Doubly fuctionalized polar norbornenes bearing the cyano and ester groups in 2,3‐positions are synthesized and enantiomers are separated by high performance liquid chromatography (HPLC) with a chiral stationary phase. These optically active monomers are polymerized by ruthenium carbene catalysts, and high yields of the polymers were obtained. The chiral monomer bearing ethyl ester gave an optically active polymer of lower, but opposite sign of optical rotation (monomer [α]D = +61.0°, polymer [α]D = ?3.1°). 3‐Phenyllactic acid is an antimicrobial compound with broad‐spectrum activity against various bacteria and fungus. The observed difference in pharmacological activity between optical isomeric 3‐phenyllactic acid necessitates a method for enantioseparation. Chiral ligand exchange countercurrent chromatography was investigated for the enantioseparation of 3‐phenyllactic acid with a synthesized chiral ligand. A two‐phase solvent system was composed of n‐butanol/hexane/water (0.4:0.6:1, v/v/v) to which Nn‐dodecyl‐l ‐hydroxyproline was added to the organic phase as chiral ligand and cupric acetate was added in the aqueous phase as a transitional metal ion. The influence factors were optimized by enantioselective liquid–liquid extraction. Baseline enantioseparation of racemic 3‐phenyllactic acid by analytical high‐speed countercurrent chromatography was achieved. The optical purities of enantiomeric 3‐phenyllactic acid reached 99.0%, as determined by chiral high‐performance liquid chromatography.  相似文献

以L-色氨酸(L-trp)和1,2-二(4-吡啶基)乙烯(bpe)为配体与钴离子反应,通过水热法合成了一种具有二维手性层面结构的手性金属-有机骨架(MOFs)材料{[Co(L-trp)(bpe)(H2O)]·H2O· NO3}n;利用X射线粉末衍射(XRD)分析和扫描电子显微镜(SEM)对材料的结构和形貌进行了表征.将该MOFs材料用作固定相,制备了液相色谱手性柱,在正己烷/异丙醇的流动相体系下,考察了该手性柱对一系列手性化合物的拆分性能.实验结果表明,该手性柱对3种苯系物的位置异构体和11种外消旋化合物表现出较好的拆分性能.在正相色谱条件下,该手性柱表现出良好的重现性和稳定性.  相似文献

Diastereomers of (RS)‐propranolol were synthesized using (S)‐levofloxacin‐based new chiral derivatizing reagents (CDRs). Levofloxacin was chosen as the pure (S)‐enantiomer for its high molar absorptivity (εo ~ 24000) and availability at a low price. Its ‐COOH group had N‐hydroxysuccinimide and N‐hydroxybenzotriazole, which acted as good leaving groups during nucleophilic substitution by the amino group of the racemic (RS)‐propranolol; the CDRs were characterized by UV, IR, 1H‐NMR, high resolution mass spectrometry (HRMS) and carbon, hydrogen, nitrogen, and sulphur fundamental elemental components analyser (CHNS). Diastereomers were separated quantitatively using open column chromatography; absolute configuration of the diastereomers was established and the reagent moiety was detagged under microwave‐assisted acidic conditions. (S)‐ and (R)‐propranolol as pure enantiomers and (S)‐levofloxacin were separated, isolated and characterized. Optimized lowest‐energy structures of the diastereomers were developed using Gaussian 09 Rev. A.02 program and hybrid density functional B3LYP with 6‐31G* basis set (based on density functional theory) for explanation of elution order and configuration. In addition, RP HPLC conditions for separation of diastereomers were optimized with respect to pH, concentration of buffer, flow rate of mobile phase and nature of organic modifier. HPLC separation method was validated as per International Conference on Harmonization guidelines. With the systematic application of various analytical techniques, absolute configuration of the diastereomers (and the native enantiomers) of (RS)‐propranolol was established. Abstract

Cellulose and cellulose derivatives are biopolymers that are often used as stationary phases for the separation of enantiomers. Describing the mechanism of such separations is a difficult task due to the complexity of these phases. In the present study, direct enantiomeric resolution of ethofumesate has been achieved, using hexane as the mobile phase with various alcoholic modifiers on cellulose tri(3,5‐dimethylphenylcarbamate) chiral stationary phase (CDMPC CSP). The influence of the mobile phase composition and the column temperature on the chiral separation was studied. It was found that at a constant temperature and within a certain range of alcohol modifier concentration, the conformation of the polymeric phase, and the selective adsorption sites were not affected by alcohol modifier concentration. The type and the concentration of the alcoholic modifiers influenced the retention factor and the separation factor. Ethofumesate gained the best enantioseparation using sec‐butanol as alcoholic modifier at 25°C with α‐value 1.70. And the separation factor decreased with the increase of the column temperature. The van't Hoff plots were linear (R 2>0.96) for ethofumesate from 25°C to 50°C. That showed the enantioselective interactions do not change over the temperature range studied. Furthermore the values of ΔH° and ΔS° were both negative, which indicated an enthalpy‐driven separation. And the possible chiral recognition mechanism of the analyte and CDMPC was discussed. It was found that hydrogen bonding plays an important role on enantioseparation of CDMPC CSP. The inclusion and fitness of solute shape in the chiral cavity significantly contributed to the enantioseparation of solute.  相似文献

High-performance liquid chromatographic methods were developed for separation of the enantiomers of mirtazapine and its four process-related substances. The direct separations were achieved on chiral stationary phases containing amylose tris(3,5-dimethylphenylcarbamate) (Chiralpak AD-H), cellulose tris(3,5-dimethylphenylcarbamate) (Chiralcel OD-H) and cellulose tris(4-methylbenzoate) (Chiralcel OJ-H ). The experimental data were utilized to discuss the effects of the mobile phase composition, the nature of the alcoholic modifier and the specific structural features of the analytes on retention and separation. The elution sequence was determined under the optimized separation conditions.  相似文献

Novel HPLC methods were developed for the analytical and semipreparative resolution of new antianginal drug ranolazine enantiomers. Good baseline enantioseparation was achieved using cellulose tris (3,5-dimethylphenylcarbamate) (CDMPC) chiral stationary phases (CSPs) under both normal-phase and polar organic modes. The validation of the analytical methods including linearity, LODs, recovery, and precision, and the semipreparative resolution of ranolazine racemate were carried out using methanol as mobile phase without any basic and acidic additives under polar organic mode, using CDMPC CSPs. At analytical scale, the elution times of both enantiomers were less than 7.5 min at 20 degrees C and 1.0 mL/min, with the separation factor (a) 1.88 and the resolution factor (R(s)) 2.95. At semipreparative scale, about 14.3 mg/h enantiomers could be isolated and elution times of both enantiomers were less than 13 min at 2.0 mL/min. To increase the throughput, the technique of overlapping injections was used. The first eluted enantiomer was isolated with a purity of 99.6% enantiomer excess (e.e.) and > 99.0% yield. The second enantiomer was isolated with a purity of 98.8% e.e. and > 99.0% yield. In addition, optical rotation and circular dichroism spectroscopy of both ranolazine enantiomers isolated were also investigated.  相似文献

A novel cyclodextrin derivative: mono(6A-N-allylamino-6A-deoxy)perphenylcarbamoylated β-cyclodextrin was synthesized. Hydrosilylation with (EtO)3SiH and then reaction of the reactive siloxane with pristine silica gel afforded a facile entry into a durable, structurally well-defined chiral stationary phase capable of enantioseparation of a variety of racemic drugs.  相似文献

在4种自制的涂敷型纤维素衍生物手性柱,即纤维素三苯甲酸酯(CTB)、纤维素三(4-甲基苯甲酸酯)(CTMB)、纤维素三苯基氨基甲酸酯(CTPC)和纤维素三(3,5-二甲基苯基氨基甲酸酯)(CDMPC)与小分子的Prikle型(S,S)-Whelk—O1手性柱上对取代环己酮及取代环己醇进行了对映体分离。研究了溶质的立体结构因素对手性分离的影响,并初步探讨、比较了溶质在这两种手性柱上的手性识别的机理。结果发现,在(S,S)-Whelk—O1柱上溶质与固定相之间的吸引作用是手性识别的主要原因,而对于纤维素衍生物手性柱,溶质的空间结构在手性空腔中的空间适应性可能是手性识别的关键。  相似文献
