A simple, effective, and robust magnetic solid‐phase extraction method was developed using magnetite/reduced graphene oxide nanoparticles as the adsorbent for the simultaneous determination of Sudan dyes (I, II, III, and IV) in foodstuffs. The magnetite/reduced graphene oxide nanoparticles were characterized by X‐ray diffraction, scanning electron microscopy, and vibrating sample magnetometry. The extraction parameters including extraction time, elution solution, and elution time and volume were investigated in detail. Such magnetite/reduced graphene oxide nanoparticles based magnetic solid‐phase extraction in combination with high‐performance liquid chromatography and variable wavelength detection gave the detection limits of 3–6 μg/kg for Sudan I–IV in chili sauce, tomato sauce, chili powder, and chili flake samples. The recoveries were 79.6–108% at three spiked levels with the intra‐ and inter‐day relative standard deviations of 1.2–8.6 and 4.5–9.6%, respectively. The feasibility was further performed by a comparison with commercial alumina‐N. This method is suitable for the routine analysis of Sudan dyes due to its sensitivity, simplicity, and low cost. 相似文献

Changes of antioxidative enzymes and cell membrane osmosis in tomato colonized by arbuscular Mycorrhizae under NaCl stress 总被引:7,自引:0,他引:7
Salinity toxicity is a worldwide agricultural and eco-environmental problem. Many literatures show that arbuscular mycorrhizal fungi (AMF) can enhance salt tolerance of many plants and some physiological changes occurred in AM symbiosis under salt stress. However, the role of ROS-scavenging enzymes in AM tomato is still unknown in continuous salt stress. This study investigated the effect of Glomus mosseae on tomato growth, cell membrane osmosis and examined the antioxidants (superoxide-dismutase, SOD; catalase, CAT; ascorbate peroxidase, APX; peroxidase, POD) responses in roots of mycorrhizal tomato and control under different NaCl stress for 40 days in potted culture. NaCl solution (0, 0.5 and 1%) was added to organic soil in the irrigation water after 45 days inoculated by AMF (Glomus mosseae). (1) AMF inoculation improved tomato growth under salt or saltless condition and reduced cell membrane osmosis, MDA (malonaldehyde) content in salinity. So the salt tolerance of tomato was enhanced by AMF; (2) SOD, APX and POD activity in roots of AM symbiosis were significantly higher than corresponding non-AM plants in salinity or saltless condition. However, CAT activity was transiently induced by AMF and then suppressed to a level similar with non-AM seedlings; (3) higher salinity (1% level) and long stress time suppressed the effect of AMF on SOD, APX, POD and CAT activity; (4) this research suggested that the enhanced salt tolerance in AM symbiosis was mainly related with the elevated SOD, POD and APX activity by AMF which degraded more reactive oxygen species and so alleviated the cell membrane damages under salt stress. Microalgae consortia were photoautotrophically cultivated in sequencing batch photobioreactors (SBPRs) with an alteration of the normal growth and starvation (nutrient limitation) phases to select consortia capable of polyhydroxyalkanoate (PHA) accumulation. At the steady state of SBPR operation, the obtained microalgae consortia, selected under nitrogen and phosphate limitation, accumulated up to 11.38% and 10.24% of PHA in their biomass, which was identified as poly(3-hydroxybutyrate) (P3HB). Photoautotrophic and mixotrophic batch cultivation of the selected microalgae consortia was conducted to investigate the potential of biomass and PHA production. Sugar source supplementation enhanced the biomass and PHA production, with the highest PHA contents of 10.94 and 6.2%, and cumulative PHA productions of 100 and 130 mg/L, with this being achieved with sugarcane juice under nitrogen and phosphate limitation, respectively. The analysis of other macromolecules during batch cultivation indicated a high content of carbohydrates and lipids under nitrogen limitation, while higher protein contents were detected under phosphate limitation. These results recommended the selected microalgae consortia as potential tools for PHA and bioresource production. The mixed-culture non-sterile cultivation system developed in this study provides valuable information for large-scale microalgal PHA production process development following the biorefinery concept. 相似文献

This study was performed to evaluate the responses of Escherichia coli 0157:H7 inoculated in an apple-carrot blended juice to manothermosonication (MTS) treatments. The MTS treatments were conducted in a continuous-flow MTS system. The juice samples were exposed to ultrasound treatment at combinations of three temperatures (60, 50 and 40 °C) and three pressure levels (100, 200, and 300 kPa) for five residence times (15, 30, 45, 60, and 75 s). The results showed that higher treatment temperature (i.e. 60 °C) and hydrostatic pressure in the MTS system significantly enhanced the microbial reduction. A FDA mandated 5-log CFU/ml reduction of E. coli 0157:H7 for juice processing was achieved in 30 s for MTS treatment at 60 °C, in comparison to 60 s at 50 °C. The Weilbull and Log-logistic models provided the best fitting of the inactivation data for the MTS treatments. Extensive damage of E. coli 0157:H7 cells treated with MTS was observed on micro-images of scanning electron microscopy and transmission electron microscopy. 相似文献

为了快速、准确鉴别预包装纯菠萝汁是否掺有外源性糖,采用傅里叶变换衰减全反射中红外光谱技术采集900~1 500 cm-1范围内不同批次的预包装纯菠萝汁样品和掺入甜菜糖浆、大米糖浆、木薯糖浆的菠萝汁掺假样品的中红外光谱共计234例,以线性判别分析和支持向量机分析为掺假鉴别模型的建模方法,比较了全波段谱图与通过主成分载荷系数分析选取特征波长图谱的两种掺假鉴别模型。研究表明,全波段图谱的线性判别分析和支持向量机分类模型对验证集的判断正确率均高于88%;选取8个特征波长之后,线性判别分析模型验证集判断正确率提高至96.15%,支持向量机模型验证集判断正确率提高至94.87%,且模型输入变量由312个减少到8个。利用傅里叶变换衰减全反射中红外光谱技术结合化学计量学方法选取特征波长后建立的模型可以较好的应用于预包装纯菠萝汁外源糖的鉴别。 相似文献

提出了利用可见/近红外高光谱成像技术检测高温障碍胁迫下番茄叶片色差的方法。首先采集380~1 023 nm波段范围内60个高温障碍胁迫和60个健康番茄叶片的高光谱图像,同时获取全部叶片的色差值(L*, a*和b*),然后提取所有样本的高光谱图像中感兴趣区域(region of interest, ROI)的光谱反射率值。基于不同预处理方法建立偏最小二乘(partial least squares, PLS)预测模型,再利用连续投影算法(successive projections algorithm, SPA)提取特征波长并建立SPA-PLS预测模型。最后分别基于全波段和特征波段建立偏最小二乘-判别分析(partial least squares-discriminant analysis, PLS-DA)模型。结果显示,全波段中基于原始光谱信息建立的模型效果最好,3个色差值的预测集决定系数(determination coefficient, R2)分别是0.818,0.109和0.896;基于特征波长建立的模型预测集R2分别是0.591,0.244和0.673;所有模型预测集的总体识别率均大于77.50%。结果表明,可见/近红外高光谱成像技术检测番茄叶片色差值(L*和b*)和识别高温障碍样本是可行的。 相似文献

采用高效液相色谱-紫外-荧光和高效液相色谱-紫外-质谱联用法,证明胃癌患者胃液中存在的荧光物质主要为色氨酸.建立了反相高效液相色谱-双波长同时紫外检测法定量分析胃液中色氨酸和利多卡因的方法.使用Kromasil C18柱(150 mm×4.6 mm, 5 μm),以含0.1%三氟乙酸的甲醇溶液与含0.1%三氟乙酸水溶液为流动相,梯度洗脱分析.色氨酸使用278 nm检测,浓度在0.5~200 mg/L范围内峰面积对浓度的线性相关系数为0.9994;利多卡因使用254 nm检测,浓度在20~5000 mg/L范围内相关系数为0.9992.色氨酸和利多卡因的检出限分别为0.15和5 mg/L;平均加标回收率分别为70.8%~110.4%(色氨酸) 和87.1%~116.2% (利多卡因).利用本方法对38例胃癌患者和48例非胃癌患者的胃液进行测试,两组胃液中色氨酸的含量存在一定的差异. 相似文献

Organic acids, sugars, phenolic compositions and antioxidant capacities of orange juice and orange wine obtained from the cv. Kozan of Turkey were determined. High-performance liquid chromatographic methods were used to identify and quantify of these compounds. Three organic acids (citric, malic and ascorbic acids) and three sugars (sucrose, glucose and fructose) were determined. The major organic acid was found as citric acid. With regard to sugars, sucrose was present in the largest amounts for orange juice and wine. A total of 13 phenolic compounds were identified and quantified in orange juice and wine, including hydroxybenzoic acids (2), hydroxycinnamic acids (5), and flavanones (6). Hesperidin, narirutin and ferulic acid were the most abundant phenolic compounds in orange juice and wine. Antioxidant activities of orange juice and wine were measured using the DPPH• (2,2-diphenyl-1-picrylhydrazyl) assay, and the antioxidant capacity of orange juice was found to be higher than that of orange wine. 相似文献
