采用快速高分离度液相色谱-质谱技术(RRLC-MS)检测经格列美脲治疗的2型糖尿病大鼠尿液中代谢物的变化, 对糖尿病组、 给药组和健康对照组大鼠的尿液代谢物谱进行了分析. 采用主成分分析(PCA)对3组大鼠进行分类并寻找潜在生物标记物. 结果表明, 3组大鼠的尿液代谢物谱得到了很好的区分, 发现并鉴定了2个潜在生物标记物, 分别为4-脱氧三羟基丁酸和4-胍基丁酸. 格列美脲对2型糖尿病大鼠的药物作用可能体现为对氨基酸代谢的调节作用.  相似文献

Levamisole is the most common adulterant found in cocaine samples and its electrochemical determination in cocaine seized samples is a challenge due to peak overlapping with cocaine. Herein, we propose a deconvolution procedure for levamisole determination in seized cocaine samples using screen‐printed carbon electrodes (SPE). Square‐wave voltammetry in 0.04 mol L?1 Britton Robinson buffered solution (pH 8.0) was selected in combination with optimized SWV parameters (f=8 s?1, a=10 mV and ΔEs=1 mV) to result in the best peak resolution to apply the deconvolution procedure. Deconvoluted responses of levamisole in the presence of cocaine were similar to untreated signals of standard levamisole solutions in absence of cocaine. A linear response was obtained in the range of 20–100 μmol L?1 (r=0.995). The results obtained for the analysis of a seized cocaine sample was statistically similar to that obtained by gas chromatography. Other adulterants found in cocaine street samples (paracetamol, glucose, phenacetin, caffeine, boric acid and lidocaine) did not affect the treated of voltammetric responses of levamisole. A pharmaceutical sample containing levamisole was also analyzed on SPEs and a recovery of 93±2 % was obtained (no deconvolution required for this sample), showing great applicability of SPEs for forensic and pharmaceutical analyses.  相似文献

Endothelialization is an effective approach to prevent thrombus formation and enhance vascular graft survival. Surface modification of biomolecules has been proved to be effective in regulating endothelial cell behaviors. In this study, several peptides including YIGSR, RGD, and REDV sequences are covalently immobilized on the surface of electrospun silk fibroin scaffolds and the effects of combined application of these peptides on cell behaviors are studied. The results show that, compared with the scaffolds modified with single peptides, the scaffolds modified with dual peptides (YIGSR+RGD) could significantly enhance the proliferation of human umbilical vein endothelial cells (HUVECs). However, the combination of REDV+RGD or YIGSR+REDV does not promote the adhesion or proliferation of HUVECs. Notably, YIGSR‐modified scaffolds improved HUVEC migration significantly in comparison to REDV‐ or RGD‐modified groups. Moreover, its combination with either of these two peptides also presents excellent effect on cell migration. Thus, all the data suggest that the combined application of peptides might be a promising method to enhance the endothelialization of small‐diameter vascular grafts.  相似文献

Artificial small‐caliber vascular grafts are still limited in clinical application because of thrombosis, restenosis, and occlusion. Herein, a small‐caliber vascular graft (diameter 2 mm) is fabricated from poly(ε‐caprolactone)‐b‐poly(isobutyl‐morpholine‐2,5‐dione) (PCL‐PIBMD) and silk fibroin (SF) by electrospinning technology and then biofunctionalized with low‐fouling poly(ethylene glycol) (PEG) and two cell‐adhesive peptide sequences (CREDVW and CAGW) with the purpose of enhancing antithrombogenic activity and endothelialization. The successful grafting of PEG and peptide sequences is confirmed by X‐ray photoelectron spectroscopy. The suitable surface wettability of the modified vascular graft is testified by water contact angle analysis. The surface hemocompatibility is verified by platelet adhesion assays and protein adsorption assays, and the results demonstrate that both platelet adhesion and protein adsorption on the biofunctionalized surface are significantly reduced. In vitro studies demonstrate that the biofunctionalized surface with suitable hydrophilicity and cell‐adhesive peptides can selectively promote the adhesion, spreading, and proliferation of human umbilical vein endothelial cells. More importantly, compared with control groups, this biofunctionalized small‐caliber vascular graft shows high long‐term patency and endothelialization after 10 weeks of implantation. The biofunctionalization with PEG and two cell‐adhesive peptide sequences is an effective method to improve the endothelialization and long‐term performance of synthetic vascular grafts.  相似文献

The task of chromatographically separating a complex mixture containing constituents with a wide range of molecular weight has been referred to in the past as the “general elution problem”. A different operational mode for conducting gas chromatography, thermal gradient programmed gas chromatography, presently in the developmental stage, has been investigated and is proposed as a means whereby such separations might be accomplished rapidly. The inherent versatility of the technique for conducting rapid analyses (50 to 300 s duration) of complicated samples containing constituents with a wide range of volatility is considered in terms of the possibility of achieving the maximum amount of analytical information per unit time.  相似文献

This paper constitutes an experimental and numerical study, using uncertainty analysis of the most important parameters, to evaluate the mechanism for the combustion of CO + H2 mixtures at high pressures in the range 15-50 bar and temperatures from 950 to 1100 K. Experiments were performed in a rapid compression machine. Autoignition delays were measured for stoichiometric compositions of CO + H2 containing between 0 and 80% CO in the total fuel mixture. The experimental results showed an unequivocal monotonic increase as the proportion of CO in the mixture was raised. Comparisons were made also with the measured ignition delays in mixtures of H2 with increasing dilution by N2, corresponding to the proportions of CO present. These times also increased monotonically, albeit with a greater sensitivity to the extent of dilution than those measured in the CO + H2 mixtures. By contrast, numerical simulations for the same mixtures, based on a kinetic model derived by Davis et al. displayed a qualitative discrepancy as there was virtually no sensitivity of the ignition delay to the changing ratio of CO + H2, certainly up to 80% replacement. No exceptions to this trend were found, despite tests being made using seven other kinetic models for CO + H2 combustion. Global uncertainty analyses were then applied to the Davis et al. model in order to trace the origins of this discrepancy. The analyses took into account the uncertainties in all rate parameters in the model, which is a pre-requisite for evaluation against ignition delay data. It is shown that the reaction rate constant recommended by Baulch et al. for the HO2 + CO reaction, at T ∼ 1000 K, could be up to a factor of 10 too high and that lowering this rate corrected the qualitative anomaly between experiment and numerical prediction.  相似文献

研究四君子汤在2型糖尿病大鼠体内的药代动力学。 将四君子汤水提物以18 g/kg给正常大鼠和2型糖尿病大鼠灌胃,分别在不同时间点取尿液和粪便,检测其成分及代谢产物的含量;比较正常大鼠和2型糖尿病大鼠的药代动力学参数。 采用快速高分辨液相色谱-质谱联用技术对人参皂苷Rc、甘草甜素及其脱糖代谢物进行表征。 结果表明,四君子汤中人参皂苷Rc和甘草甜素在2型糖尿病大鼠体内的累计排泄率均高于正常大鼠,大部分人参皂苷通过尿液排出体外、粪便代谢为稀有人参皂苷;大部分甘草甜素在尿液和粪便中转化为甘草次酸,2型糖尿病大鼠与正常大鼠人参皂苷Rc和甘草甜素的药代动力学参数存在明显差异。 初步总结了四君子汤在体内的药代动力学机制和代谢途径。本分析方法具有高度自动化,灵敏性和准确性,这将有助于四君子汤的临床研究的不断发展。  相似文献

This review considers the ethical and technical problems currently associated with employing mouse bioassays for marine-toxin analysis and the challenges and the difficulties that alternative methods must overcome before being deemed applicable for implementation into a regulatory monitoring regime. We discuss proposed alternative methods, classified as functional, immunological and analytical, for well-established European toxins as well as emerging toxins in European waters, highlighting their advantages and disadvantages. We also consider emerging tools and technologies for future toxin analysis.Even though regulatory bodies have recently recommended analytical methods for a number of toxins, there is still scope for functional and immunological methods in rapid screening and detecting emerging toxins. Future developments foreseen in the analysis of marine biotoxins are multiplex-based analysis, miniaturization and portability for on-site testing. However, the longstanding lack of reference materials and standards continues to pose a severe limitation on progress in development, validation and therefore implementation of any alternative method based on the criteria stipulated by European Union legislation.  相似文献

For bottom-up MS, the digestion step is critical and is typically performed with trypsin. Solvent-assisted digestion in 80% acetonitrile has previously been shown to improve protein sequence coverage at shorter digestion times. This has been attributed to enhanced enzyme digestion efficiency in this solvent. However, our results demonstrate that tryptic digestion in 80% acetonitrile is less efficient than that of conventional (aqueous) digestion. This is a consequence of decreased enzyme activity beyond ∼40% acetonitrile, increased enzyme autolysis and lower protein solubility in 80% acetonitrile. We observe multiple missed cleavages and reduced concentration of fully cleaved digestion products. Nonetheless we confirm, through room temperature solvent-assisted digestion, a consistent improvement in protein sequence coverage when analyzed by mass spectrometry. These results are explained through the increased number of unique digestion products available for detection. Thus, while solvent-assisted digestion has clear merits for proteome analysis, one should be aware of the inefficiency of protein digestion though this protocol, particularly with absolute protein quantitation experiments.  相似文献
