A new coumarin laser dye 3-(benzothiazol-2-yl)-7-hydroxycoumarin

The electronic absorption, and emission spectra as well as fluorescence quantum yield of 3-(benzothiazol-2-yl)-7-hydroxycoumarin (BTHC) were measured in different solvents and are affected by solvent polarity (Δf). The deprotonation of BTHC by triethylamine is a reversible process. BTHC is relatively photostable, the quantum yield of photodecomposition (φ_c) was found to be 2×10⁻⁴ and 2.7×10⁻⁴ in EtOH and DMF, respectively. The fluorescence lifetimes of BTHC were measured in the absence and in the presence of molecular oxygen and were found to be 2.82 and 2.78 ns, respectively. BTHC acts as good laser dye upon pumping with nitrogen laser (λ_ex=337.1 nm) in ethanol and gives laser emission with maxima at 508 and 522 nm.     

Generalized criterion of solvent polarity as a tool for control of chromatographic process

The generalized (reductive) criterion of solvent polarity was obtained by the method of multiparametric optimization of the Snyder index P", Hildebrand parameter _T , permittivity _r , and solvatochromism parameter E _T (30). Possibilities of employing this criterion for estimation of the elution power of normal and reversed mobile phases used in high-performance liquid chromatography were considered.     

The Importance of Polarity/Polarizability Interaction on the Acidity Behavior of 9,10-Anthraquinone and 9-Anthrone Derivatives in Methanol-Water Mixed Solvents Using Target Factor Analysis and QSPR Approaches

The influence of solvent properties on acidity constants of some newly synthesized 9,10-anthraquinone and 9-anthrone derivatives was studied in methanol-water mixtures in a composition range of 0.57 to 1.0 methanol mole fraction. The model was established by using both multiple linear regression and target factor analysis. Both methods revealed that the solvent polarity/polarizability parameter ^* is a major factor in controlling the acidity behavior of the anthraquinones and anthrones studied in binary methanol-water mixed solvents. A QSPR study was conducted to drive the relationships between the ^* coefficient s and the polarity/polarizability of molecules. Both dipole moment and polarizability were found to have a linear relationship with s. The results confirm that, in the dipolar protic solvents used, the dipole-dipole interaction (for neutral molecules) and the ion-dipole interaction (for ionized molecules) are the major factors controlling the acidity behavior of these compounds.     

X射线照射Hep-2细胞对其培养基吸收光谱的影响

采用分光光度计分别测量了受不同剂量X射线照射后继续培养48h时的人上皮样喉癌细胞株Hep-2的培养基RPMI1640的紫外吸收光谱,分析了该培养基中蛋白质的紫外吸收特性.结果发现:各组培养基的吸收光谱存在明显的差别,新鲜培养基RPMI1640(10%胎牛血清)233nm处的吸收峰在细胞生长过程中位移至235nm,275nm处的吸收峰位移至278nm.反映出各组培养基中芳香族氨基酸中各种氨基酸如色氨酸、酪氨酸、苯丙氨酸的含量的变化,说明癌细胞在生长过程中对这几种氨基酸的改变不是按等量比的.实验还发现细胞培养基的吸收光谱强度与各剂量组细胞数基本成正相关,进而与细胞所受X射线照射的剂量有内在的联系.这些结果将为利用光谱技术研究人喉癌最佳X射线放射剂量奠定基础.     

Coupled LC-GC: The capacity of silica gel (HP)LC columns for retaining fat

Many applications of coupled LC-GC in food analysis require that LC separates large quantities of triglycerides from the components of interest. The capacities of silica gels to retain triglycerides have been determined for n-hexane as mobile phase as well as some eluent mixtures. Conclusions are drawn for practical applications.

Unusual peak defocusing in capillary GC on hexakis(2,6-diO-pentyl-3-O-acetyl)-α-cyclodextrin stationary phase

An unusual peak defocusing effect influencing chromatographic performance over a limited range of elution temperatures is described for hexakis(2,6-di-O-pentyl-3-O-acetyl)-α-cyclodextrin stationary phase. Since this phenomenon is likely to be dependent on minor details of the cyclodextrin molecule, full assignment of the ¹H- and ¹³C-NMR-spectra are given.     

Rapid preparative isolation of erythrocentaurin from Enicostemma littorale by medium‐pressure liquid chromatography,its estimation by high‐pressure thin‐layer chromatography,and its α‐amylase inhibitory activity

Erythrocentaurin is a relatively simple natural product present among the members of Gentianaceae. A preparative method for the isolation of erythrocentaurin from the ethyl acetate fraction of Enicostemma littorale using medium‐pressure liquid chromatography has been reported. The method consisted of a simple step gradient from 10 to 20% ethyl acetate in n‐hexane. Using a 70 × 460 mm Si60 column, this method is capable of processing 20 g of material in <3 h (purity ≈ 97%). The recovery of erythrocentaurin was 87.77%. Estimation of erythrocentaurin in extracts and fractions based on high‐pressure thin‐layer chromatography was carried out on silica gel 60 F₂₅₄ plates with toluene/ethyl acetate/formic acid (80:18:2 v/v/v) as the mobile phase. The densitometric analysis was performed at 230 nm. A well‐separated compact band of erythrocentaurin appeared at R_f 0.54 ± 0.04. The analytical method showed good linearity in the concentration range of 200–1500 ng/band with a correlation coefficient of 0.99417. The limits of detection and quantification were found to be ≈60 and ≈180 ng/band, respectively. Erythrocentaurin exhibited a concentration‐dependent α‐amylase inhibition (IC₅₀ 1.67 ± 0.28 mg/mL). The outcome of the study should be considered for pharmacokinetic and biotransformation studies involving E. littorale.     

高内相乳液法制备整体柱及其分离性能研究

建立了高内相乳液(HIPE)法制备以甲基丙烯酸十二烷酯为功能单体(LMA),二乙烯基苯(DVB)为交联剂的新型毛细管电色谱整体柱。polyHIPE整体柱利用扫描电镜与BET进行了表征,表明了制备的整体柱具有较大的孔径与较好的比表面积,更好的通透性,可更好地满足高效快速分离分析的需要。实验中K2S2O8不仅可以作为引发剂,同时加热后残基能为固定相提供电渗流。优化表面活性剂span80含量、W/O比例对整体柱制备的影响,以硫脲与苯系物为目标化合物考察了整体柱的分离性能与效果。该方法用实际水样品苯系物分析,取得良好的效果。
