固相萃取-气相色谱法测定北京市水样中的邻苯二甲酸酯   总被引:16,自引:0,他引:16  
研究了使用毛细管柱(OV-1701)分离、FID作为检测器的气相色谱法测定水中5种邻苯二甲酸酯(PAEs)[邻苯二甲酸二乙酯(DEP),邻苯二甲酸二丁酯(DBP),邻苯二甲酸丁基苄基酯(BBP),邻苯二甲酸二(2-乙基己基)酯(DEHP),邻苯二甲酸二环己酯(DCHP)]的色谱条件。5种PAEs检出限在0.1~0.3μg/L之间,所测PAEs的质量浓度在0.5~100μg/L范围内,RSD在2.2%~3.3%之间,各物质校正曲线(质量浓度与峰面积)的相关系数在0.9970~0.9993之间。利用固相萃取技术萃取水中PAEs,回收率在82.5%~110.5%之间,RSD在1.1%~4.0%之间。北京市吴家村污水处理厂4个采样点中邻苯二甲酸酯浓度在(0.7~193.3)μg/L之间。北京市南护城河的3个采样点中邻苯二甲酸酯浓度在(0.1~241.8)μg/L之间。  相似文献

Thermodynamics of the interaction between Ni2+ and human growth hormone (hGH) were determined at 27 °C in Nail solution by isothermal titration calorimetry. A new method to predict protein penetration and the effect of metal ions on the stability of proteins is introduced. The new solvation model was used to reproduce the enthalpies of Ni2+-hGH interaction over the whole range of Ni2+ concentrations. The solvation parameters recovered from the new equation, attributed to the structural change of hGH and its biological activity.  相似文献

In this study, the steroid hormone levels in river and tap water samples were determined by using a novel dispersive liquid-liquid microextraction method based on the solidification of a floating organic drop (DLLME-SFO). Several parameters were optimized, including the type and volume of the extraction and dispersive solvents, extraction time, and salt effect. DLLME-SFO is a fast, cheap, and easy-to-use method for detecting trace levels of samples. Most importantly, this method uses less-toxic solvent. The correlation coefficient of the calibration curve was higher than 0.9991. The linear range was from 5 to 1000 μg L−1. The spiked environmental water samples were analyzed using DLLME-SFO. The relative recoveries ranged from 87% to 116% for river water (which was spiked with 4 μg L−1 for E1, 3 μg L−1 for E2, 4 μg L−1 for EE2 and 9 μg L−1 for E3) and 89% to 102% for tap water (which was spiked with 6 μg L−1 for E1, 5 μg L−1 for E2, 6 μg L−1 for EE2 and 10 μg L−1 for E3). The detection limits of the method ranged from 0.8 to 2.7 μg L−1 for spiked river water and 1.4 to 3.1 μg L−1 for spiked tap water. The methods precision ranged from 8% to 14% for spiked river water and 7% to 14% for spiked tap water.  相似文献

Side‐effects from allograft, limited bone stock, and site morbidity from autograft are the major challenges to traditional bone defect treatments. With the advance of tissue engineering, hydrogel injection therapy is introduced as an alternative treatment. Therapeutic drugs and growth factors can be carried by hydrogels and delivered to patients. Abaloparatide, as an analog of human recombinant parathyroid hormone protein (PTHrp) and an alternative to teriparatide, has been considered as a drug for treating postmenopausal osteoporosis since 2017. Since only limited cases of receiving abaloparatide with polymeric scaffolds have been reported, the effects of abaloparatide on pre‐osteoblast MC3T3‐E1 are investigated in this study. It is found that in vitro abaloparatide treatment can promote pre‐osteoblast MC3T3‐E1 cells’ viability, differentiation, and mineralization significantly. For the drug delivery system, 3D porous structure of the methacrylated gelatin (GelMA) hydrogel is found effective for prolonging the release of abaloparatide (more than 10 days). Therefore, injectable photo‐crosslinked GelMA hydrogel is used in this study to prolong the release of abaloparatide and to promote healing of defected bones in rats. Overall, data collected in this study show no contradiction and imply that Abaloparatide‐loaded GelMA hydrogel is effective in stimulating bone regeneration.  相似文献

Summary The synthesis of the heptacosapeptide amide with the primary structure of Human-secretin is described. For this purpose 7 fragments were designed, i.e. H-Gly-Leu-Val-NH2 25–27b,Z-Arg(Z 2)-Leu-Leu-Gln-OH 21–24,Z-Arg(Z 2)-Leu-Gln-OH 18–20,Z-Arg(Z 2)-Glu(OtBu)-Gly-Ala-OH 14–17,Z-Arg(Z 2)-Leu-OH 12–13,Z-Thr(tBu)-Ser(tBu)-Glu(OtBu)-Leu-Ser(tBu)-OH 7–11,Adoc-His(Adoc)-Ser(tBu)-Asp(OtBu)-Gly-Thr(tBu)-Phe-OH 1–6 these fragments were consequently assembled to the overall protected total sequence using the Wünsch/Weygand-method with dicyclohexylcarbodiimide. After deprotection by exposure to trifluoroacetic acid in presence of 1,2-ethanedithiol and water as scavenger, the isolated crude product was purified by column chromatography on CM-Sepharose, fast flow. This synthetized Human-secretin showed the full biological activity in comparison to Porcine-secretin.

We describe an accurate method for protein quantification based on conventional acid hydrolysis and an isotope dilution-HPLC-mass spectrometry (ID-HPLC-MS) method. Sample purity was confirmed using capillary zone electrophoresis, HPLC and MS. The analyte protein, human growth hormone (hGH), was effectively hydrolyzed by incubation with 8 M hydrochloric acid at 130 °C for 48 h, where at least 1 μM of hGH was treated to avoid possible degradation of released amino acids during hydrolysis. Using a reversed-phase column, the analytes (isoleucine, phenylalanine, proline and valine) were separated within 5 min using an isocratic eluent comprising 10% acetonitrile containing 0.1% trifluoroacetic acid. The detection limit (signal to noise ratio of 3) of amino acids was 5.5-6.2 fmol per injection. The quantification precision (RSD) of amino acids for intra- and inter-day assays was less than 0.98% and 0.39%, respectively. Comparison with other biochemical and instrumental methods revealed substantially higher accuracy and reproducibility of the ID-HPLC-MS/MS method as expected. The optimized hydrolysis and analytical conditions in our study were suitable for accurate quantification of hGH.  相似文献

建立了猪血浆中雌激素、雄激素、皮质激素、孕激素以及部分违禁添加激素的高效液相色谱-串联质谱(HPLC-MS/MS)检测方法。以叔丁基甲醚为提取溶剂,涡旋振荡进行提取,采用Xselect-C18和Extend-C18(3.5μm,2.1 mm×100 mm)色谱柱进行目标物分离,外标法定量。结果表明,33种类固醇激素在其线性浓度范围内线性关系良好,相关系数均大于0.99;在3个不同浓度下的加标回收率为50.9%~119.5%,相对标准偏差为2.3%~9.8%,定量下限(LOQ)为0.05~40.00 ng/mL。采用所建立方法对北京某大型养猪场所采样品进行检测,共有10种激素被测出。该方法操作简单、灵敏度高、重现性好,可满足对猪血浆中类固醇激素的检测和验证要求。  相似文献
