Fluorescence quenching and synchronous fluorescence methods were used to study the interactions of fluore-scence-active quercetin (Qct) with casein (Cas) and bovine serum albumin (BSA) in phosphate buffer solution (PBS, pH=7.4) with or without coexisting carbon nanotubes (CNTs). Formulae for binding constant (K) and molar binding ratio (n) were established for methods 1 (fixing protein concentration, changing Qct concentration, and monitoring the fluorescence of protein) and 2 (fixing Qct concentration, changing protein concentration, and monitoring the fluorescence of Qct), to which values of K and n were calculated via nonlinear least-square fitting of the experimental data, and the “optical inner filtering induced fluorescence quenching” effect was thus quantitatively evaluated. The quenching effects of coexisting CNTs on the fluorescence of Qct, BSA, and Cas, as well as the effects of coexisting CNTs on Qct-BSA and Qct-Cas interactions, were examined. Synchronous fluorescence was also used to examine the effects of coexisting CNTs and Qct on the conformations of BSA and Cas, with relevant K and n values for tyrosine (Tyr) and tryptophan (Trp) residues estimated. It was concluded that the CNTs mainly interacted with the Trp residues locating near the protein surfaces, but small-sized Qct molecules could further interact with the Tyr residues locating inside the protein molecules.  相似文献

研究了电子受体氯冉酸(CL)和2,3 二氰5,6 二氯1,4 对苯醌(DDQ)与电子给体氧氟沙星之间的荷移反应。实验发现,氧氟沙星与以上两种电子受体在氯仿中可生成稳定的n π络合物,其荧光强度较之氧氟沙星分别增强了22和54倍。据此建立了两种基于荷移反应简便可靠地测定氧氟沙星的荧光光谱新方法。CL法和DDQ法测定氧氟沙星的质量浓度分别为0 06~2.4μg/mL和0 16~2.0μg/mL时,荧光强度与质量浓度呈良好的线性关系。该方法已用于药物制剂中氧氟沙星的测定,其回收率分别为98.4%~100.9%和98.5%~100.4%。  相似文献

在pH=6.00的KH2PO4-NaOH缓冲溶液中,波长差△λ=35 nm,采集260~325 nm波长范围内的同步荧光光谱数据,并对该数据进行一阶求导后用偏最小二乘法处理,以此建立了偏最小二乘-导数同步荧光光谱法同时测定对羟苯基乳酸(PHPLA)、多巴(Dopa)和尿黑酸(HGA)的新方法.该三种物质的线性范围分别为0.06~3.00、0.06~3.00和0.08~3.00 mg/L;检出限分别为0.0307、0.0310和0.0428mg/L.该法用于细胞培养液中PHPLA、Dopa和HGA三组分的同时测定,其测定结果与高效液相色谱法的测定结果相比无显著性差异.  相似文献

A simple, rapid and sensitive synchronous fluorescence method is put forward for the determination of enrofloxacin (ENRO) in the pharmaceutical formulation and its residue in milk based on the yttrium (III)-perturbed luminescence. When Y3+ is added into the ENRO solution, the fluorescence of ENRO is significantly enhanced. The synchronous fluorescence technology is employed in the method to determine trace amount of ENRO residue in milks. The synchronous fluorescence intensity of the system is measured in a 1-cm quartz cell with excitation wavelength of 328 nm, Δλ = 80 nm. A good linear relationship between the fluorescence intensity and the ENRO concentration is obtained in the range of 1.0 × 10−9 to 2.0 × 10−6 mol L−1 (r2 = 0.9992). The limit of detection (LOD) of this method attains as low as 3.0 × 10−10 mol L−1 (S/N = 3). The selectivity of this method is also very good. Common metal ions, rare-earth ions and some pharmaceuticals, which are usually used together with ENRO, do not interfere with the determination of ENRO under the actual conditions. The proposed method can be applied to determine ENRO residue in milks, and limit of quantification (LOQ) determined in the spiked milk is estimated to be 2.8 × 10−8 mol L−1 (10 μg L−1). Moreover, this method can be used as a rapid screening for judging whether the ENRO residues in milks exceed Minimal Risk Levels (MRLs) or not. General Stochastic Hybrid System (SHS) are characterised by Stochastic Differential Equations (SDEs) with discontinuities and Poisson jump processes. SHS are useful in model based design of Cyber-Physical System (CPS) controllers under uncertainty. Industry standard model based design tools such as Simulink/Stateflow® are inefficient when simulating, testing, and validating SHS, because of dependence on fixed-step Euler–Maruyama (EM) integration and discontinuity detection. We present a novel efficient adaptive step-size simulation/integration technique for general SHSs modelled as a network of Stochastic Hybrid Automatons (SHAs). We propose a simulation algorithm where each SHA in the network executes synchronously with the other, at an integration step-size computed using adaptive step-size integration. Ito’ multi-dimensional lemma and the inverse sampling theorem are leveraged to compute the integration step-size by making the SDEs and Poisson jump rate integration dependent upon discontinuities. Existence and convergence analysis along with experimental results show that the proposed technique is substantially faster than Simulink/Stateflow®when simulating general SHSs.  相似文献

Viral hepatitis is a common infectious disease caused by five viruses (hepatitis virus A, B, C, D, and E). Given the diversity of hepatitis virus, rapid screening and accurate typing of viral hepatitis are the prerequisites for hepatitis therapy. Here, a multicolor fluorescence system was constructed by combining with the multi-color fluorescence properties of CdSe/ZnS quantum dots (QDs, emission wavelengths: 525 nm, 585 nm and 632 nm) and the broad-spectrum fluorescence quenching performance of GO. Taking advantage of the specific recognition of ssDNA modified CdSe/ZnS QDs to target hepatitis virus DNA, the constructed system could effectively distinguish hepatitis A virus DNA (HAV-DNA), hepatitis B virus DNA (HBV-DNA), and hepatitis C virus DNA (HCV-DNA) in a homogeneous solution. Based on the different adsorption property of GO for ssDNA and dsDNA, the fluorescence Forster resonance energy transfer (FRET) process between ssDNA modified QDs and GO could be regulated. The fluorescence signal of the constructed system presented a sensitive response to HAV-DNA, HBV-DNA, and HCV-DNA content in the range of 1.0–192 nM, 8.0–192 nM, and 1.0–128 nM, respectively. The limit of detection for HAV-DNA, HBV-DNA, and HCV-DNA is 0.46 nM, 1.53 nM, and 0.58 nM. The constructed system can be used to screen hepatitis virus DNA in real samples, which provides an alternative strategy for rapid screening and diagnosis of viral hepatitis.  相似文献

The supramolecular interaction of β-cyclodextrin dimer with berberine hydrochloride was studied in aqueous KH2PO4-H3PO4 buffer solution of pH 2.00 at room temperature by spectrofluorimetry. The apparent association constant of the complex was 1.53 × 104 L mol−1. Based on the significant enhancement of fluorescence intensity of supramolecular sandwich complexes, a spectrofluorimetric method with high sensitivity and selectivity was developed for the determination of berberine hydrochloride in aqueous solution in presence of ethylenediamine linked β-CD dimer. The linear range of the method was 12.8-1.00 × 104 ng mL−1 with the detection limit 3.6 ng mL−1. There was no interference from the normally used in tablets and serum constituents. The proposed method was successfully applied to the determination of berberine hydrochloride in tablets and serum. And then it has a promising potential in therapeutic drug monitoring, pharmokinetis and clinical application.  相似文献
