Abstract This report describes a radioimmunoassay method for the measurement of unconjugated corticosterone (B) in plasma. Utilizing an anti-Cortisol antiserum cross reacting with B combined with a one step celite microcolumn chromatographic system, we have measured this latter steroid accurately in small aliquots of plasma from both male and female subjects. The plasma levels of B obtained by this method agreed well with levels observed in adult men and women, using other techniques. During a menstrual cycle studied, wide fluctuations were observed with levels ranging from 1, 3 to 14, 4 ng/ml. Adrenal suppression decreased the levels of B below 1 ng/ml. The mineralocorticoid B is predominantly ACTH -dependent. 相似文献

Conclusion The outlined technique of combining automatic HPLC and RIA has been proved to be a practicable and specific method for the estimation of multiple adrenal steroids from different body fluids. It is equivalently suited for the simultaneous and specific assessment of any other steroid groups.

Abstract A relatively simple and rapid radioimmunoassay (RIA) for the measurement of aldosterone in serum and urine has been developed. The method involves extraction of 1 ml of serum or urine (after acid hydrolysis) with dichloromethane, followed by partition chromatography on celite microcolumns prior to RIA. Dextran coated charcoal is used for separation of free from antibody-bound aldosterone. The method is very sensitive, blanks are negligible and recovery is approximately The%. 80 coefficient of variation is 4. 3% (within assay) and 10. 4% (between assay). When known amounts of aldosterone were added to serum and urine pools containing low endogenous levels of this steroid recovery was quantitative. Aldosterone concentrations measured under various physiological conditions were in agreement with published data. Up to 150 samples can be assayed by 1 technician during 5 working days. 相似文献

The plasma levels of 17-5δ-P obtained by this method agreed well with levels observed in adult men and women, using two other techniques.
