在酸性介质中用氧化还原滴定法研究了铈(IV)离子在痕量铱(III)离子催化作用下,于25~40 ℃区间氧化异丁醇(BA)的反应动力学.结果表明反应对铈(IV)离子为一级,对异丁醇的表观反应级数为正分数.准一级速率常数kobs随[H+]及催化剂[Ir(Ⅲ)]增加而增大,随[HSO4-]增加而减小.在氮气保护下,反应不能引发丙烯酰胺聚合,说明在反应中没有自由基产生.提出了催化剂、底物和氧化剂间生成双核加合物的反应机理,通过kobs与HSO4-的依赖关系,找到本反应体系的动力学活性物种是Ce(SO4)2+,并计算出平衡常数、速控步骤的速率常数及相应的活化参数.  相似文献

In the Aspergillus oryzae protease-catalyzed ester hydrolysis, substitution of N-unprotected amino acid esters for the corresponding N-protected amino acid esters resulted in a large enhancement of the hydrolysis rate, while the enantioselectivity was deteriorated strikingly when the substrates employed were the conventional methyl esters. This difficulty was overcome by employing esters bearing a longer alkyl chain such as the isobutyl ester. Utilizing this ester, amino acids carrying an aromatic side chain were resolved with excellent enantioselectivities (E=50 to >200). With amino acids bearing an aliphatic side chain also, good results in terms of the hydrolysis rate and enantioselectivity were obtained by employing such an ester as the isobutyl ester. Moreover, the enantioselectivity proved to be enhanced further by conducting the reaction at low temperature. This procedure was applicable to the case where the enantioselectivity was not high enough even by the use of the isobutyl ester.  相似文献

Although glyphosate (Gly) is one of the most widely used agrochemicals, it is also of the most difficult to measure. Gly, its metabolites, and related compounds cannot be sought within the scope of multi-residue methods. Specific so-called single-residue methods are used instead. Liquid chromatography-mass spectrometry (LC-MS) is currently the most widely used technique for determining Gly and its metabolites. This review addresses the different LC-MS-based methods proposed for the determination of Gly and related species in food and environment matrices. Sample preparation (food and environment), as well as their determination based in novel liquid chromatography/mass spectrometry approaches including different specific stationary phases are presented and the specific analytical challenges, strengths and drawbacks are critically discussed.  相似文献

In recent years, knowledge of the different chemical forms of the elements has gained increasing importance. There has been significant progress in methods that hyphenate chromatographic separations with atomic spectrometry. These hyphenated methods can provide the most complete information on the species distribution and even structure. However, they can be lengthy, relatively costly and difficult to bring to the routine. On the other hand, it is important to remember that chromatographic techniques represent only a minor part of the separation procedures available and, in certain cases, the application of basic chemistry to sample treatments can give quantitative information about specific chemical forms. In this sense, non-chromatographic procedures can provide methods that offer sufficient information on the elemental speciation for a series of situations. Moreover, these non-chromatographic strategies can be less time consuming, more cost effective and available, and present competitive limits of detection. Thus, non-chromatographic speciation analysis continues to be a promising research area and has been applied to the development of several methodologies that facilitate this type of analytical approach. In view of their importance, the present work overviews and discusses different non-chromatographic methods as alternatives for the speciation analysis of clinical, environmental and food samples using atomic spectrometry for detection.  相似文献

采用一种新型紫外、荧光衍生试剂咔唑-9-乙基氯甲酸酯对9种氨基酸(丝氨酸、苏氨酸、甘氨酸、谷氨酸、天冬氨酸、缬氨酸、异亮氨酸、亮氨酸、苯丙氨酸)进行柱前衍生,采用胶束电动色谱模式在14 min内完成分离。分离条件:以pH 9.0的 20 mmol/L硼酸盐-30 mmol/L十二烷基硫酸钠(SDS)溶液(含3%(体积分数)乙腈)为缓冲溶液,柱温25 ℃,分离电压18 kV,紫外检测波长214 nm。该方法的线性范围为0.025~0.25 mmol/L,检出限为2.15~2.46 μmol/L。  相似文献

In this study, a simple analytical method for the determination of γ‐aminobutyric acid, gabapentin, and baclofen by using high‐performance liquid chromatography with fluorescence detection was developed. An amidogen‐reactive fluorescence labeling reagent, 4‐(carbazole‐9‐yl)‐benzyl chloroformate was first used to sensitively label these analytes. The completed labeling of these analytes can be finished rapidly only within 5 min at the room temperature (25°C) to form 4‐(carbazole‐9‐yl)‐benzyl chloroformate labeled fluorescence derivatives. These labeled derivatives expressed strong fluorescence property with the maximum excitation and emission wavelengths of 280 and 380 nm, respectively. The labeled derivatives were analyzed using a reversed‐phase Eclipse SB‐C18 column within 10 min with satisfactory shapes. Excellent linearity (R2 > 0.995) for all analytes was achieved with the limits of detection and the limits of quantitation in the range of 0.25?0.35 and 0.70?1.10 μg/L, respectively. The proposed method was used for the simultaneous determination of γ‐aminobutyric acid and its analogs in human serum with satisfactory recoveries in the range of 94.5–97.5%.  相似文献

A MEKC methodology with UV detection was developed for the enantioselective separation of selenomethionine (SeMet). The use of (+)‐1‐(9‐fluorenyl)ethyl chloroformate (FLEC) as chiral derivatization reagent to form SeMet diastereomers enabled their subsequent separation using ammonium perfluorooctanoate (APFO) as a volatile pseudostationary phase. The effect of APFO concentration and pH, temperature, injection volume, and derivatization conditions (time and FLEC/SeMet ratio) were evaluated in order to select the best separation conditions. A chiral resolution of 4.4 for DL‐SeMet was achieved in less than 6 min using 100 mM APFO at pH 8.5 as electrophoretic buffer. Satisfactory results were obtained in terms of linearity, precision (RSD from 3.4 to 5.1% for migration times and from 1.8 to 4.6% for corrected peak areas), accuracy, and LODs (3.1 × 10?6 M and 3.7 × 10?6 M for d and l enantiomers, respectively). The method was successfully applied to the determination of l ‐SeMet in food supplements.  相似文献

以肌氨酸为内标物,邻苯二甲醛-9-芴甲基氯甲酸酯为柱前衍生剂,用ODS色谱柱在柱温40℃下,采用二元梯度洗脱,DAD检测器在338nm波长处检测,建立了一种利用反相高效液相色谱同时测定小麦籽粒中17种氨基酸的方法。氨基酸浓度在5~800μmol/L范围内,其峰面积与内标物峰面积的比值和氨基酸浓度的线性相关系数均大于0.996;17种氨基酸的加标回收率在97.5%~103.1%范围内。应用本方法对小麦籽粒中氨基酸含量进行测定,取得了较理想的结果。同时,本法还可应用于糙米和玉米等粮食中氨基酸含量的测定。  相似文献

This article reviews the most common, useful methods for the chiral determination of amphetamine (AM) and AM-derived designer drugs in different of matrix, including blood, hair, urine, medicaments or standard solutions, taking into consideration articles published in the past 15 years. We consider chromatographic methods (e.g., gas, liquid, high-performance liquid, and thin layer). We describe several types of chiral derivatization reagent, mobile-phase additive and chiral stationary phase commonly used in the chromatographic methods. Tables summarize basic information about conditions (e.g., type of column and mobile phase), detection mode and reference data for each procedure.  相似文献
