Poly(ethylene glycol) (PEG) is frequently used to coat various medical nanoparticles (NPs). As PEG is known to minimize NP interactions with biological specimens, the question remains whether PEGylated NPs are intrinsically less toxic or whether this is caused by reduced NP uptake. In the present work, the effect of gold NP PEGylation on uptake by three cell types is compared and evaluated the effect on cell viability, oxidative stress, cell morphology, and functionality using a multiparametric methodology. The data reveal that PEGylation affects cellular NP uptake in a cell‐type‐dependent manner and influences toxicity by different mechanisms. At similar intracellular NP numbers, PEGylated NPs are found to yield higher levels of cell death, mostly by induction of oxidative stress. These findings reveal that PEGylation significantly reduces NP uptake, but that at similar functional (= cell‐associated) NP levels, non‐PEGylated NPs are better tolerated by the cells.  相似文献

It is very likely that the main driving force of enzyme evolution is the requirement to improve catalytic and regulatory efficiency which results from the intrinsic performance as well as from the spatial and functional organization of enzymes in living cells.Kinetic co-operativity may occur in simple monomeric proteins if they display “slow” conformational transitions, at the cost of catalytic efficiency. Oligomeric enzymes on the other hand can be both efficient and co-operative. We speculate that the main reason for the emergence of co-operative oligomeric enzymes is the need for catalysts that are both cooperative and efficient. As it is not useful for an enzyme to respond to a change of substrate concentration in a complex kinetic way, the emergence of symmetry has its probable origin in a requirement for “functional simplicity”.In a living cell, enzyme are associated with other macromolecules and membranes. The fine tuning of their activity may also be reached through mutations of the microenvironment. Our hypothesis is that these mutations are related to the vectorial transport of molecules, to achieve the hysteresis loops of enzyme reactions generated by the coupling of reaction and diffusion, through the co-operativity brought about by electric interactions between a charged substrate and a membrane, and last but not least, through oscillations. As the physical origins of these effects are very simple and do not require complex molecular devices, it is very likely that the functional advantage generated by the spatial and functional organization of enzyme molecules within the cell have appeared in prebiotic catalysis or very early during the primeval stages of biological evolution.We shall began this paper by presenting the nature of the probable earliest catalysts in the RNA world.  相似文献

Existing experimental studies of the thermal denaturation of DNA yield sharp steps in the melting curve suggesting that the melting transition is first order. This transition has been theoretically studied since the early sixties, mostly within an approach in which the microscopic configurations of a DNA molecule consist of an alternating sequence of non-interacting bound segments and denaturated loops. Studies of these models neglect the repulsive, self-avoiding, interaction between different loops and segments and have invariably yielded continuous denaturation transitions. In the present study we take into account in an approximate way the excluded-volume interaction between denaturated loops and the rest of the chain. This is done by exploiting recent results on scaling properties of polymer networks of arbitrary topology. We also ignore the heterogeneity of the polymer. We obtain a first-order melting transition in d = 2 dimensions and above, consistent with the experimental results. We also consider within our approach the unzipping transition, which takes place when the two DNA strands are pulled apart by an external force acting on one end. We find that the under equilibrium condition the unzipping transition is also first order. Although the denaturation and unzipping transitions are thermodynamically first order, they do exhibit critical fluctuations in some of their properties. For instance, the loop size distribution decays algebraically at the transition and the length of the denaturated end segment diverges as the transition is approached. The development of highly potent chiral discrimination methods that solve the problems of the diastereomer method, in which it is impossible to discriminate the diastereomers having chiral centers separated by more than four bonds, is described. On the basis of the results obtained, a new hypothesis, Induced Chiral Fields that the achiral reversed phase can provide chiral fields depending on the structures of the eluents, is proposed to explain the significant results of separation of the diastereomers derived from newly developed chiral and fluorescent labeling reagents and optical isomers by reversed-phase HPLC, which was hitherto impossible.  相似文献

探索了影响氯化亚锡还原法制备碳纳米管和不同粒径纳米碳黑的99mTc标记化合物的多种因素, 在给定实验条件下, 99mTc的标记率能够稳定达到90%以上。 细胞培养液中标记物的放射化学纯度在2.5 h中保持在(86±4)%范围内。 制备的标记化合物满足碳纳米颗粒细胞摄取率测定和细胞毒性机制研究的实验要求。 实验结果提示, 99mTc标记过程是基于还原得到的低价Tc在碳纳米颗粒表面上的物理吸附机理。 The effects of experimental conditions on preparation of 99mTc labeled carbon nanotubes and nanocarbon blacks by SnCl2 were investigated. At given conditions the labeling yields were over 90%. In a culture medium, the radiochemical purity of the labeling compounds kept (86±4)% within 2.5 h. The 99mTc labeled multi walled carbon nanotubes (MWNTs) and nanocarbon blackes (NCBs) obtained in this work met satisfactory experimental demands for study of cellular uptake and toxicity. The experiments showed that the labeling process was based on physical adsorption of low valent technetium resulted from reduction reaction on the surface of the carbon nanomaterials.  相似文献

Five two-photon excitable dipyrrylmetheneboron difluoride labels (dipyrrylmethene-BF2 labels) with fluorescence emission maximum between 530 and 590 nm, and a frequently used rhodamine label, TAMRA, were conjugated to aminomodified oligonucleotides. The performance of the labeled oligonucleotides was studied in a separation-free nucleic acid hybridization assay using ArcDia TPX bioaffinity assay technology. The results show that oligonucleotide conjugates of dipyrrylmethene-BF2 labels provide higher two-photon excited fluorescence yield and better assay sensitivity than corresponding TAMRA conjugate. The effect of conjugation on photophysical properties of the labels and performance of the labeled oligonucleotides in separation-free hybridization assay is discussed.  相似文献

近年来,羟自由基(^.OH)对DNA氧化损伤已受到广泛关注,但是很少研究^.OH对RNA的氧化损伤。其实,RNA与DNA一样,也是核酸的两大组分之一,也有许多重要功能。所以^.OH攻击RNA也会引起重后果,会造成细胞功能衰退甚至细胞死亡等。为此,我们建立了Vit.C-CuSO4-Phen-H2O2-PNA这一产生和测定^.OH氧化损伤RNA的化学发光体系,以便加强^.OH氧化损伤RNA的研究。通过对本体系测定条件的研究,得出了本体系最佳组方是:Vit.C,CuSO4,Phen,H2O2和RNA,浓度分别为350μmol/L,55μmol/L,350μmol/L,0.2mol/L和20μg/mL,体系pH为5.5,体系终体积为1mL。随后,利用本体系检测了槲皮素,咖啡酸,黄芩甙和芦丁抗^.OH氧化损伤RNA的作用,发现这四种抗氧化剂均能有效抑制^.OH氧化损伤RNA的分子机理,结果发现,^.OH清除剂硫脲几乎抑制全部发光,推测是因硫脲清除了引发剂^.OH所致;O^-.2清除剂SOD只能抑制小部分发光;^1O2清除剂叠氮化钠和苯甲酸都能抑制绝大部分发光。这些事实提示,^.OH是RNA氧化损伤的引发剂;O^-.2只是导致RNA氧化损伤的次要因素,^1O2才是导致RNA氧化损伤的最主要因素。  相似文献

Bioprobes based on fluorescent ruby nanoparticles, which are suitable for ultrasensitive imaging, are reported. A stable aqueous/buffer colloid, permitting facile conjugation to proteins, is produced by femtosecond laser ablation of ruby and the nanoparticles (mean size 17 nm) are photostable, with long lifetime (1–4 ms) 694 nm emission. With time‐gating complete (>20 dB) suppression of cell autofluorescence and suppression of exogenous fluorophores is observed. Nanoparticles are imaged in as‐grown cells and those immunolabeled with quantum dots. Immunoassay binding to target biomolecules is also demonstrated.  相似文献
