of molecules so that the amplification of the response signal will be realized due to the big molecular size of the complex. It could be proved from the impedance technique that this amplification strategy caused dramatic improvement of the detection sensitivi-ty. A simple and sensitive ligand affinity capture method (LAC) was developed to detect biotinylated biomolecules bound to a biotin–avidin base by matrix‐assisted laser desorption ionization time‐of‐flight mass spectrometry (MALDI ToF MS). Glass slides covered with a metal film for MALDI MS applications were treated with amino‐silane and derivatized with biotin followed by binding of avidin. Washing buffers with high ionic strength increased the specificity of the subsequent binding of biotinylated biomolecules to the avidin layer. A combined thin layer‐dried droplet method using α‐cyano‐4‐hydroxycinnamic acid (CHCA) in acetone or ethyl acetate resulted in the most intense ions of biotinylated polymyxin B, whereas the matrix conditions did not influence the detection of angiotensin II. Addition of biotinylated biomolecules in the low femtomole to low picomole range resulted in sufficient ion intensity for detection by the LAC method. The LAC concept was extended by binding of biotinylated lipopolysaccharide to the biotin–avidin base followed by preferential capture and specific detection of the binding antagonist polymyxin B. Films of electrogenerated polypyrrole and hydrophilic alginate, both functionalized with biotin moieties, were used to allow for the transfer of polyphenol oxidase activity in organic media. Enzyme electrodes, based on multilayered structures, were protected at the molecular level by the affinity binding of alginate as a hydrophilic additive, and were then transferred into chlorobenzene, dichloromethane, chloroform, ethyl acetate or acetonitrile. The biosensor performance for the detection of catechol at ?0.2 V was investigated, highlighting the main influence of the hydrophobicity of the solvent and, to a lesser extent, the dielectric constant. The effect of the substrate hydrophobicity on the biosensor response was examined in chlorobenzene. 相似文献

The kinetics of free glucose-6-phosphate dehydrogenase (G-6-PDH), biotinylated G-6-PDH, and biotinylated G-6-PDH complexed
with avidin were investigated. The kinetics of the free enzyme were consistent with a sequential rather than a ping-pong mechanism.
The kinetics of the biotinylated enzyme were similar to that of the free enzyme, but the kinetic constants were different;

m value for NADP was halved, whereas theK
m for G-6-P decreased only slightly. In the presence of avidin, theK
m of biotinylated G-6-PDH for G-6-P nearly doubled whereas theK
m for NADP did not change significantly. Avidin complexed with biotinylated G-6-PDH inhibited the enzyme from acting. Based
upon these reactions, it was possible to devise assays for either free biotin or free avidin using biotinylated G-6-PDH as
the indicator enzyme. Concentrations of biotin between 40 and 60 mg/mL, or of 25–95 Μg/mL of avidin could be measured within

Cell adhesion to a scaffold is a prerequisite for tissue engineering. Many studies have been focused on enhancing cell adhesion to synthetic materials that are used for scaffold fabrication. In this study, we applied an avidin-biotin binding system to enhance chondrocyte adhesion to biodegradable polymers. Biotin molecules were conjugated to the cell membrane of chondrocytes, and mediated cell adhesion to avidin-coated surfaces. We demonstrated that immobilization of biotin molecules to chondrocyte surfaces enhanced cell adhesion to avidin-coated biodegradable polymers such as poly(L-lactic acid), poly(D,L-lactic acid), and polycaprolactone, compared to the adhesion of normal chondrocytes to the same type of biodegradable polymer. The biotinylated chondrocytes still maintained their proliferation ability. This study showed the promise of applying the avidin-biotin system in cartilage tissue engineering. [diagram in text]. 相似文献
