Synthesis of thieno[3.2-<Emphasis Type="Italic">b</Emphasis>]pyrrolenine derivatives under the Fischer reaction conditions

The reaction of alkyl 4-(2-acetylhydrazino)-2-methyl-3-thiophenecarboxylates with 3-methylbutan-2-one was carried out under the Fischer reaction conditions. The influence of Lewis acids and solvents on the process was studied. A convenient method for the synthesis of thieno [3.2-b]pyrrolenine derivatives was proposed.__________Published in Russian in Izvestiya Akademii Nauk. From D-glucose to biologically potent L-hexose derivatives: synthesis of alpha-L-iduronidase fluorogenic detector and the disaccharide moieties of bleomycin A2 and heparan sulfate

A novel and convenient route for the synthesis of biologically potent and rare L-hexose derivatives from D-glucose is described. Conversion of diacetone-alpha-D-glucose (14) into 1,2:3,5-di-O-isopropylidene-beta-L-idofuranose (19) was efficiently carried out in two steps. Orthogonal isopropylidene rearrangement of compound 19 led to 1,2:5,6-di-O-isopropylidene-beta-L-idofuranose (27), which underwent regioselective epimerization at the C3 position to give the L-talo- and 3-functionalized L-idofuranosyl derivatives. Hydrolysis of compound 19 under acidic conditions furnished 1,6-anhydro-beta-L-idopyranose (35) in excellent yield, which was successfully transformed into the corresponding L-allo, L-altro, L-gulo, and L-ido derivatives via regioselective benzylation, benzoylation, triflation and nucleophilic substitution as the key steps. Applications of these 1,6-anhydro-beta-L-hexopyranoses as valuable building blocks to the syntheses of 4-methylcoumarin-7-yl-alpha-L-iduronic acid and the disaccharide moieties of bleomycin A(2) as well as heparan sulfate are highlighted.     

1H and 13C assignments of two new triterpenes from Cladocolea grahami

The structure elucidation and 1H and 13C assignments of the new triterpenes 3beta-palmitoxy-7beta-hydroxyolean-12-ene (1) and its hydrolysis product 3beta,7beta-dihydroxyolean-12-ene (2) and 3beta-sn-glyceroyl-(1'-palmitoxy)urs-12-ene (3), isolated from the aerial parts of Cladocolea grahami (Loranthaceae), are reported.     

Study on oxidation of polymers treated by high LET radiation

The oxidation of medical silicone rubber and segmented polyether urethane which were bombarded with 11.2 MeV Si⁺ or F⁺ at the dose of 5 × 10¹⁴ ions/cm² was studied. In XPS measurements, it was found that the oxygen concentration on the surface of the implanted polymeric membranes had been increasing in three months at least. ESR measurement showed that some free radicals existed in these membranes and their quantities decreased with time. Chemical analysis proved that peroxides were generated in the γ irradiated membranes. The reason for the increasing of oxygen concentration was that free radicals combined with oxygen in the air and produced peroxides on the membrane surface. Moreover, in order to demonstrate the radiation mechanism, low LET (γ rays) radiation of such polymeric membranes was carried out for comparison     

Evaluation of the immobilized artificial membrane phosphatidylcholine: Drug discovery column for high-performance liquid chromatographic screening of drug–membrane interactions

Chromatographic retention factors (k′) of a series of eight β-adrenoceptor antagonist compounds (β-adrenolytic drugs) were determined employing an immobilized artificial membrane column (IAM.PC.DD). The influence of mobile phase pH, ionic strength, and organic modifier composition was studied in order to examine column performance. After the IAM.PC.DD columns were exposed to approximately 7000 column volumes of a 0.01 M PBS mobile phase, five out of six columns tested showed significant peak broadening and decreased k′ values indicative of premature column failure. The data suggested that the immobilized phospholipids stationary phase was removed by the 0.01 M PBS mobile phase. The β-adrenolytic drug's log k′_IAM values obtained with an IAM.PC.DD column were compared to an ^esterIAM.PC.MG column for predicting drug membrane interactions. For the linear regression analysis between log k′_IAM and the logarithm of the n-octanol–water partition coefficients (r_IAM.PC.DD=0.8710 vs. r_IAM.PC.MG=0.9538), the C₁₈ HPLC retention factors (r_IAM.PC.DD=0.8408 vs. r_IAM.PC.MG=0.9380), the liposome partition coefficients (r_IAM.PC.DD=0.8887 vs. r_IAM.PC.MG=0.9187), and various pharmacokinetic parameters, significantly better correlations were obtained with the ^esterIAM.PC.MG column than the IAM.PC.DD column.     

Carboxylic Acids from <Emphasis Type="Italic">Phyllanthus urinaria</Emphasis>

Five compounds, terephthalic acid mono-[2-(4-carboxy-phenoxycarbonyl)-vinyl] ester (1), (E)-3-(5′-hydroperoxy-2,2′-dihydroxy[1,1′-biphenyl]-4-yl)-2-propenoic acid (2), 3,4,5-trihydroxybenzoic acid (3), succinic acid (or butanedioic acid) (4), and 2,3,4,5,6-pentahydroxybenzoic acid (5), were isolated from Phyllanthus urinaria. The structures of these compounds were elucidated by means of spectral techniques including IR, MS, and 1D/2D NMR. 1 and 2 are new compounds.__________Published in Khimiya Prirodnykh Soedinenii, No. 1, pp. 14–17, January–February, 2005.     

Efficient synthesis of p-bis-(chlorodifluoromethyl)benzene

Selective, high yield partial fluorination of p-bis-(trichloromethyl)benzene to p-bis-(chlorodifluoromethyl)benzene has been accomplished by warming a slurry of the p-bis-(trichloromethyl)benzene in anhydrous HF which also contains a small quantity of inert solvent, such as 1,2-dichloroethane.     

Synthesis and polymerization of N-[N′-(α-methylbenzyl)aminocarbonyl-n-alkyl]maleimide

Two types of optically active N-[N′-(α-methylbenzyl)amino/carbonyl-n-alkyl]maleimides (MBAC) were synthesized from maleic anhydride, 6-amino-n-caproic acid (or 12-amino-n-dodecanoic acid), and (R)-(+)-α-methylbenzylamine. Radical homopolymerizations of MBAC were performed in several solvents at 60 and 110°C for 24 h to give optically active polymers. Radical copolymerizations of MBAC were performed with styrene (ST) and methyl methacrylate (MMA) in dioxane at 60°C. The monomer reactivity ratios and the Alfrey-Price Q-e values were determined. Chiroptical properties of the polymers and copolymers were investigated. © 1995 John Wiley & Sons, Inc.     

Cross-metathesis of vinyl aromatic heterocycles with 1-octene in the presence of a Schrock catalyst

Metathesis of 2-vinyl aromatic heterocycles such as furan, thiophene, pyrrole and pyridine in the presence of a molybdenum-based Schrock catalyst has been investigated from a synthetic point of view. The self-metathesis of 2-vinyl aromatic heterocycles was not successful. However, in cross-metathesis of thiophene, furan and styrene with 1-octene, the cross-metathesis product, heterodimer, was readily obtained selectively, together with only small amounts of the two corresponding self-metathesis products. The origin of the surprisingly high selectivity of heterodimer formation was elucidated through metallacyclobutane intermediate mechanism, observations of carbenes by in situ ¹H NMR, and reaction products.     

Preparation of Y2Ba4Cu7O15-δ Superconductor without High Oxygen Pressure

The synthesis of bulk Y₂Ba₄Cu₇O_15-δ superconductor at atmospheric oxygen pressure via solid state sintering is reported. Temperature ranging from 860 to 890 °C as well as time interval over 2 to 15 days were used to investigate the formation of the Y₂Ba₄Cu₇O_15-δ phase. A time-temperature profile characterizing the conditions for the preparation of Y₂Ba₄Cu₇O_15-δ phase suggests the optimal condition to be sintering at 890 °C for over 10 days. Detailed results of X-ray diffraction, electrical resistivity, iodometric titration and magnetization measurements are described.
