Type 2 diabetes mellitus (T2DM) and its attendant complications, such as diabetic nephropathy (DN), impose a significant societal and economic burden. The investigation of discovering potential biomarkers for T2DM and DN will facilitate the prediction and prevention of diabetes. Phospholipids (PLs) and their metabolisms are closely allied to nosogenesis and aggravation of T2DM and DN. The aim of this study is to characterize the human plasma phospholipids in T2DM and DN to identify potential biomarkers of T2DM and DN. Normal phase liquid chromatography coupled with time of flight mass spectrometry (NPLC-TOF/MS) was applied to the plasma phospholipids metabolic profiling of T2DM and DN. The plasma samples from control (n = 30), T2DM subjects (n = 30), and DN subjects (n = 52) were collected and analyzed. The significant difference in metabolic profiling was observed between healthy control group and DM group as well as between control group and DN group by the help of partial least squares discriminant analysis (PLS-DA). PLS-DA and one-way analysis of variance (ANOVA) were successfully used to screen out potential biomarkers from complex mass spectrometry data. The identification of molecular components of potential biomarkers was performed on Ion trap-MS/MS. An external standard method was applied to quantitative analysis of potential biomarkers. As a result, 18 compounds in 7 PL classes with significant regulation in patients compared with healthy controls were regarded as potential biomarkers for T2DM or DN. Among them, 3 DM-specific biomarkers, 8 DN-specific biomarkers and 7 common biomarkers to DM and DN were identified. Ultimately, 2 novel biomarkers, i.e., PI C18:0/22:6 and SM dC18:0/20:2, can be used to discriminate healthy individuals, T2DM cases and DN cases from each other group.  相似文献

Decreased oxygen saturation in asymmetrically prominent cortical veins (APCV) seen in ischemic stroke has been hypothesized to correlate with an increase of de-oxygenated hemoglobin. Our goal is to quantify magnetic susceptibility to define APCV by establishing a cutoff above which the deoxyhemoglobin levels are considered abnormal. A retrospective study was conducted on 26 patients with acute ischemic stroke in one cerebral hemisphere that exhibited APCV with 30 age- and sex-matched healthy controls. Quantitative susceptibility mapping (QSM) was used to calculate the magnetic susceptibility of the cortical veins. A paired t-test was used to compare the susceptibility of the cortical veins in the left and right hemispheres for healthy controls as well as in the contralateral hemisphere for stroke patients with APCV. The change in oxygen saturation in the APCV relative to the contralateral side was calculated after thresholding the susceptibility using the mean plus two standard deviations of the contralateral side for each individual. The thresholded susceptibility value of the APCVs in the stroke hemisphere was 254 ± 48 ppb which was significantly higher (p < 0.05) than that in the contralateral hemisphere (123 ± 12 ppb) and in healthy controls (125 ± 8 ppb). There was a decrease of oxygen saturation in the APCV ranging from 16% to 44% relative to the veins of the contralateral hemisphere. In conclusion, APCV seen in SWI correspond to reduced levels of oxygen saturation and these abnormal veins can be identified using a susceptibility threshold on the QSM data.  相似文献

We present a scalar triplet extension of the standard model to unify the origin of inflation with neutrino mass, asymmetric dark matter and leptogenesis. In presence of non-minimal couplings to gravity the scalar triplet, mixed with the standard model Higgs, plays the role of inflaton in the early Universe, while its decay to SM Higgs, lepton and dark matter simultaneously generate an asymmetry in the visible and dark matter sectors. On the other hand, in the low energy effective theory the induced vacuum expectation value of the triplet gives sub-eV Majorana masses to active neutrinos. We investigate the model parameter space leading to successful inflation as well as the observed dark matter to baryon abundance. Assuming the standard model like Higgs mass to be at 125–126 GeV, we found that the mass scale of the scalar triplet to be ?O(109) GeV?O(109) GeV and its trilinear coupling to doublet Higgs is ?0.09 so that it not only evades the possibility of having a metastable vacuum in the standard model, but also lead to a rich phenomenological consequences as stated above. Moreover, we found that the scalar triplet inflation strongly constrains the quartic couplings, while allowing for a wide range of Yukawa couplings which generate the CP asymmetries in the visible and dark matter sectors.  相似文献
