Catalytic Asymmetric Synthesis of Allylic Alcohols and Derivatives and their Applications in Organic Synthesis

Allylic alcohols represent an important and highly versatile class of chiral building blocks for organic synthesis. This Review summarizes the plethora of methods developed for the catalytic asymmetric synthesis of enantioenriched allylic alcohols. These include: dynamic kinetic resolution (DKR/DKAT), nucleophilic 1,2‐addition to carbonyl groups, allylic substitution, oxidation of C? H bonds, the addition of O nucleophiles to π systems, reduction of unsaturated carbonyl compounds, and an alternative route from enantioenriched propargylic alcohols. Furthermore, these catalytic asymmetric processes are exemplified by their applications in the syntheses of complex molecules such as natural products and potential therapeutic agents.     

The Psymberin Story—Biological Properties and Approaches towards Total and Analogue Syntheses

Psymberin is a marine natural product which has attracted a great deal of interest since its isolation: While the highly cytotoxic compound was detected early on as an ingredient in a marine sponge, it took over a decade and 600 additional samples for the structure to eventually be assigned. In the last eight years fascinating synthetic and biosynthetic investigations have led to a more detailed understanding as well as a new starting point for structure–activity studies towards new antitumor compounds. The Review gives an in‐depth insight into the progress in the field of the marine polyketide psymberin and demonstrates how organic synthesis is influencing neighboring scientific subjects.     

Simultaneous determination of seven prohibited substances in cosmetic products by liquid chromatography-tandem mass spectrometry

In this paper,we developed and validated a simple,sensitive,and selective high-performance liquid chromatography-tandem mass spectrometry(HPLC-MS/MS) method to identify and measure the following prohibited substances that may be found in cosmetic products:minoxidil,hydrocortisone, spironolactone,estrone,canrenone,triamcinolone acetonide and progesterone.Chromatographic separation was performed on a Waters Symmetry C18(100 mm×2.1 mm,3.5μm particle size) with a gradient elution system composed of 0.2%(v/v) formic acid aqueous solution and methanol containing 0.2%(v/v) formic acid at a flow rate of 0.3 mL/min.The substances were detected using a triple quadrupole mass spectrometer in the multiple reaction monitoring mode with an electrospray ionization source.All of the calibration curves showed good linearity(r > 0.999) within the tested concentration ranges.The limit of detection was <25 pg.The relative standard deviations for intraday precision for each of the prohibited substances were <3.5%at two concentration levels(2μg/g,10μg/g). The relative recovery rate for each of the prohibited substances ranged from 91.8%to 111%at three concentration levels(0.1μg/g,2μg/g,10μg/g),including the limit of quantification.In conclusion,we have developed and validated a method that can identify seven prohibited substances in cosmetic products.     

离子色谱法测定面制品和面包改良剂中的溴酸盐

用离子色谱法测定面制品和面粉改良剂中的溴酸盐。用30 mL水超声提取样品15 min(脂肪含量高的样品先用正己烷除脂),过HLB柱和银氢柱净化,以AS9–HC色谱柱分离,NaHCO3作为淋洗液,电导检测器检测,实现了复杂基质面制品中溴酸盐的检测。方法线性相关系数大于0.999,最低检测浓度为0.5 mg/kg,加标回收率为78.4%~106.3%。利用建立的方法对面制品和面包改良剂中的溴酸盐进行检测,共检测各类面制品144份,总体合格率为97.2%;检测面包改良剂类产品10份,合格率为40.0%。     

LC–MS/MS法测定动物性食品中万古霉素残留

建立液相色谱–串联质谱(LC–MS/MS)法检测动物性食品中万古霉素药物残留的方法。用磷酸盐缓冲溶液对动物性食品中的万古霉素进行提取,经HLB固相萃取柱净化,采用电喷雾离子源,以正离子检测方式进行质谱分析。实验结果表明,万古霉素质量浓度在1~500μg/L范围内与色谱峰面积呈良好的线性,相关系数r=0.998。低、中、高3个质量浓度添加水平的回收率为84.8%~118.2%,相对标准偏差为2.2%~7.2%(n=5),检出限为2μg/kg。     

Practical Synthesis of (20S)‐10‐(3‐Aminopropyloxy)‐7‐ethylcamptothecin,a Water‐Soluble Analogue of Camptothecin

A robust, practical synthesis of (20S)‐10‐(3‐aminopropyloxy)‐7‐ethylcamptothecin (T‐2513, 5 ), which is a water‐soluble analogue of camptothecin, has been developed. The key step in this synthesis is a highly diastereoselective ethylation at the C20 position by using N‐arylsulfonyl‐(R)‐1,2,3,4‐tetrahydroisoquinoline‐3‐carboxylic acid ester as a chiral auxiliary, which affords the key intermediate ethyl‐(S)‐2‐acyloxy‐2‐(6‐cyano‐5‐oxo‐1,2,3,5‐tetrahydroindolizin‐7‐yl)butanoate ( 8 k ) in 93 % yield and 87 % de. Optically pure compound 8 k was obtained by a single recrystallization from acetone and its further elaboration through Friedlander condensation afforded compound 5 . This synthesis does not require any chromatographic purification steps and can provide compound 5 on a multi‐gram scale in 6.3 % overall yield (16 steps).
