18α(H)-, 18β(H)-oleanane and lupane are angiosperm-derived biomarkers that are used as age indicators for the Late Cretaceous onwards when the first proliferation of angiosperms occurred. In addition, the 18α(H)-/18β(H)-oleanane ratio is employed as a thermal maturity parameter of crude oil. However, evidence has shown that accurate quantification of these compounds has been impeded by inadequate chromatographic separation by traditional one-dimensional gas chromatography. In this study, we present the separation of 18α(H)-, 18β(H)-oleanane and lupane with comprehensive two-dimensional gas chromatography (GC×GC). Furthermore, it was observed that 18β(H)-oleanane elutes earlier than 18α(H)-oleanane in second dimension (polarity) which we attribute to steric hindrance effects. Two GC conditions have been developed in order to achieve baseline separation of the triterpenoids of interest in complex mixtures such as sediment extracts and crude oils.  相似文献

Abstract

Friedelan-3-one (1) and friedelane-3,16-dione (2) isolated from leaves and branches of Maytenus robusta Reissek were subjected to structural modifications via nucleophilic addition to the carbonyl group and Baeyer-Villiger oxidation in order to synthesize potential cytotoxic compounds. The oximes friedelane-3-hydroxyimino (3) and 3-hydroxyiminofriedelan-16-one (4) together with the lactones friedelane-3,4-lactone (5) and 3,4-lactonefriedelan-16-one (6) were characterized by IR and NMR spectroscopic analyses. Compounds 4 and 6 are reported for the first time. Cytotoxic screening via MTT assay in human leukemia cell lines (THP-1 and K562) demonstrated no significant improvement of compounds 3-6 when compared to the starting materials. Only compounds 3 and 5 demonstrated an improvement against K562 cells. However, the same assay on ovarian and breast cancer cell lines (TOV-21G and MDA-MB-231) showed a reduction in the IC50 for compounds 4-6, indicating that ring A modifications may enhance the biological potential.  相似文献

Abstract

A novel lactone-type norcucurbitacin, designated as neocucurbitacin D (1), together with five known cucurbitane triterpenes were isolated from traditional Tibetan medicine “Se Ji Mei Duo”, which is the seed of Herpetospermum pedunculosum (Ser.) C.B. Clarke. The structure of neocucurbitacin D was elucidated by spectroscopic analysis, including 2D NMR and X-ray techniques. Compounds 1–6 were screened for their xanthine oxidase (XOD) inhibitory activity. Compound 1, 2 and 4 exhibited significant XOD inhibition with IC50 values ranging from 10.16 to 18.41?μM. The absolute stereochemistry and XOD inhibitiory activity of lactone-type norcucurbitacins was reported firstly.  相似文献

Abstract

Brazil has a great variety of native plants which could be explored and among them guabiju (Myrcianthes pungens) stands out. Thus, this study consisted of isolating pentacyclic triterpenes, α and β-amyrin from guajibu leaves, and determine the antioxidant activity.The leaves were dried, pulverized and submitted to a dynamic maceration process with ethanol 96° GL, concentrated to obtain crude leaf extract (CLE). CLE was eluted with dichloromethane until total depletion, resulting in a dichloromethane fraction (FRDicl) which was concentrated and analyzed by GC/MS and NMR. The result of the chemical analysis revealed the presence of pentacyclic triterpenes of oleanane (β-amyrin), and ursane (α-amyrin) groups. The β-carotene bleaching method revealed a high antioxidant activity for the CLE as well as for FRDicl. The antioxidant protection equivalent to the trolox was of 137 and 129%, respectively at 500 µg/mL. The antioxidant potential of FRDicl can be explained by the presence of α and β-amyrins.  相似文献

Arjunolic acid, the major extractable constituent of the heavy wood ofTerminalia Arjuna, has the potential to be used as a rigid and functional molecular framework for the construction of nanosized supramolecular architectures and nanomaterials. The nanosized triterpenoid, arjunolic acid, showed efficient gelation of various organic solvents at low concentrations. The low molecular mass gelator molecules self-assembled in the solvents to form fibers of nanometer diameters. The analytical performances of three atmospheric-pressure sources, electrospray (ESI), atmospheric-pressure chemical ionization (APCI), and atmospheric-pressure photoionization (APPI), were evaluated for the analysis of pentacyclic triterpenes in liquid chromatography-mass spectrometry (LC-MS). Among these sources, APPI and APCI are particularly well adapted to sensitive analyses of pentacyclic triterpenes by LC-MS. Detection parameters were optimized for both the sources, and the effects of three dopants (toluene, acetone and anisole) on the detection (sensitivity and ion fingerprints in MS spectra) were studied in detail for APPI-MS.The limits of quantification were measured under selected ion monitoring conditions, in the range of 0.005-0.015 mg l(-1) and 0.002-0.84 mg l(-1) in APPI and APCI, respectively, depending on the studied pentacyclic triterpene. Overall, APPI was found more sensitive than APCI in positive ion mode, whereas APCI shows the greatest sensitivity for acidic triterpenes in negative ion mode.Following this study, the developed LC-MS method was used for the characterization of pentacyclic triterpenes in three plant extracts. High amounts of betulinic acid, betulinic aldehyde and betulinic aldehyde acetate were observed in plane bark. The main component of birch bark is betulin and extracts of okoume resin exhibit high amounts of alpha- and beta-amyrin.  相似文献

Euphorbia heterophylla L. is regarded as a major weed worldwide. Its high aggressiveness in agricultural environment prompted us to investigate the allelopatic activity and chemical constitution of extracts from roots of this plant. Hexane extract showed low phytotoxic activity. Methanol extract at 2.0 mg mL?1 inhibited 100% of germination, root and shoot growth of the indicator plants Sorghum bicolor and Lactuca sativa. β-sitosterol, stigmasterol, and esters of lupeol, germanicol, taraxasterol, pseudotaraxasterol, α-amyrin and β-amyrin were isolated from the hexane extract and their structures elucidated on the basis of MS and 1H, 13C and DEPT-135 NMR data. GC-MS analysis of the derivatized methanol extract allowed for identifying a series of allelopathic organic acids potentially involved in allelopathic interactions of E. heterophylla. This is the first study on the allelopathic activity of extracts and identification of metabolites from roots of E. heterophylla.  相似文献

Several pentacyclic triterpenoid acids including betulinic, oleanolic, and ursolic acids were reported to have health beneficial properties such as antiviral and anti‐inflammatory properties, as well as the capability to inhibit “in vitro” the development of various cancer cell types. For this reason betulinic, oleanolic, and ursolic acids are used as neutraceuticals. For the analysis of the pentacyclic triterpenoid acids in complex plant materials, an improved scheme was developed, involving a qualitative screening using silylation and gas chromatography with mass spectrometry analysis, followed by quantitation using a novel liquid chromatography with tandem mass spectrometry procedure. The use of the two methods provides more reliable information regarding the plant materials with unknown composition. Besides betulinic, oleanolic, and ursolic acids that were analyzed, by this procedure a fourth pentacyclic triterpenoid acid was identified and quantitated that was not previously reported to be present in plants. This acid has been identified as 3β‐3‐hydroxy‐lupa‐18,20(29)‐dien‐28‐oic acid. The newly identified acid has a structure as a derivative of lupane, although lupane with a double bond in the 18‐position was not previously reported as present in plants. The new liquid chromatography with tandem mass spectrometry procedure developed for this study offers a very low limit of quantitation, excellent precision, and robustness. Rosemary was found to contain the largest levels of pentacyclic triterpenoid acids among all the analyzed botanicals.  相似文献
