A high-performance liquid chromatographic method for the determination of wogonoside in plasma of rats administrated orally with the traditional Chinese medicinal preparation Huang-Lian-Jie-Du decoction was developed. Sample preparation was carried out by protein precipitation with a mixture of acetonitrile and methanol (1:1, v/v). The extracted sample was separated on a Hypersil C(18) (150 x 5 mm i.d., 5 microm) analytical column by linear gradient elution using 0.05% (v/v) phosphoric acid (containing 5 mm sodium dihydrogen phosphate) and acetonitrile as mobile phase at a flow rate of 1.5 mL/min. The eluate was detected using a UV detector at 276 nm. The assay was linear over the range 0.109-7.0 microg/mL (R(2) = 0.9999, n = 5). Mean recovery was determined as 98.39%. Intra- and inter-day precisions (RSD) were < or =7.59%. The limit of quantitation was 0.109 microg/mL. After validation, the HPLC method developed was applied to investigate the preliminary pharmacokinetics of wogonoside in rat after oral administration of Huang-Lian-Jie-Du decoction.  相似文献

Abstract

In Tunisia, Pelargonium graveolens is widely consumed as a food aromatizing hydrosol. Recent studies have shown the potential of plant solvent-free extracts as food and pharmaceutical natural additives. Accordingly, in this study, we investigate the phenolic content, the volatile fractions of green P. graveolens extracts such as infusion and decoction, and we evaluate their biological activities. The total phenolic content of the infusion (27.05?mg GAE/gDM) is significantly different from that of decoction (31.2?mg GAE/gDM). The GC-MS analysis identified about twenty volatile components in both extracts. The DPPH inhibition and the β-carotene bleaching tests of the infusion and the decoction had considerable results. Besides, infusion and decoction exhibited a relatively high anti-acetyl-cholinesterase activity and a considerable antimicrobial activity against S. aureus, among three tested pathogenic bacteria.  相似文献

采用高效液相色谱-二极管阵列检测/质谱(HPLC-DAD/MS)联用技术,以10 mmol/L醋酸铵和乙腈混合溶液梯度洗脱 系统为流动相,应用C18色谱柱对生脉饮煎剂中人参皂甙类成分进行分离鉴定。分析结果表明:生脉饮煎剂中主要含有17个 人参皂甙类成分,即20(R)-人参皂甙Rh1、Rh2、Rg3、Rg2,20(S)-人参皂甙Rh1、Rh2、Rg3、Rg2,人参皂甙Rf、Rg6、Rg5 、F4、Rk1、Rk3、Rh4,20(S)-和20(R)-原人参三醇。人参皂甙成分在煎煮过程中发生了很大变化,主要变成了一些中低 极性产物,这是因为煎煮过程中发生了水解、差向异构、脱水等反应。该方法简便、精确、灵敏度高,可以用来分析生脉 饮煎剂中人参皂甙的变化。  相似文献

In this paper, to evaluate the effect of the region of origin on the quality consistency of Shaoyao‐Gancao Decoction (SGD), the SGD fingerprint was developed for the first time. Chemometric methods including similarity analysis, hierarchical clustering analysis and principal component analysis were employed to study the quality consistency of SGD. Meanwhile, high‐performance liquid chromatography coupled with electrospray ionization quadrupole time‐of‐flight mass spectrometry was applied for comprehensive analysis of SGD and 93 compounds were tentatively characterized. Furthermore, a high‐performance liquid chromatography method with multi‐wavelength switching for simultaneous determination of 16 characteristic ingredients comprising gallic acid, oxypaeniflorin, albiflorin, paeoniflorin, liquiritin apioside, liquiritin, isoliquiritin apioside, galloylpaeoniflorin, 1,2,3,4,6‐penta‐O‐galloyl‐d ‐galactopyranose (PGG), ononin, isoliquiritin, liquiritigenin, benzoylpaeoniflorin, glycyrrhizic acid, isoliquiritigenin and formononetin, was established. All 16 analytes show excellent linearity (R2 ≥ 0.9990) with recoveries ranging from 96.58 to 104.61% and limits of detection and quantification of 0.022–0.291 and 0.037–0.635 μg/mL, respectively. Finally, it was successfully applied to determine 15 batches of SGD. The results of our research indicate that different regions of origin have a significant effect on the quality consistency of SGD, and its fingerprint combined with chemometrics and multi‐ingredient determination comprise an efficient and reliable approach for quality consistency evaluation.  相似文献

Traditional Chinese medicine targeted at gut microbiota has good effects in relieving the clinical manifestation of Alzheimer's disease, and intestinal metabolites are considered as a bridge of communication between the brain-gut axis. In order to explore the molecular mechanism of Ganmaidazao decoction treatment, first, the model rats induced by Aβ25-35 and d-gal were used to test the therapy of Ganmaidazao extract using the Morris Water Maze, Western Blot and Elisa. Then the 16S rDNA gene sequencing of the gut microbiota as well as UPLC-QTOF/MS-based metabolomic analysis of feces were carried out. Last, the relationship between Alzheimer's disease, gut microbiota and metabolites was analyzed. Results showed that the abundance and diversity of gut microbiota were rescued and the changes of fecal metabolites in rats with Alzheimer's disease were reversed after Ganmaidazao decoction administration, which were mainly related to lipid metabolism, steroid hormone metabolism, energy metabolism, amino acid metabolism and bile acid metabolism. After associating with Spearman’s correlation analysis, we concluded that gut microbiota and metabolites were closely related and Ganmaidazao decoction could interfere with the balance of gut microbiota and their corresponding metabolites to exert anti- Alzheimer’s disease effect. Combined with PICRUSt2 functional prediction of gut microbiota and metabolomics results, phenylalanine metabolism has been focused as a key metabolic pathway, and Ganmaidazao decoction can reduce the abnormal accumulation of phenylalanine and phenylpyruvate and promote their metabolism by restoring the activity of phenylalanine hydroxylase. This integrated omics approach has potential roles in understanding the complex mechanisms of Ganmaidazao decoction in treating Alzheimer’s disease.  相似文献
