Capillary sieving electrophoresis utilizing SDS (CE(SDS)) is one of the most applied methods for the analysis of antibody (mAb) size heterogeneity in the biopharmaceutical industry. Inadequate peak identification of observed protein fragments is still a major issue. In a recent publication, we introduced an electrophoretic 2D system, enabling online mass spectrometric detection of generic CE(SDS) separated peaks and identification of several mAb fragments. However, an improvement regarding system stability and handling of the approach was desired. Here, we introduce a novel 8-port valve in conjunction with an optimized decomplexation strategy. The valve contains four sample loops with increased distances between the separation dimensions. Thus, successively coinjection of solvent and cationic surfactant without any additional detector in the second dimension is enabled, simplifying the decomplexation strategy. Removal efficiency was optimized by testing different volumes of solvents as presample and cationic surfactant as postsample zone. 2D measurements of the light and heavy chain of the reduced NIST mAb with the 8-port valve and the optimized decomplexation strategy demonstrates the increased robustness of the system. The presented novel set-up is a step toward routine application of CE(SDS)-CZE-MS for impurity characterization of proteins in the biopharmaceutical field.  相似文献

为了进一步探讨非还原端饱和结构的肝素寡糖在UV 232 nm的吸收情况, 制备了4种饱和结构的肝素二糖, 并用离子对反相液相色谱/离子阱飞行时间质谱(RPIP-LC/MS-IT-TOF)光电二极管阵列检测器分析了它们在UV 232 nm的吸收情况. 分析结果表明, 饱和结构的肝素二糖在UV 232 nm的检出限为9 μg(S/N=10), UV 232 nm/UV 206 nm约为不饱和结构肝素二糖UV 232 nm的7%~40%. 结果还表明, 肝素二糖UV 232 nm的吸收强度受亚硫酸基团(SO32?)影响较大. 另外, 通过比较不饱和结构的肝素/硫酸类肝素(Hep/HS)标样二糖发现, 含N-未取代葡萄糖胺(GlcNH3+)基团的二糖在UV 232 nm的吸收值较低. 最后, 通过简单的UV检测方法, 结合 HNO2(pH=4.0)裂解法和RPIP-LC/MS-IT-TOF分析, 简化了含GlcNH3+肝素六糖的测序方法. 本研究为以后用 HNO2(pH=1.5)裂解法对混合组分N-硫酸化的肝素寡糖结构序列分析提供了可能.  相似文献

Compound annotation using MS/MS data is the major bottleneck in interpretation of mass spectrometry data during non-targeted screening and suspect screening exposomics studies. Apart from compound identification using available databases or mass spectral libraries, the true challenge comes when completely new compounds have to be identified. Along with recent advances in MS instrumentation that set grounds to a new revolutionary age in environmental exposomics, a multitude of cheminformatics annotation approaches has been developed. Herein, we review the basic principles of the cutting-edge cheminformatics MS-based approaches employed in eco-exposome annotation.We give a solid background discussing the eco-exposome concept in relation to the advances in MS instrumentation, and define the three crucial cheminformatics tasks used in the eco-exposome annotation: molecular formula assignment, compound prioritization and compound annotation. The basic principles of compound annotation are discussed, which are based on three approaches of utilizing structural information inherent to MS data. These involve direct, indirect and joint annotation approaches. We assess their performance through the ability to annotate eco-exposome constituents. We discuss future perspectives and give directions to new annotation strategies and performance evaluation protocols aiming to solve current issues hampering the incorporation of cheminformatics annotation approaches in regular eco-exposome annotation workflows.  相似文献

The analysis of volatile compounds, and particularly of certain hydrocarbons, e.g. pentane, and some aldehydes, e.g. hexanal, has been used to estimate the degree of oxidation of vegetable oil. We have tested a dynamic headspace technique without heating the sample during the purge time of the analysis. Some tests have been performed after heating the sample at 180°C for a specified time before the analysis, to compare the results of analysis with and without heating. The aim of this study was ultrasound assisted removal of Ceftriaxone sodium (CS) based on CCD model. Using sonochemical synthesized Bi2WO6 implanted on graphitic carbon nitride/Multiwall carbon nanotube (g-C3N4/MWCNT/Bi2WO6). For this purpose g-C3N4/MWCNT/Bi2WO6 was synthesized and characterized using diverse approaches including XRD, FE-SEM, XPS, EDS, HRTEM, FT-IR. Then, the contribution of conventional variables including pH, CS concentration, adsorbent dosage and ultrasound contact time were studied by central composite design (CCD) under response surface methodology (RSM). ANOVA was employed to the variable factors, and the most desirable operational conditions mass provided. Drug adsorption yield of 98.85% obtained under these defined conditions. Through conducting five experiments, the proper prediction of the optimum point were examined. The respective results showed that RSD% was lower than 5% while the t-test confirmed the high quality of fitting. Langmuir isotherm equation fits the experimental data best and the removal followed pseudo-second order kinetics. The estimation of the experimentally obtained maximum adsorption capacities was 19.57 mg.g of g-C3N4/MWCNT/Bi2WO6 for CS. Boundary layer diffusion explained the mechanism of removal via intraparticle diffusion.  相似文献

Here we present a simple yet effective gas chromatography-mass spectrometry (GC-MS) identification approach for the detection of heteroatom-containing compounds (HACCs) in petroleum fractions. The MS/AMDIS (Automated Mass Spectral Deconvolution and Identification System) program was used to identify parts per million (ppm) HACC concentrations in petroleum fractions in place of traditional techniques (extraction and standard injection). Polycyclic aromatic sulfur heterocycles (S-PAHs) were used as model compounds to confirm the validity of the AMDIS identifiers, which were compared with extracted results using the off-line X-calibur software. AMDIS was able to identify ppm concentrations of S-PAHs in oil condensate. There was good agreement between experimental and AMDIS identification results for S-PAHs in oil condensate. AMDIS was also used to detect nitrogen-containing compounds (NCCs) and alkylphenols in oil condensate. Our results confirmed the presence of 2-methylbenzothiazole, carbazole, and 2,4-ditertbutyl phenol. In a crude oil sample, AMDIS identification of m/z=191 biomarkers was consistent with empirical results. Therefore, AMDIS can help to reduce the number of experimental steps in identification protocols.  相似文献

Peak broadening in ion mobility (IM) is a relatively predictable process and abnormally broad peaks can be indicative of the presence of unresolved species. Here, we introduce a new ion mobility peak fitting (IM_FIT) software package for automated and systematic determination of traveling wave ion mobility (TWIM) unresolved species. To identify IM unresolved species, the IM_FIT software generates a trend line by plotting ions' mobility peak widths as a function of their arrival times. Utilizing user-defined thresholds, IM_FIT allows for automated and rapid detection of ions that deviate from the peak width trend line. To demonstrate the advantages of IM_FIT for automated detection of IM unresolved species, IM-mass spectrometry (IM-MS) data from a sample mixture containing polypropylene glycol and multiple peptides were analyzed. A total of 14 out of the 34 observed singly-charged IM peaks above 5% relative abundance (i.e., signal-to-noise ratios above ∼200) were tagged as potentially co-eluting ions by IM_FIT. Subsequently, the 14 IM peaks tagged as potentially unresolved (presumably, peaks corresponding to co-eluting compounds), were further analyzed by automated IM deconvolution (AIMD), liquid chromatography-IM-MS (LC-IM-MS), and/or ultra-high resolution mass spectrometry. Using the aforementioned techniques, more than 85% of the tagged IM peaks (12 out of 14) were confirmed to contain co-eluting ions. As an additional new finding, IM_FIT facilitated the discovery of an unexpected sequence-scrambled y-type fragment ion.  相似文献

This study was undertaken to establish one of the most reliable sampling methods and to precisely evaluate the bias involved in the collection of airborne pollutant samples. For the purpose of our study, we investigated the performance of three different types of sampling techniques by measuring major aromatic volatile organic compounds (VOC) in outdoor air; the target analytes specifically include benzene, toluene, ethylbenzene, and xylene (commonly called BTEX). As the first step of our approach, we designed and developed a multi-channel sampling system consisting of a six-port mass flow controller (SJU-MFC) system. Because this system allowed the collection of up to six replicate samples, our measurement results were analyzed and screened statistically for the derivation of high-quality BTEX data. The feasibility of this sampling system was further tested through a comparison with concurrent measurement data sets obtained by two additional, but independent, sampling techniques: (1) automatic continuous sampler (ACS) and (2) on-line GC (O-GC) system. Based on the data sets collected concurrently by three different sampling methods, we attempted to evaluate the compatibility of sampling techniques. Although the results obtained by SJU-MFC system were not statistically different from those of the O-GC system, they were moderately distinguishable from those of ACS. Such patterns were seen consistently, when examined by correlation analysis. The overall results of our study thus generally point out that the compatibility of data sets, when the proper caution is taken, improve significantly among different sampling methodologies.  相似文献

Polycyclic aromatic hydrocarbons (PAHs) are frequently measured in the atmosphere for air quality assessment, in biological tissues for health-effects monitoring, in sediments and mollusks for environmental monitoring, and in foodstuffs for safety reasons. In contemporary analysis of these complex matrices, gas chromatography (GC), rather than liquid chromatography (LC), is often the preferred approach for separation, identification, and quantification of PAHs, largely because GC generally affords greater selectivity, resolution, and sensitivity than LC. This article reviews modern-day GC and state-of-the-art GC techniques used for the determination of PAHs in environmental samples. Standard test methods are discussed. GC separations of PAHs on a variety of capillary columns are examined, and the properties and uses of selected mass spectrometric (MS) techniques are presented. PAH literature on GC with MS techniques, including chemical ionization, ion-trap MS, time-of-flight MS (TOF-MS), and isotope-ratio mass spectrometry (IRMS), is reviewed. Enhancements to GC, for example large-volume injection, thermal desorption, fast GC, and coupling of GC to LC, are also discussed with regard to the determination of PAHs in an effort to demonstrate the vigor and robustness GC continues to achieve in the analytical sciences.  相似文献
