Abstract 4-[(5,6,7,8-Tetrahydro-5,5,8,8-tetramethyl-2-naphthyl)-carbamoyl] benzoic acid (Am-80) is a novel retinobenzoic acid derivative possessing retinoid activity. Plasma concentration of this drug in clinical use is very low, less than 1 ng/ml, and could not be measured with direct immunoassays. A combination of high-performance liquid chromatography (HPLC) and radioimmunoassay (RIA) was developed for this drug in human plasma. Am-80 in 0.5 ml of human plasma was extracted by solid-phase extraction, and the extract was purified by reversed-phase HPLC. The immunoreactivity in the fraction of the eluate was measured by competitive RIA. The within- and between-assay variances were 4.1 to 15.5% and 4.1 to 7.0%, respectively. The limit of quantification was 0.04 ng/ml in human plasma, which was much lower than that of direct RIA, 0.6 ng/ml, previously reported. This assay system could be used to observe the pharmacokinetics of Am-80 in human even after dermal application at very low dose. 相似文献

Abstract A radioimwunoassay for the quantitation of cytochrome P-450 (cyt P-450) from the yeast Candida maltosa in the microsomab fraction was established using a purified secondary antibody adsorbed to cellulose nitrate discs, a primary rabbit anti (cyt P-450) serum and [125I] labeled cyt P-450. The present assay is highly sensitive, specific and simple, It allows the detection of as little as 5 f moles cyt P-450 in a volume of 40 μ1. Equal immunological reactivities were found for the highly purified, solubilized microsomal, denatured and the apo-form of the yeast cyt P-450. 相似文献

Abstract A radioimmunoassay method for the direct measurement of cortisol in plasma after precipitation of the proteins with ethanol is described. Utilizing a specific antiserum against cortisol, with minimal or no cross reaction with other steroids, cortisol was meas-red accurately in a volume of 0.001 ml of plasma. The range of levels that could be measured per ml of plasma varied from 10 to 500 ng. The mean value of plasma cortisol at 8 a. m. obtained in twelve subjects by this method was about half the mean levels reported by another competitive protein binding assay. 相似文献
