高效液相色谱法同时测定稻田中苄嘧磺隆和苯噻酰草胺残留 总被引:4,自引:0,他引:4
建立了同时测定稻田(稻田土壤、水和植株)中苄嘧磺隆和苯噻酰草胺残留量的高效液相色谱(HPLC)分析方法。稻田水样品用二氯甲烷直接萃取;稻田土壤样品用碱性乙腈-二氯甲烷(1:1, v/v)混合液直接提取;水稻植株样品用碱性二氯甲烷提取后,二氯甲烷提取液经弗罗里硅土柱净化。上述样品溶液采用C18不锈钢色谱柱(150 mm×4.6 mm, 5 μm)分离,流动相为水-甲醇(30:70, v/v),流速为0.5 mL/min,柱温为30 ℃,紫外检测波长为238 nm,外标法定量。苄嘧磺隆和苯噻酰草胺在0.05~5.00 mg/L范围内的线性关系均很好(r>0.9999)。在稻田水、土壤和水稻植株中添加3个水平(0.05, 0.10, 1.00 mg/kg)的苄嘧磺隆和苯噻酰草胺,两者的回收率均在85.39%~113.33%之间,相对标准偏差为0.91%~10.24%。这表明该方法的灵敏度、准确度和精密度均符合农药残留测定的技术要求。 相似文献

Understanding the interactions between proteins and ligands is critical for protein function annotations and drug discovery. We report a new sequence‐based template‐free predictor (TargetATPsite) to identify the Adenosine‐5′‐triphosphate (ATP) binding sites with machine‐learning approaches. Two steps are implemented in TargetATPsite: binding residues and pockets predictions, respectively. To predict the binding residues, a novel image sparse representation technique is proposed to encode residue evolution information treated as the input features. An ensemble classifier constructed based on support vector machines (SVM) from multiple random under‐samplings is used as the prediction model, which is effective for dealing with imbalance phenomenon between the positive and negative training samples. Compared with the existing ATP‐specific sequence‐based predictors, TargetATPsite is featured by the second step of possessing the capability of further identifying the binding pockets from the predicted binding residues through a spatial clustering algorithm. Recently, we developed a convenient microfluidic droplet generation device based on vacuum‐driven fluid manipulation with a piezoelectric diaphragm micropump. In the present study built on our previous work, we investigate the influence of settings applied to the piezoelectric pump, such as peak‐to‐peak drive voltage (Vp‐p) and wave frequency, on droplet generation characteristics. Stepwise adjustments to the drive voltage in ±10‐Vp‐p increments over the range of 200?250 Vp‐p during droplet creation revealed that the droplet generation rate could be reproducibly controlled at a specific drive voltage. The droplet generation rate switched within <0.5 s after the input of a new voltage. Although the droplet generation rate depended on the drive voltage, this setting had almost no influence on droplet size. The frequency over the selected range (50?60 Hz) did not markedly influence the droplet generation rate or droplet size. We show that the current fluid manipulation system can be conveniently used for both droplet generation and for rapid droplet reading, which is required in many microfluidic‐based applications. 相似文献

Filamentous fungi secrete various oxidative enzymes to degrade the glycosidic bonds of polysaccharides. Cellobiose dehydrogenase (CDH) (E.C.1.1.99.18) is one of the important lignocellulose degrading enzymes produced by various filamentous fungi. It contains two stereo specific ligand binding domains, cytochrome and dehydrogenase - one for heme and the other for flavin adenine dinucleotide (FAD) respectively. The enzyme is of commercial importance for its use in amperometric biosensor, biofuel production, lactose determination in food, bioremediation etc. Termitomyces clypeatus, an edible fungus belonging to the basidiomycetes group, is a good producer of CDH. In this paper we have analyzed the structural properties of this enzyme from T. clypeatus and identified a distinct carbohydrate binding module (CBM) which is not present in most fungi belonging to the basidiomycetes group. In addition, the dehydrogenase domain of T. clypeatus CDH exhibited the absence of cellulose binding residues which is in contrast to the dehydrogenase domains of CDH of other basidiomycetes. Sequence analysis of cytochrome domain showed that the important residues of this domain were conserved like in other fungal CDHs. Phylogenetic tree, constructed using basidiomycetes and ascomycetes CDH sequences, has shown that very surprisingly the CDH from T. clypeatus, which is classified as a basidiomycetes fungus, is clustered with the ascomycetes group. A homology model of this protein has been constructed using the CDH enzyme of ascomycetes fungus Myricoccum thermophilum as a template since it has been found to be the best match sequence with T. clypeatus CDH. We also have modelled the protein with its substrate, cellobiose, which has helped us to identify the substrate interacting residues (L354, P606, T629, R631, Y649, N732, H733 and N781) localized within its dehydrogenase domain. Our computational investigation revealed for the first time the presence of all three domains - cytochrome, dehydrogenase and CBM - in the CDH of T. clypeatus, a basidiomycetes fungus. In addition to discovering the unique structural attributes of this enzyme from T. clypeatus, our study also discusses the possible phylogenetic status of this fungus. 相似文献

Since 1983 we have analyzedaqua regia extracts from environmental reference materials of 5 different soils and 4 different sludges by neutron activation analysis. Aqua regia as such is not accepted for irradiation in a nuclear reactor, and therefore the solution has to be evaporated to dryness, mixed and dried to constant weight before an aliquot can be taken and subjected to instrumental neutron activation analysis (INAA). Results for leachable Cr and Zn were found to display greater variability than results for total contents of these elements, and the reasons for this will be discussed.For the 1994 certification campaign for one soil and two sludges we have chosen to determine the total content of trace elements in these reference materials by INAA before and afteraqua regia leaching. The leachable contents are then found indirectly as a difference between the two results; in this way we eliminate the sources of uncertainty associated with the drying and handling of theaqua regia extracts, and we reduce the contributions from counting statistics. 相似文献

The Golay-Giddings and Poiseuille equations are used to derive equations for the calculation of the maximum plate number and minimum time conditions for given columns at fixed, but selectable, outlet pressures. In addition, expressions are presented for the determination of minimum analysis times for separation problems requiring given plate numbers. In this instance, the optimum column length can be calculated as a function of outlet pressure. A Basic computer program, incorporating the equations for the various optima, together with the H-ū curves, is described. Input variables are either column length or desired plate number, column diameter, film thickness, capacity ratio of the solute, column outlet pressure, seperation temperature, and carrier gas. The carrier gas viscosity is automatically calculated in the case of hydrogen, helium, or nitrogen. For these gases, and if the solute is a n-alkane, the diffusivity of the solute in the mobile phase is calculated. In this case, the carbon number of the solute is needed in the computation. For high molecular weight polydimethylsilicone phases (e.g. SE-30), the program can approximate the diffusivity of n-alkanes in the stationary phase at the given temperature as a function of the carbon number. Of course, manually entered values of viscosity and diffusion coefficients can be included in the calculations. 相似文献

催化加氢作用, 当金属组分A和组分B浓度分别为 15 μg/g ~ 25 μg/g 和 1100 μg/g~1300 μg/g时催化剂具有较好的催化加氢活性。两元水溶性催化剂硫化后的XPS、XRD和TEM分析结果表明:活性组分经硫化反应后生成了金属硫化物,但组分A硫化率不足50%;组分B易硫化结晶,其晶粒成为组分A的硫化物晶粒或颗粒分散的“载体”,使组分A的硫化物具有较高的分散度,提高了催化剂的加氢活性;A-B双金属水溶性催化剂经硫化反应后所形成的颗粒尺寸在100 nm左右,说明两元金属催化剂在硫化结晶过程中确实存在相互分散和抑制晶粒增长的作用。 相似文献

应用微量化学法和固相萃取技术 ,建立了粮谷中恶草酮残留量的气相 质谱 (GC MS)测定方法。用苯 正己烷 (体积比为 1∶1)萃取 ,中性氧化铝小柱净化。净化液用GC MS测定 ,采用外标法定量。恶草酮在大米中的回收率为 90 4%~ 115 .7% ,RSD为 2 %~ 6 % ,在玉米中的回收率为 81 3%~ 10 9 7% ,RSD为 4%~ 9% ,最低定量检出限为 0 0 0 5mg/kg。该法快速、灵敏、准确 ,各项技术指标均满足农药残留检测的要求。 相似文献
