The changes in structural organization of the major light-harvesting chlorophyll a/b–protein complex of photosystem II (LHC II) at the level of trimeric subcomplexes were studied in spinach plants grown under low light conditions (50 μmol quanta m−2 s−1) and then acclimated to elevated irradiances. By monitoring photochemical quenching of fluorescence yield (qP), photosystem II (PS II) functional status was assessed in leaves of plants acclimated to a range of elevated irradiances. Three separate acclimative irradiances were selected for the experiments, reflecting: limiting light conditions (150 μmol quanta m−2 s−1), near to the inflexion point on the irradiance curve conditions (300 μmol quanta m−2 s−1) and an excessive light, causing a moderate stress in the form of down regulation of PS II (450 μmol quanta m−2 s−1). An immunoblot analysis showed that there was a clear decline in an abundance on chlorophyll basis of Lhcb1-3 apoproteins as an acclimative irradiance increased from 50 to 450 μmol quanta m−2 s−1, with Lhcb1 decreasing to a lesser extent than Lhcb2 and Lhcb3 (only at excessive irradiance). When analyzed by non-denaturing isoelectric focusing BBY membrane fragments (PSII-enriched, stacked thylakoid membranes) isolated from low light-grown plants were resolved into nine fractions, seven of which (labelled 3–9) were established by us previously [Jackowski and Pielucha, J. Photochem. Photobiol. B: Biol. 64 (2001) 45] to be LHC II subcomplexes representing mixed populations of closely similar trimers, comprising permutations of Lhcb1 and Lhcb2 (subcomplexes 3–7) or Lhcb1-3 (subcomplexes 8 and 9). A heterogeneity with regard to accumulation behaviour of LHC II subcomplexes in response to elevated irradiances was revealed. The subcomplexes 5 and 6 were accumulating at similar level, regardless of the light irradiance experienced. Another group consisting of the subcomplexes 3 and 4 (the most basic ones) showed a progressive increase in relative abundance with increasing an irradiance intensity whereas the subcomplexes 7–9 (the most acidic ones) exhibited a progressive decline in their relative abundance during an acclimation of spinach plants to elevated irradiances thus they may collectively represent an elevated irradiance-responsive subunit of LHCII.  相似文献

A novel hybrid system composed of fluorescent core/shell semiconductor quantum dots and the light harvesting complex II (LHCIIb), a membrane protein of higher plants, has been assembled. Experiments with different mutants show that hybrid formation can be mediated by a C-terminal His6 tag attached to the protein as well as by positive charges of the first N-terminal amino acids of LHCIIb. Quenching of the quantum dot fluorescence upon binding of LHCIIb was partially attributed to energy transfer from the quantum dots to LHCIIb.  相似文献

Thermochromatium (Tch.) tepidum是一种中等嗜热的紫色光合细菌, 最佳生长温度为48-50 ℃; 其捕光天线复合物2 (LH2)含有非均一性脱辅基蛋白和类胡萝卜素(Car), 且高分辨率晶体结构未知. 我们通过超快光谱研究了分别采用去垢剂n-dodecyl-β-D-maltoside (DDM)和lauryldimethylamine oxide (LDAO)制备的LH2的激发态动力学, 观测到由细菌叶绿素(BChl)的Qy态介导的B800-to-B850单重态能量传递过程(时间尺度~1.2 ps, 用DDM制备的LH2), 以及由类胡萝卜素S2态介导的Car-to-Car和Car-to-BChl 单重态能量传递过程(~100 fs). 结果表明C=C共轭双键数目(NC=C)为11和12的两类Car共处于同一LH2复合物中; 相对于源自其它菌种、构成组分相对简单的LH2, Tch. tepidum的LH2中B800-B850的相对取向有较大差异. 本工作发现LH2中低含量类胡萝卜素组分anhydrorhodovibrin (NC=C=12)起着高效“能量陷阱”的作用, 可能是一种重要的光保护机制; 基于类胡萝卜素的超快谱带位移现象提出(OH-)spirilloxanthin(NC=C=13)距BChl分子可能比其它类胡萝卜素更近. 这些研究结果有助于进一步理解苛刻自然条件下生长的Tch. tepidum的捕光和光保护机制.  相似文献

采用稳态荧光光谱技术在低温83 K下用波长为436 nm和507 nm的连续光激发对LHCⅡ进行研究,得到两种波长光激发下LHCⅡ的荧光光谱,并采用高斯组分光谱解析的方法,分别解析出四个谱带,结合吸收光谱和发射光谱分析,认为各自其中两个反映了两种光谱特性:Chl a683.6680/681、Chl a694.0690.0和Chl a/b671.4670.0和Chl a683.8680/681,其余两个长波长组分可能是Chl a分子主发射峰的振动副带.另外还将两种波长光激发下得到的荧光光谱特性做了比较,436 nm光激发下LHCⅡ发出的荧光强度要高于507 nm光的激发,这是由于接收436 nm光的Chl a分子数目多于接收507 nm光的类胡萝卜素分子,且436 nm下Chl a的吸收率也大于507 nm下类胡萝卜素的.从峰值上看,436 nm较507 nm光激励下的荧光光谱峰值产生红移,表明在不同波长光激励下,色素分子之间的能量传递途径是不同的.  相似文献

To explore the effect of cell membrane stacking on the function of light-harvesting complex 2 (LH2) in purple nonsulfur photosynthetic bacteria, LH2 from Rhodobacter sphaeroides 2.4.1 (R. sph 2.4.1) was reconstituted into lipid bilayer vesicles (LH2@liposome) and further formed multi-layer structure by electrostatic interaction with poly-l-lysine (LH2@liposome/PLL), which was characterized by cryo-electron microscopy (cryo-EM) and TEM. When embedded in liposomes and additionally in multi-layer liposomes, the absorption band, zero-crossing point of CD signals and fluorescence emission of LH2 B850 excitons were observed to uniformly have 1–2 nm red-shifting. Combining with the corresponding fluorescence quench and the generation of shorter-living fluorescence species, a new excitonic species generated through B850 structural splitting was proposed. By FT-Raman and triplet carotenoid dynamics, the structural mechanism was deduced and discussed. Briefly, all environmental changes, including LH2 aggregating and multi-layer membrane stacking, eventually applied forces on B850 exciton molecules mainly through the hydrogen bonding between the C3-acetyl carbonyl groups of B850 BChls and Tyr44 and 45 residues at C-terminus of α-polypeptides. The effect of multi-layer structure on LH2 could be assigned as a kind of photoprotection.  相似文献

We simulated two‐dimensional electronic spectra (2DES) of the chlorophyll a manifold of light‐harvesting complex II (LHCII) at various temperatures (77, 110, 150, 190, 230, 273, and 293 K) using the hierarchical equations of the motion‐phase matching approach. We confirm the main excitation energy transfer pathways assignments within the chlorophyll a manifold of LHCII measured in a recent work (J. Phys. Chem. B 2019, 123, 6765–6775). The calculated transfer rates are also in general agreement with the measured rates. We also provided theoretical confirmation for the experimental assignments, as uphill and downhill energy transfer processes, of 2D spectral features that were reported in recent experimental reports. These temperature‐dependent features were also ascertained to follow the detailed‐balance principle.  相似文献

The gels and co-gels from glycine (Gly) and glutamic acid (Glu)-based dendrons with either tyrosine (Tyr) or tryptophan (Trp), two of the luminescent amino acid residues in natural proteins, at the focal point were prepared (Tyr-AB4 and Trp-AB4). It was found that such gels, especially the co-gels from Tyr-AB4/Trp-AB4, showed a high efficient energy transfer (ET) and light-harvesting behaviors. Moreover, luminescent gels with tunable emission ranging from blue to green were also observed owing to the cascade intra- and intermolecular ET from dendritic gelators to the guest molecules (PDNS) in the host-guest gel sample (co-gel with PDNS as the guest molecule), which mimicked the natural light-harvesting systems.  相似文献
