In this work, an electrochemical sensor coupled with an effective flow-injection amperometry (FIA) system is developed, targeting the determination of gallic acid (GA) in a mild neutral condition, in contrast to the existing electrochemical methods. The sensor is based on a thin electroactive poly(melamine) film immobilized on a pre-anodized screen-printed carbon electrode (SPCE*/PME). The characteristics of the sensing surface are well-characterized by field emission scanning electron microscopy (FE-SEM), X-ray photoelectron spectroscopy (XPS) and surface water contact angle experiments. The proposed assay exhibits a wide linear response to GA in both pH 3 and pH 7.0 phosphate buffer solutions (PBS) under the optimized flow-injection amperometry. The detection limit (S/N = 3) is 0.076 μM and 0.21 μM in the pH 3 and pH 7 solutions, respectively. A relative standard deviation (RSD) of 3.9% is obtained for 57 successive measurements of 50 μM GA in pH 7 solutions. Interference studies indicate that some inorganic salts, catechol, caffeine and ascorbic acid do not interfere with the GA assay. The interference effects from some orthodiphenolic compounds are also investigated. The proposed method and a conventional Folin–Ciocalteu method are applied to detect GA in green tea samples using the standard addition method, and satisfactory spiked recoveries are obtained.  相似文献

Plastic disposable choline biosensors based on ruthenized-carbon screen-printed electrodes were prepared and their use for monitoring organophosphorus pesticides and carbamates is described. The presence of 0.5% ruthenium on activated carbon mixed to form a simple graphite-based ink for the working electrode surface increased the sensitivity towards hydrogen peroxide. The choline biosensor is based on such an electrode coupled with choline oxidase immobilized by adsorption and was used to detect the inhibition effect of carbamates and organophosphorus pesticides on acetylcholinesterase. With the optimized procedure described (pH, buffer composition, incubation time, substrate concentration), concentrations of pesticides (Carbofuran) as low as 1 nM could be detected.  相似文献

Summary A highly sensitive on-line amperometric detection of platinum compounds in HPLC effluents was possible with the use of a polarographic detector. For TNO-1 (=cis-1,1-di(aminomethyl)-cyclohexane Pt(II)chloride) a linear dynamic range of at least three decades could be obtained (0.034–108g TNO-1/ml) with a detection limit of three times the noise signal at 0.7 ng TNO-1 (20ng Pt/ml). The sensitivity was 0.1 nA/ng TNO-1 (0.2nA/ng Pt). The within-day presicion was 1.1% at a concentration of 10.8g TNO-1/ml (n=10). With this system the compatibility was measured of TNO-6 with infusion fluids. In 0.15 M NaCl, TNO-6 (=cis-1,1-di(aminomethyl)-cyclohexanePt(II)sulphate) was converted into TNO-1 within 40 minutes. Molecular changes of TNO-6 were also observed in a 5% aqueous glucose solution.  相似文献

Voltametric studies on glassy-carbon and carbon-fiber electrodes result in the development and testing of a carbon-fiber microelectrode capable of operating in model buffer and biological solutions when monitoring nitric oxide in vivo. The Nafion-modified electrode confidently determines the NO presence in solutions containing anions NO 2 ? , which interfere with the assay. Once properly calibrated, it permits quantitative determination of the NO concentration in a buffer solution.  相似文献

A promising electrochemical sensor based nickel‐carbon nanotube (Ni‐CNT) modified on glassy carbon (GC) electrode had been developed and the properties of the modified electrode were characterized by multispectroscopic analysis. The fabricated sensor (GC/Ni‐CNT) electrode was utilized to determine the catecholamines such as epinephrine and dopamine simultaneously. Differential pulse voltammetry and amperometry were used to verify the electrochemical behavior of the studied compounds. The GC/Ni‐CNT based amperometric sensor showed a wide linear range and low detection limit with high analytical sensitivity of 8.31 and 6.61 μA μM?1 for EP and DA, respectively which demonstrates better characteristics compared to other electrodes reported in the literature. Further, no significant change in amperometric current response was observed in presence of biological interference species such as glucose, cysteine, citric acid, uric acid and ascorbic acid in the detection of EP and DA. The utility of this GC/Ni‐CNT electrode was well established for the determination of EP and DA in human urine samples.  相似文献

We describe a microfluidic device that can be used to detect interactions between red blood cells (RBCs) and endothelial cells using a gold pillar array (created by electrodeposition) and an integrated detection electrode. Endothelial cells can release nitric oxide (NO) via stimulation by RBC‐derived ATP. These studies incorporate on‐chip endothelial cell immobilization, direct RBC contact, and detection of NO in a single microfluidic device. In order to study the RBC‐EC interactions, this work used a microfluidic device made of a PDMS chip with two adjacent channels and a polystyrene base with embedded electrodes for creating a membrane (via gold pillars) and detecting NO (at a glassy carbon electrode coated with platinum‐black and Nafion). RBCs were pharmacologically treated with treprostinil in the absence and presence of glybenclamide, and ATP release was determined as was the resultant NO release from endothelial cells. Treprostinil treatment of RBCs resulted in ATP release that stimulated endothelial cells to release on average 1.8±0.2 nM NO per endothelial cell (average±SEM, n=8). Pretreatment of RBCs with glybenclamide inhibited treprostinil‐induced ATP release and, therefore, less NO was produced by the endothelial cells (0.92±0.1 nM NO per endothelial cell, n=7). In the future, this device can be used to study interactions between many other cell types (both adherent and non‐adherent cell lines) and incorporate other detection schemes.  相似文献

A micromolar concentration of zinc has been shown to significantly change the dynamics of exocytosis as well as the vesicle contents in a model cell line, providing direct evidence that zinc regulates neurotransmitter release. To provide insight into how zinc modulates these exocytotic processes, neurotransmitter release and vesicle content were compared with single cell amperometry and intracellular impact vesicle cytometry with a range of zinc concentrations. Additionally, time-of-flight secondary ion mass spectrometry (ToF-SIMS) images of lipid distributions in the cell membrane after zinc treatment correlate to changes in exocytosis. By combining electrochemical techniques and mass spectrometry imaging, we proposed a mechanism by which zinc changes the fusion pore and the rate of neurotransmitter release by changing lipid distributions and results in the modulation of synaptic strength and plasticity.  相似文献

In this work, a screen‐printed carbon electrode (SPCE) was modified with a cobalt/porous silicon (Co@PSi) nanocomposite powder to develop a nonenzymatic sensor for the detection of hydrogen peroxide. The Co@PSi nanocomposite was synthesized through the chemical reaction between silicon powder in a HF/HNO3 solution and cobalt cations. In this process, cobalt nanoparticles were anchored on the porous silicon. The structure and morphology of the synthesized nanocomposite were investigated by X‐ray diffraction, Fourier transform infrared spectroscopy, X‐ray photoemission spectroscopy, energy dispersive X‐ray spectroscopy, and field‐emission scanning electron microscopy. The constructed nonenzymatic, screen‐printed sensors based on the Co@PSi nanocomposite showed perfect electrocatalytic oxidation response to hydrogen peroxide over the range 1–170 and 170–3,770 μmol/L with the limit of detection of 0.8 μmol/L. In addition, the Co@PSi‐SPCE sensor exhibited good selectivity for the determination of H2O2 in the presence of common interfering species including glucose, ascorbic acid, uric acid, dopamine, nitrate, and nitrite ions. The constructed electrochemical sensor was successfully used for the determination of H2O2 in real samples.  相似文献

A portable electroanalytical system applied for rapid and simultaneous determination of uric acid (UA) and nitrite (NIT) in human biological fluids (urine, saliva and blood) is reported. The system is based on batch‐injection analysis with multiple‐pulse amperometric (BIA‐MPA) detection using screen‐printed electrodes (SPEs) modified with multi‐walled carbon nanotubes. Sample dilution in optimized electrolyte (0.1 mol L−1 Britton‐Robinson buffer pH 2) followed by injection of 100 μL on the electrode surface using an electronic micropipette is performed. UA is detected at +0.45 V and both UA+NIT at +0.70 V. Linear calibration plots for UA and NIT were obtained over the range of 1–500 μmol L−1 with detection limits of 0.05 and 0.06 μmol L−1, respectively. For comparison, a differential‐pulse voltammetric (DPV) method was optimized, and linear calibration plots for UA and NIT were obtained over range of 1–30 μmol L−1 and 1–40 μmol L−1 with detection limits of 0.1 and 0.3 μmol L−1, respectively. BIA‐MPA is highly precise (RSD<1.3 %), fast (160 h−1) and free from sample‐matrix interferences as recovery values ranged from 77 to 121 % for spiked samples (short contact time of sample aliquot with SPE). Contrarily, recovery tests conducted using DPV did not provide adequate recovery values (>150 %), probably due to the longer contact time of the SPE with the biological samples during analysis leading to a severe interference of sample matrices.  相似文献

One-dimensional Ni/Au/PPy-COOH nanowires with multiple segments were synthesized in this study. Smooth surfaces and magnetic properties of nanowires were investigated by scanning transmission electron microscopy (SEM), Energy-dispersive X-ray spectroscopy (EDX), and Electron Spin Resonance (ESR) techniques. The nanowires were used to modify the screen-printed electrode surface and as a micro-environment for Trametes versicolor laccase. The ability of this enzyme biosensor to detect dopamine change in human biological samples was demonstrated by a wide linear range (0.01–50 μM) and a low LOD (2.265 nM). In addition, the biosensor exhibited excellent selectivity allowing the detection of dopamine in the presence of ascorbic acid, uric acid, L-Cys, serotonin, and glucose, with high sensitivity of reduction currents obtained at −0.2 V (vs. Ag/AgCl). The proposed biosensor allowed the detection of dopamine in commercial serum and artificial urine with recovery values close to 100 %. It also demonstrated reproducibility, reusability, and long-term storage stability. The sensitivity, Kmapp, and Imax values of the biosensor were determined as 2.05 μM and 1.03 μA, respectively. The LAC-Ni/Au/PPy-COOH/NAF/SPE biosensor is a reliable design for detecting dopamine with a wide linear range.  相似文献
