A rapid, selective, and sensitive liquid chromatography–tandem mass spectrometry method was developed and validated for the simultaneous determination of unbound sunitinib and its active metabolite N‐desethyl sunitinib in plasma. Plasma and post‐dialysis buffer samples were extracted using a liquid–liquid extraction procedure with acetonitrile–n‐butylchloride (1:4, v/v). Chromatographic separation was achieved on a Waters X‐Terra® MS RP18 column with a mobile phase consisting of acetonitrile and water (60:40, v/v) containing formic acid (0.1%, v/v) using an isocratic run, at a flow‐rate of 0.2 mL/min. Analytes were detected by electrospray tandem mass spectrometry in the selective reaction monitoring mode. Linear calibration curves were generated over the ranges 0.1–100 and 0.02–5 ng/mL for sunitinib and 0.2–200 and 0.04–10 ng/mL for N‐desethyl sunitinib in plasma and in phosphate‐buffered solution, respectively. The values for both within‐day and between‐day precision and accuracy were well within the generally accepted criteria for analytical methods. The analytical range was sufficient to determine the unbound and total concentrations of both analytes. The method was applied for measurement unbound concentrations in addition to total concentrations of sunitinib and its metabolite in plasma of a cancer patient receiving 50 mg daily dose. Abstract

The separation of bromocomplexes of platinum group metals by Donnan dialysis is demonstrated with both anion and cation exchange membranes. the inclusion of ethylenediamine (en) in the sample improves the separation of Pd(II) from Pt(IV) with experiments performed with an anion exchange membrane and decreases the amount of metal retained on the membrane phase. With a cation exchange membrane, the addition of a ligand such as en is required for transport. With 5.6 mM en in the sample at pH 10, 74% of Pd(II) is transported across an anion exchange membrane into 0.5 M NH4Br after 6 hours while only 8% of the Pt(IV) is dialyzed. Rhodium(III) and iridium(III) behave like Pt(IV). Using a cation exchange membrane under the same conditions except with a 1 hour dialysis results in a 30-fold preferential preconcentration of Pd(II) relative to Pt(IV), and, based on the amount retained in the membrane, a preconcentration of Ir(III) which exceeds that of Pd(ll) and Pt(IV) by factors of 40 and 20, respectively.  相似文献

以d[TG4T]4、d[G4T4G4]2和c-myc序列为代表,采用竞争平衡透析、紫外光谱和圆二色光谱研究了oxazine750对四股螺旋结构的识别与稳定作用。 结果表明,oxazine750能够更好的结合在平行四股螺旋上,而对反平行四股螺旋结合较少。 热变性实验表明,oxazine750使d[TG4T]4形成的平行结构的四股螺旋的解旋温度提高了11 ℃,对c-myc形成的平行四股螺旋的解旋温度甚至提高了40 ℃。 而对d[G4T4G4]2形成的反平行结构解旋温度降低了近8 ℃。  相似文献

用平衡透析法详细研究了生理pH(7.43)条件下Cd(Ⅱ)与HSA或BSA的结合平衡.通过非线性最小二乘法拟合Bjerrum方程,首次报道了Cd(Ⅱ)-HSA和Cd(Ⅱ)-BSA体系的逐级稳定常数值,其K_1~K_3的数量级均为10~4;Hill系数和自由能偶合定量分析表明Cd(Ⅱ)与HSA或BSA的结合均产生在类似体系中少见的强的正协同效应,且Cd(Ⅱ)与HSA结合产生的正协同效应大于BSA;Scatchard图分析表明,Cd(Ⅱ)在HSA和BSA中均有3个强结合部位.通过Cd(Ⅱ)与Cu(Ⅱ),Zn(Ⅱ)或Ca(Ⅱ)等竞争结合HSA或BSA的结果,进一步讨论了Cd(Ⅱ)在HSA或BSA中强结合部位的可能位置和(或)配体.  相似文献

Using the preirradiation technique a kinetic study of the grafting of the 4-vinyl pyridine (V4P) and an aliphatic ammonium monomer (ALAM) onto the copolymer film of ethylene–tetrafluoroethylene (ETFE) has been performed. The influence of dose, temperature, and concentration of monomer, reticular agent, and inhibitor were investigated. The results are discussed on the basis of the interactions between monomer diffusibility and viscosity of the medium. The characteristics of some membranes were determined. Their applicability to the recovery of acid by dialysis is demonstrated.  相似文献

Detailed studies were carried out on equilibrium dialysis of the binding of Ni2++ ion to human scrum albumin (HSA) and bovine serum albumin (BSA).The successive stability constants were obtained by the Icfisi squares fitting.The eight binding sites found for both Ni(Ⅱ)-HSA and Ni(Ⅱ)-BSA systems can be divided into two different sets; and for both systems,there exist two identical prior binding sites where the bound Ni2+ ions can he con sidered as allosteric effectors,which induce the allosteric effect in accordance with the model proposed by Moeod et al As indicated by allosteric parameters,the ability of R-state to bind Ni2+ ions is ca 100 times as much as that of T state,and the conformation of HSA is markedly tenser than that of BSA.  相似文献

The possibility of selective preconcentration of platinum group metal ions by Donnan dialysis was investigated. The effect of sample matrix (glycine) on self diffusion of the following platinum group metal ions Pt(IV), Pd(II), Rh(III), Ir(III) and Ir(IV) was determined. To separate a sample from the receiver electrolyte (0.5M NH4Cl), anion or cation-exchange membrane were used. Excellent selective preconcentration of Pd(II) in the sample and Ir(III) in the receiver solution was achieved. Experiments performed enable to draw some conclusion on the charge sign of glycinepalladium complexes.  相似文献

Polyethersulfone (PES) hollow fiber membranes for kidney dialysis application were prepared by the dry-jet wet-spinning method. A dual-coagulation bath technology was first time employed for fabricating the kidney dialysis membranes with a tight inner skin and loose outer supporting layer structure. A weak coagulant isopropanol (IPA) was served as the first external coagulation bath, while water as the second bath. Experiments demonstrate their advantages of better controlling both inner and outer skin morphology. The as-spun fibers have a higher mean effective pore size (μp), pure water permeation flux (PWP) and molecular weight cut-off (MWCO) with an increase in N-methyl-2-pyrrolidone (NMP) percentage in bore fluid (i.e., internal coagulant). After being treated in 8000 ppm NaOCl solution for 1 day, fibers show larger pore sizes and porosity in both inner and outer surfaces, and thinner inner and outer layers than their as-spun counterparts. Among them, the bleached fibers spun with 50 wt.% NMP in bore fluid have the MWCO (43 kDa) and PWP (40 × 10−5 L m−2 Pa−1 h−1) suitable for kidney dialysis application. Based on SEM observations and solute rejection performance, the further heat treated fibers in an aqueous solution is found to be an effective way to fine tune membranes morphology and MWCO for kidney dialysis application. The solute rejection performance data of the hollow fiber membranes spun with 55 wt.% NMP in bore fluid after heat treated at 90 °C in water for 2 h were found to be very appropriate for the kidney dialysis application.  相似文献
