Inductively coupled plasma atomic emission spectrometry (ICP-AES) is a powerful, sensitive analytical technique with numerous applications in chemical characterization including that of ancient pottery, mainly due to its multi-element character, and the relatively short time required for the analysis. A critical step in characterization studies of ancient pottery is the selection of a suitable decomposition procedure for the ceramic matrix. The current work presents the results of a comparative study of six decomposition procedures applied on a standard ceramic potsherd reference material, SARM 69. The investigated decomposition procedures included three microwave-assisted decomposition procedures, one wet decomposition (WD) procedure by conventional heating, one combined microwave-assisted and conventional heating WD procedure, and one fusion procedure. Chemical analysis was carried out by ICP-AES. Five major (Si, Al, Fe, Ca, Mg), three minor (Mn, Ba, Ti) and two trace (Cu, Co) elements were determined and compared with their certified values. Quantitation was performed at two different spectral lines for each element and multi-element matrix-matched calibration standards were used. The recovery values for the six decomposition procedures ranged between 75 and 110% with a few notable exceptions. Data were processed statistically in order to evaluate the investigated decomposition procedures in terms of recovery, accuracy and precision, and eventually select the most appropriate one for ancient pottery analysis.  相似文献

In ethanol production from lignocellulose by enzymatic hydrolysis and fermentation, it is desirable to minimize addition of fresh-water and waste-water streams, which leads to an accumulation of substances in the process. This study shows that the amount of fresh water used and the amount of waste water thereby produced in the production of fuel ethanol from softwood, can be reduced to a large extent by recycling of either the stillage stream or part of the liquid stream from the fermenter. A reduction in fresh-water demand of more than 50%, from 3 kg/kg dry raw material to 1.5 kg/kg dry raw material was obtained without any negative effects on either hydrolysis or fermentation. A further decrease in the amount of fresh water, to one-fourth of what was used without recycling of process streams, resulted in a considerable decrease in the ethanol productivity and a slight decrease in the ethanol yield  相似文献

Reproducible measurements, reliable results and 1st order kinetics for the whole reaction are obtained during the thermal decomposition of dioxetanes only, if inclusions of impurities in commercial sample pans are blocked by additional thick (magnitude of 100 ?m) and close Al2O3 protective layers. As a rule, nearly the same activation parameters are then found both for the decomposition of solvent-free dioxetanes and diluted solutions in several solvents. Mixtures of different dioxetanes in the same solvent contribute independently to the overall heat flow rate.

A batch reactor was employed to steam explode corn fiber at various degrees of severity to evaluate the potential of using this feedstock as part of an enzymatically mediated cellulose-to-ethanol process. Severity was controlled by altering temperature (150–230°C), residence time (1–9 min), and SO2 concentration (0–6% [w/w] dry matter). The effects of varying the different parameters were assessed by response surface modeling. The results indicated that maximum sugar yields (hemicellulose-derived water soluble, and cellulose-derived following enzymatic hydrolysis) were recovered from corn fiber pretreated at 190°C for 5 minutes after exposure to 3% SO2. Sequential SO2-catalyzed steam explosion and enzymatic hydrolysis resulted in a conversion efficiency of 81% of the combined original hemicellulose and cellulose in the corn fiber to monomeric sugars. An additional posthydrolysis step performed on water soluble hemicellulose stream increased the concentration of sugars available for fermentation by 10%, resulting in the high conversion efficiency of 91%. Saccharomyces cerevisiae was able to ferment the resultant corn fiber hydrolysates, perhydrolysate, and liquid fraction from the posthydrolysis steps to 89, 94, and 85% of theoretical ethanol conversion, respectively. It was apparent that all of the parameters investigated during the steam explosion pretreatment had a significant effect on sugar recovery, inhibitory formation, enzymatic conversion efficiency, and fermentation capacity of the yeast.  相似文献

The following paper presents a fast, selective, sensitive, and automated procedure for the residue analysis of the pyrethroid insecticide acrinathrin in various fruit samples by on-line combination of Liquid Chromatography (LC) with capillary Gas Chromatography (GC) and an Electron-Capture Detector (ECD). The loop-type interface and Fully Concurrent Solvent Evaporation (FCSE) were chosen due to their ease of use and low volatility of acrinathrin. Commercially available LC diol columns with small internal diameters (2.1 mm) provided the selectivity required for the separation of the solute from matrix components and permitted the transfer of fraction volumes up to 500 μl into the retention gap, located inside the gas chromatograph. Optimization of the operating conditions with standard solutions showed the influence of the GC initial transfer temperature on the response of acrinathrin and the quantitative nature of the transfer was established by liquid scintillation counting. The linearity and precision were studied with the aid of calibration plots, whereas quantitative recoveries of the analyte (at least 95 %) were determined in spiked fruits. Compared to the corresponding off-line technique, the on-line approach allows us to reach lower detection limits (ppb level when 20 μl LC injections are performed) with excellent repeatability between 0.6 and 6.6 %. Moreover, the integrated analytical set-up is robust, less time-consuming and subsequently brings a marked improvement in the sample preparation step of complex biological matrices. Finally, the versatility of the system renders it highly suitable for the determination of other similar pyrethroids in order to develop a multiresidue method.  相似文献

基于固相萃取原理和微电子机械系统(Micro-Electro-Mechanical System, MEMS)技术研制了一种多孔氧化硅微流控样品预处理芯片, 并利用具有大比表面积的多孔氧化硅作为提取DNA的固相载体, 从而大大提高了DNA的提取产率. 分析了影响DNA提取产率的因素, 改进了芯片制备工艺和DNA提取实验方案, 成功地提取了小鼠外周血DNA, 提取产率为24 ng/(μL全血), 达到商用试剂盒水平. 同时以该DNA作为PCR扩增模板, 扩增效果良好.  相似文献
