设计了一种单螺旋通道的聚二甲基硅氧烷(Poly(dimethylsiloxane),PDMS)微流控芯片,用于副溶血性弧菌气溶胶的快速有效富集。该芯片的特征在于其通道呈螺旋分布,且通道内部含有均匀分布的鱼骨形结构。结果表明,在不同富集时间段内,采用该芯片方法捕获的细菌总数均远高于传统落板法。对于传统落板法无法有效捕获的低浓度样本(10~4CFU/mL)的缺陷,该方法的优势在于:芯片内部的螺旋通道可增大对气溶胶中微生物的离心力;鱼骨形结构的设计增加了待测样品与芯片内壁间的接触几率。此外,以无鱼骨形的螺旋芯片作为对照,验证了鱼骨形结构对于高效富集的意义。此芯片设计巧妙、易于制备、高效便携、富集效果较好,在气溶胶污染严重的水产加工等场所具有较大的应用前景。 相似文献

Acute toxicity of pesticides in water was assessed singly and in mixtures using the responses of the luminescent bacterium Vibrio fischeri (BioTox™), the aquatic invertebrate Daphnia magna (Daphtoxkit™), and the MitoScan™ assay. The latter utilized fragmented mitochondria to enzymatically convert β-nicotinamide adenine dinucleotide (NADH) to its oxidized form, NAD+. The rate of the conversion being sensitive to type and concentration of toxicants. The pesticides tested were Carbofuran (2,3-dihydro-2,2-dimethylbenzofuran-7-yl methylcarbamate), Cyromazine (N-cyclopropyl-1,3,5-triazine-2,4,6-triamine), Fenamiphos (ethyl 4-methylthio-m-tolyl isopropylphosphoramidate), and Formetanate (3-dimethylaminomethyleneiminophenyl methylcarbamate). The toxicity bioassays were characterized in terms of relative sensitivity, reproducibility, range of the linear response, and the ability to reveal synergistic/antagonistic interactions among toxicants. The D. magna assay was the most sensitive and best able to detect toxic interactions of mixtures. Also, unlike the other assays used, the response of the daphnid system was linear over a 10-fold change in pesticide concentration. Relative to the BioTox™, the MitoScan™ was 2- to 11-fold more sensitive for the compounds and mixtures tested. The EC50 reproducibility of all tests was within ±20% coefficient of variation; however, the lowest observable effect concentration (LOEC) were only reproducible to ±35% on average. Cyromazine was the least toxic of the pesticides tested. To test the predictive value of the concept of concentration addition, toxicities of binary and quaternary mixtures of four different pesticides were analyzed. Synergistic/antagonistic responses were most frequently observed in testing with D. magna. Synergistic/antagonistic effects were seen only in 25 and 50% of the cases with the BioTox™ and the MitoScan™ assays, respectively. 相似文献

研究了浙江省沿海网箱养殖大黄鱼溃疡病病原菌的生物学特性、对化学药物和中草药的敏感性及其胞外产物活性等.从症状典型的网箱养殖患病大黄鱼体内分离到1株可疑细菌824-1,经人工感染试验证实为是引起该病的病原菌.经鉴定,菌株824-1为溶藻弧茵(Vibrio alginolyticus).利用杯碟法研究了其胞外产物的性质,结果表明:其胞外产物具有淀粉酶、明胶酶、酪蛋白酶、卵磷脂酶、脂酶和几丁质酶活性以及溶血活性,其中以淀粉酶、明胶酶和酪蛋白酶的活性最强,但没有脲酶活性.检测了18种化学药物和15种中草药对病原菌的抑菌作用.结果表明,复方新诺明和庆大霉素等2种化学药物和石榴皮、地榆、五味子、大黄等4种中草药的抑菌能力最强,可作为防治该病的有效药物。 相似文献

Summary This work introduces a neural network methodology for developing QSTR predictors of toxicity to Vibrio fischeri. The method adopts the Radial Basis Function (RBF) architecture and the fuzzy means training strategy, which is fast and repetitive,
in contrast to most traditional training techniques. The data set that was utilized consisted of 39 organic compounds and
their corresponding toxicity values to Vibrio fischeri, while lipophilicity, equalized electronegativity and one topological index were used to provide input information to the
models. The performance and predictive ability of the RBF model were illustrated through external validation and various statistical
tests. The proposed methodology can be used to successfully model toxicity to Vibrio fischerifor a heterogeneous set of compounds. 相似文献

Fragmentation mechanisms of electron ionization (EI) mass spectrometry of the title compounds have been elucidated by high-resolution (HR) mass spectrometric measurements of the elemental composition and measurements of the metastable transitions (B(2)/E, CID). The experimental results were interpreted with the help of Mass Frontier 3.0 software, which aided the elucidation of fragmentation mechanisms and helped to deduce structures of the ions formed. Characteristic under the conditions of EI-MS measurement was the production of protonated adducts. Three distinct pathways observed include the formation of oxonium type ions, the conjugated transfer of electrons in the pyranose ring, and cleavage of the acylamide side chains. By applying the results obtained, the molecular mass, as well as the structures of both the saccharide and acylamide side chain involved in related substances, can be determined. 相似文献
