The study on the mechanism and kinetics of mRNA degradation provides a new vision for chemical intervention on protein expression. The AU enrichment element (ARE) in mRNA 3′-UTR can be recognized and bound by the ARE binding protein (AU-rich Element factor (AUF1) to recruit RNase for degradation. In the present study, we proposed a novel strategy for expression regulation that interferes with the AUF1-RNA binding. A small-molecule compound, JNJ-7706621, was found to bind AUF1 protein and inhibit mRNA degradation by screening the commercial compound library. We discovered that JNJ-7706621 could inhibit the expression of AUF1 targeted gene IL8, an essential pro-inflammatory factor, by interfering with the mRNA homeostatic state. These studies provide innovative drug design strategies to regulate mRNA homeostasis.  相似文献

 The design of new heterogeneous photooxygenation systems able to employ visible light, oxygen, mild temperatures, and solvent with a low environmental impact has been investigated. In particular, the heterogenization of decatungstate (W10O4-32), a polyoxometalate with photocatalytic activity in oxidation reactions, has been carried out in polymeric membranes of polyvinylidenefluoride. The polymeric catalytic membranes prepared by phase inversion technique have been successfully applied in the aerobic mineralization of phenol in water, which was used as an example of organic pollutant. In order to evaluate the effect of the polymeric environment on the overall catalyst behavior, we have also heterogenized the decatungstate (opportunely functionalized) in perfluorinated membrane made of Hyflon. The photocatalytic composite membranes are characterized by different and tuneable properties depending on the nature of the polymeric micro-environment, in which the catalyst is confined. Moreover, the selective separation function of the membrane results in enhanced performance in comparison with homogeneous reactions.  相似文献

采用高效液相色谱-二极管阵列(HPLC-DAD)检测方法定量分析仿生杀菌剂邻烯丙基苯酚在小麦纹枯病菌中的降解情况。检测条件为:流动相为乙腈/水(0.1%醋酸),HYPERSIL BDS C18(250 mm×4.6 mmi.d.,5μm)色谱柱,检测波长280 nm。邻烯丙基苯酚的线性回归方程为y=5.019 8.0266x(r=0.9999),在2~500 mg/L浓度范围内线性关系良好。检出限和定量下限分别为0.15和2.00 mg/L;平均回收率为72.63%。所建立的分析方法灵敏、准确,并且具有较好的稳定性和重现性,可以用于邻烯丙基苯酚在小麦纹枯病菌中消解动态的定性定量分析。检测0~144 h内邻烯丙基苯酚在小麦纹枯病菌培养液中的降解情况,0~8h间邻烯丙基苯酚在培养液中迅速降解,至8 h时药物的降解率达到47.13%。此后,邻烯丙基苯酚呈平稳的递减趋势,在144 h药物已消解71.02%。此过程中药物对病菌菌丝生长的抑制作用并未因其降解而降低,而是呈现较平稳的抑菌势态。  相似文献

In this study, we aimed to isolate bacteria capable of degrading the polysaccharide ulvan from the green algae Ulva sp. (Chlorophyta, Ulvales, Ulvaceae) in marine environments. We isolated 13 ulvan-degrading bacteria and observed high diversity at the genus level. Further, the genera Paraglaciecola, Vibrio, Echinicola, and Algibacter, which can degrade ulvan, were successfully isolated for the first time from marine environments. Among the 13 isolates, only one isolate (Echinicola sp.) showed the ability not only to produce externally expressed ulvan lyase, but also to be periplasmic or on the cell surface. From the results of the full-genome analysis, lyase was presumed to be a member of the PL25 (BNR4) family of ulvan lyases, and the bacterium also contained the sequence for glycoside hydrolase (GH43, GH78 and GH88), which is characteristic of other ulvan-degrading bacteria. Notably, this bacterium has a unique ulvan lyase gene not previously reported.  相似文献

将固化后的酚醛树脂在不同温度下进行热处理,对固化后的样品进行热重分析,对热处理后的试样进行傅立叶红外光谱分析.实验结果表明,酚醛树脂的热降解与亚甲基的取代位有关.酚醛树脂中的亚甲基分两个阶段进行热解降,350~450℃的温度区间主要是部分邻-邻(o-o′)位亚甲基和邻-对(o-p)位亚甲基的分解,400~620℃温度区间为对-对(p-p′)位亚甲基的分解,p-p′位亚甲基比o-o′位亚甲基的起始热分解温度高50℃左右.  相似文献

Step-stress experiments are performed in this paper to investigate the degradation mechanism of an AlGaN/GaN high electron mobility transistor(HEMT).It is found that the stress current shows a recoverable decrease during each voltage step and there is a critical voltage beyond which the stress current starts to increase sharply in our experiments.We postulate that defects may be randomly induced within the AlGaN barrier by the high electric field during each voltage step.But once the critical voltage is reached,the trap concentration will increase sharply due to the inverse piezoelectric effect.A leakage path may be introduced by excessive defect,and this may result in the permanent degradation of the AlGaN/GaN HEMT.  相似文献

The effect of UV irradiation on the morphology and structure of syndiotactic polypropylene (sPP) is investigated both for pure films and those filled with multiwall carbon nanotubes (MWCNTs). UV treatment causes a structural reorganization of pure sPP films. It seems that the incorporation of MWCNTs has a strong stabilizing effect, with nanotubes acting as a limiter of the UV‐induced chain breakage, especially when a good dispersion of carbon nanotubes in the polymer is achieved. The incorporation of MWCNTs introduces a high concentration of defects in the crystal structure of sPP, which limits the UV‐induced growth of crystallites, and prevents the development and propagation of cracks caused by UV irradiation. © 2012 Wiley Periodicals, Inc. A new class of polyurethanes has been designed, containing tertiary carbamate groups in the main chain of the polymer, which enable the resulting polymer to degrade completely under acid and thermal treatment. The decomposition temperatures of the polymers were determined by measuring the evolution of carbon dioxide and other decomposition products using TGA‐MS. Until decomposition of the polymer, no glass transition was found. The polymers exhibit excellent solubility in common organic solvents like chloroform and tetrahydrofuran, making them to suitable materials for film formation. From the obtained polymers, nanoparticles were synthesized by the solvent evaporation method combined with the miniemulsion technique. The resulting nanoparticles can be used as intelligent fillers in films and sensors, since they degrade at temperatures of above 180 °C, which can be detected by a color change reaction with ninhydrin. Polymeric nanocapsules were prepared by an interfacial polyaddition reaction from 2,4‐toluene diisocyanate and tertiary diols performed at the droplet's interface in inverse (water‐in‐oil) miniemulsions. These nanocapsules with an encapsulated photoacid generator can act as a release system, whereby an acidic release through irradiation with ultraviolet light can be triggered. © 2011 Wiley Periodicals, Inc.
