建立了高速逆流色谱分离纯化木蝴蝶黄芩素和白杨黄素的方法。两相溶剂系统为石油醚-乙酸乙酯-甲醇-水,固定相为5:5:5:5(V/V)体系的上相,以5:5:5:5(V/V)和5:5:7:3(V/V)体系的下相为流动相进行梯度洗脱。从300mg木蝴蝶粗提物中一步分离纯化得到25.5mg黄芩素和36.6mg白杨黄素。经高效液相色谱分析,纯度分别为99.2%和100%。其化学结构由^1H-NMR和^13C-NMR鉴定。  相似文献

Introduction: Identification of Oroxylum indicum and Scutellaria baicalensis provides an interesting challenge in selection of biomarker compound to be used in routine analysis. Both plants have similar phytochemical profile and are rich sources of flavones and flavone glycosides. The objective of this study was to prepare the chemical fingerprinting of O. indicum bark and S. baicalensis roots using the liquid chromatography and mass spectroscopy in single chromatographic method. Materials and methods: Extracts prepared using various solvent systems (methanol, aqueous methanol, chloroform, hexane, and water) of both plants were analyzed using C18 reverse phase column with solvent system containing acetonitrile and 0.1% formic acid. Major flavonoids were identified based on mass spectra, fragmentation pattern, and UV spectra. Results: In this article, well-resolved high-performance liquid chromatographic (HPLC) separation in both plant extracts was obtained and chemical fingerprints for both plant extracts were established and flavonoids present (baicalin, oroxylin A-7-O-glucuronide, chrysin-7-O-glucuronide, baicalein, chrysin, oroxylin-A, wogonin, skullcap flavone II) were identified as possible biomarkers. Conclusion: Mass spectrometry coupled with HPLC can be a tool for fingerprinting of various natural products used in dietary supplement industry. The fingerprint developed in the article can be used for quality evaluation as well as identifying possible adulteration of extracts of both the plants.  相似文献

The study was aimed to identify cytotoxic leads from Abutilon indicum leaves for treating glioblastoma. The petroleum ether extract, methanol extract (AIM), chloroform and ethyl acetate sub-fractions (AIM-C and AIM-E, respectively) prepared from AIM were tested for cytotoxicity on U87MG human glioblastoma cells using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. These extracts exhibited considerable activity (IC50 values of 42.6–64.5 μg/mL). The most active AIM-C fraction was repeatedly chromatographed to yield four known compounds, methyl trans-p-coumarate (1), methyl caffeate (2), syringic acid (3) and pinellic acid (4). Cell viability assay of 14 against U87MG cells indicated 2 as most active (IC50 value of 8.2 μg/mL), whereas the other three compounds were much less active. Interestingly, compounds 14 were non-toxic towards normal human cells (HEK-293). The content of 2 in AIM-C was estimated as 3% by HPLC. Hence, presence of some more active substances besides methyl caffeate (2) in AIM-C is anticipated.  相似文献

报道了用毛细管气相色谱和色-质谱联用方法分离鉴定紫花野菊和小红菊的挥发油化学成分。初步鉴定了以反-丁香烯,β-蒎烯,1,8-桉叶素等为主的二十八种成分,并测定了它们的含量。同时与野菊花进行了比较。结果表明两者与野菊花主要成分有所不同。  相似文献

A preparative high‐speed counter‐current chromatography method for isolation and purification of flavonoid compounds from Oroxylum indicum was successfully established by using ionic liquids as the modifier of the two‐phase solvent system. Two flavonoid compounds including baicalein‐7‐O‐diglucoside and baicalein‐7‐O‐glucoside were purified from the crude extract of O. indicum by using ethyl acetate–water–[C4mim][PF6] (5:5:0.2, v/v) as two‐phase solvent system. 36.4 mg of baicalein‐7‐O‐diglucoside and 60.5 mg of baicalein‐7‐O‐glucoside were obtained from 120 mg of the crude extract. Their purities were 98.7 and 99.1%, respectively, as determined by HPLC area normalization method. The chemical structures of the isolated compounds were identified by 1H‐NMR and 13C‐NMR.  相似文献

以乙醇为溶剂,采用溶剂回流提取方法结合微波、超声波辅助提取野菊花中的蒙花苷,用HPLC法测定。采用正交试验法研究了野菊花中蒙花苷提取和HPLC实验条件,并比较了采用不同的微波和超声波辅助提取方式对蒙花苷提取结果的影响。结果表明,溶剂回流提取最佳条件为:提取温度80℃,回流时间2.0 h,溶剂用量40 mL。应用于安徽等产地野菊花中蒙花苷的提取分析,结果显示,广东和广西产地药材含量明显更高;在相同的实验条件下采用微波辅助提取可使蒙花苷达到较高的提取率,特别是采用微波辐射药材后再进行溶剂回流提取,提取时间可缩短一半。  相似文献

In Southeast Asia, traditional medicine has a longestablished history and plays an important role in the health care system. Various traditional medicinal plants have been used to treat diseases since ancient times and much of this traditional knowledge remains preserved today. Oroxylum indicum (beko plant) is one of the medicinal herb plants that is widely distributed throughout Asia. It is a versatile plant and almost every part of the plant is reported to possess a wide range of pharmacological activities. Many of the important bioactivities of this medicinal plant is related to the most abundant bioactive constituent found in this plant—the baicalein. Nonetheless, there is still no systematic review to report and vindicate the biological activities and therapeutic potential of baicalein extracted from O. indicum to treat human diseases. In this review, we aimed to systematically present in vivo and in vitro studies searched from PubMed, ScienceDirect, Scopus and Google Scholar database up to 31 March 2020 based on keywords “Oroxylum indicum” and “baicalein”. After an initial screening of titles and abstracts, followed by a full-text analysis and validation, 20 articles that fulfilled all the inclusion and exclusion criteria were included in this systematic review. The searched data comprehensively reported the biological activities and therapeutic potential of baicalein originating from the O. indicum plant for anti-cancer, antibacterial, anti-hyperglycemia, neurogenesis, cardioprotective, anti-adipogenesis, anti-inflammatory and wound healing effects. Nonetheless, we noticed that there was a scarcity of evidence on the efficacy of this natural active compound in human clinical studies. In conclusion, this systematic review article provides new insight into O. indicum and its active constituent baicalein as a prospective complementary therapy from the perspective of modern and scientific aspect. We indicate the potential of this natural product to be developed into more conscientious and judicious evidencebased medicine in the future. However, we also recommend more clinical research to confirm the efficacy and safety of baicalein as therapeutic medicine for patients.  相似文献

One new flavonoid, 5,6,7-trimethoxyflavone-8-O-β-d-glucopyranoside (1), along with six known compounds 27, was isolated from Oroxylum indicum. Their structures were determined on the basis of spectral data. The antibacterial activities of compounds 14 were studied. Compounds 1 and 3 showed medium antibacterial activity against Staphylococcus aureus with MIC/MBC at 32–128 μg/ml.  相似文献

在0.1 mol.L-1的NH4Cl-氨水(pH 9.4)溶液中白杨黄酮于-1.22 V(vs.SCE)处产生一个灵敏的极谱波,其二阶导数峰峰电流i″p与白杨黄酮浓度在3.7×10-7~2.6×10-5mol.L-1范围内呈良好的线性关系(r=0.997 2,n=9),检出限为1.0×10-7mol.L-1(S/N=3),9次测定5.0×10-6mol.L-1白杨黄酮的相对标准偏差为1.2%;研究了一些常见离子及有机物对测定的影响。通过循环伏安法等方法证明白杨黄酮在汞电极上为受吸附控制的不可逆过程,并通过试验推导出其电极反应。该方法应用于木蝴蝶种子提取物中白杨黄酮的测定,其回收率在96.7%~103.0%之间。  相似文献
