A two-step sample preparation technique based on dispersive micro solid-phase extraction combined with coacervative microextraction is presented for preconcentration and determination of tricyclic antidepressant drugs in biological samples. An important feature of the method is the application of hydrophobic magnetic nanoparticles, which in combination with coacervative microextraction method enables development of rapid and efficient extraction procedure in order to achievement of a high extraction efficiency. Simultaneous optimization by experimental design lead to improvement of method with low cost which supply useful information about interaction among variables. Under the optimized conditions, a linear range of 5–1000 ng mL−1 with detection limits from 0.51 to 1.4 ng mL−1 were obtained for target analytes. The method was successfully used for the determination of analytes in biological fluids (plasma and urine) with relative recoveries in the range of 89–105% (RSDs < 3.5%).  相似文献

Natural enantiomers of unique tricyclic sesquiterpenoids, valeriananoids A-C 1-3, have been synthesized starting from bicyclo[2.2.2]octane-2,5-dione derivative 11, which was obtained by diastereoselective catalytic domino Michael reaction of oxophorone 5 with 8-phenylmenthyl acrylate 10 by LDA or silica-gel-base (NMAP-Li). The tricyclic ring was closed selectively by intramolecular 6-endo-trig mode cyclization of the ketyl radical, which was generated from keto-allylether 25 by either lithium or sodium naphthalenide.  相似文献

The chromatographic behavior of seven tricyclic antidepressants (amitryptiline, clomipramine, doxepin, imipramine, maprotiline, nortryptiline, and trimipramine) was examined with micellar mobile phases containing the nonionic surfactant Brij-35. Acetonitrile-water mixtures were also used for comparison purposes. Tricyclic antidepressants are moderately polar basic drugs, which are positively charged in the usual working pH. This gives rise to a strong association with the alkyl chains and residual ionized silanols in silica-based stationary phases, which is translated in a high consumption of organic solvent to get appropriate retention times. Brij-35 modifies the surface of the stationary phases creating a neutral bilayer that masks silanols and reduces the polarity. Consequently, the retention times are decreased. A simple chromatographic procedure for the control of tricyclic antidepressants in pharmaceutical formulations was developed, using 0.02 M Brij-35 at pH 3 and UV detection. Satisfactory recoveries were achieved, with intra- and inter-day relative standard deviations usually below 1 and 2%, respectively. The preparation of the samples was simple and only required solubilization and filtration steps previous to injection. The proposed procedure has the advantage of not using an organic solvent in the mobile phase, and the biodegradable character of Brij-35. This makes an example of "green" liquid chromatographic analysis.  相似文献

Summary Solid-phase microextraction (SPME) is a solvent-free sample-preparation technique that enables isolation and pre-concentration of analytes from a sample on a thin film coating a fused-silica fiber. In this study SPME coupled with microcolumn liquid chromatography (micro LC) has been used for the determination of four tricyclic antidepressants (amitriptyline, imipramine, nortriptyline, and desipramine) in human urine. SPME conditions which affect extraction efficiency were optimized, and under the optimum conditions the system was a few hundred times more sensitive than direct LC analysis without SPME. For amitriptyline the detection limit was 3 ng mL−1 and the calibration curve was linear in the range of 5–500 ng mL−1. The SPME-micro LC method has been applied to the analysis of amitriptyline in patient’s urine.  相似文献

It is generally assumed that selective serotonin reuptake inhibitors (SSRIs) induce antidepressant activity by inhibiting serotonin (5-HT) reuptake transporters, thus elevating synaptic 5-HT levels and, finally, ameliorates depression symptoms. New evidence indicates that SSRIs may also modulate other neurotransmitter systems by inhibiting neuronal nicotinic acetylcholine receptors (nAChRs), which are recognized as important in mood regulation. There is a clear and strong association between major depression and smoking, where depressed patients smoke twice as much as the normal population. However, SSRIs are not efficient for smoking cessation therapy. In patients with major depressive disorder, there is a lower availability of functional nAChRs, although their amount is not altered, which is possibly caused by higher endogenous ACh levels, which consequently induce nAChR desensitization. Other neurotransmitter systems have also emerged as possible targets for SSRIs. Studies on dorsal raphe nucleus serotoninergic neurons support the concept that SSRI-induced nAChR inhibition decreases the glutamatergic hyperstimulation observed in stress conditions, which compensates the excessive 5-HT overflow in these neurons and, consequently, ameliorates depression symptoms. At the molecular level, SSRIs inhibit different nAChR subtypes by noncompetitive mechanisms, including ion channel blockade and induction of receptor desensitization, whereas α9α10 nAChRs, which are peripherally expressed and not directly involved in depression, are inhibited by competitive mechanisms. According to the functional and structural results, SSRIs bind within the nAChR ion channel at high-affinity sites that are spread out between serine and valine rings. In conclusion, SSRI-induced inhibition of a variety of nAChRs expressed in different neurotransmitter systems widens the complexity by which these antidepressants may act clinically.  相似文献

Sodium sulfate‐induced deep eutectic solvent–based solidification of floating organic droplets–dispersive liquid phase microextraction was developed prior to gas chromatography–mass spectrometry. In this method, a mixture of Na2SO4 solution (as phase separation agent and disperser) containing menthol–decanoic acid was rapidly injected into an alkaline aqueous solution containing Na2SO4. The solution was placed in an ice bath and the menthol–decanoic acid solvent was solidified on the surface of the solution. Under the optimal conditions, the enrichment factors and extraction recoveries were 122–147 and 74–89%, respectively. Finally, an aliquot of the collected organic phase was removed and mixed with acetonitrile and injected into the separation system. The limits of detection and lower limits of quantification were obtained at the ranges of 13–25 and 24–41 ng L?1, respectively. The relative standard deviations of the proposed method were ≤11% for intra‐ and inter‐day precisions at four concentrations.  相似文献

A semiautomatic extraction-fluorimetric method for the determination of tricyclic antidepressant drugs (TCAs) based in the formation of ion pairs with 9,10-dimethoxyanthracene-2-sulphonate (DMAS) has been developed. The aqueous solutions of the TCAs (imipramine, desipramine, amitriptyline, nortriptyline, clomipramine or doxepine) are injected into a carrier composed by DMAS in an acid medium and the ion pair formed is extracted into dichloromethane where the fluorescence is measured. An experimental design (Central Composite Design) together with the Response Surface Methodology has been used to find the optimal instrumental FIA and chemical variables. We have considered as the response function the product of the peak height by the sampling frequency. The calibration curves were linear over the working range (0.25-3.00 mg L−1). The limits of detection were lower than 0.30 mg L−1. The method has been satisfactorily applied to the determination of imipramine, amitriptyline, clomipramine and doxepin in pharmaceutical preparations.  相似文献
