Substance P and hemokinin‐1 were predominantly examined by immunoassays with their limitation to differentiate appropriately between both peptides. The use of liquid chromatography coupled with tandem mass spectrometry is a promising, highly selective alternative. Adsorption processes have been identified in preliminary experiments to play a crucial role in the loss of mass spectrometry intensity of both peptides. Therefore, a design of experiments concept was created to minimize nonspecific peptide adsorption. For this purpose, the most critical influencing parameters—(1) the composition of the injection solvent as well as (2) the most suitable container material—were systematically and concordantly investigated. The addition of modifiers, such as formic acid, dimethyl sulfoxide, and organic solvents, to the injection solvent led to a substantial gain of intensity of substance P and hemokinin‐1 compared to the start gradient as an injection solvent. Furthermore, the systematic investigation underlined the high impact of the container material, demonstrating polypropylene as the most favorable material. A conjoint injection solvent optimum was found to determine both peptides simultaneously by the conduction of a sweet‐spot analysis. The experimental design substantially reduced nonspecific peptide adsorption and enabled the simultaneous and selective determination of endogenous substance P and hemokinin‐1 plasma levels.  相似文献

Reaction optimisation and understanding is fundamental for process development and is achieved using a variety of techniques. This paper explores the use of self-optimisation and experimental design as a tandem approach to reaction optimisation. A Claisen-Schmidt condensation was optimised using a branch and fit minimising algorithm, with the resulting data being used to fit a response surface model. The model was then applied to find new responses for different metrics, highlighting the most important for process development purposes.  相似文献

Crack propagation on the basal planes in zinc was examined by means of in situ fracture testing of pre-cracked single crystals, with specific attention paid to the fracture mechanism. During quasistatic loading, crack propagation occurred in short bursts of dynamic crack extension followed by periods of arrests, the latter accompanied by plastic deformation and blunting of the crack-tip. In situ observations confirmed nucleation and propagation of microcracks on parallel basal planes and plastic deformation and failure of the linking ligaments. Pre-existing twins in the crack path serve as potent crack arrestors. The crystallographic orientation of the crack growth direction on the basal plane was found to influence both the fracture load as well as the deformation at the crack-tip, producing fracture surfaces of noticeably different appearances. Finite element analysis incorporating crystal plasticity was used to identify dominant slip systems and the stress distribution around the crack-tip in plane stress and plane strain. The computational results are helpful in rationalizing the experimental observations including the mechanism of crack propagation, the orientation dependence of crack-tip plasticity and the fracture surface morphology.  相似文献

A new family of p-stage methods for the numerical integration of some scalar equations and systems of ODEs is proposed. These methods can be seen as a generalization of the explicit p-stage Runge–Kutta ones, while providing better order and stability results. We will show in this first part that, at the cost of losing linearity in the formulas, it is possible to obtain explicit A-stable and L-stable methods for the numerical integration of scalar autonomous ODEs. Scalar autonomous ODEs are of very little interest in current applications. However, be begin studying this kind of problems because most of the work can be easily extended to a more general situation. In fact, we will show in a second part (entitled ‘The separated system case'), that it is possible to generalize our methods so that they can be applied to some non-autonomous scalar ODEs and systems. We will obtain linearly implicit L-stable methods which do not require Jacobian evaluations. In both parts, some numerical examples are discussed in order to show the good performance of the new schemes.  相似文献

In this article, we discuss and analyze new conforming virtual element methods (VEMs) for the approximation of semilinear parabolic problems on convex polygonal meshes in two spatial dimension. The spatial discretization is based on polynomial and suitable nonpolynomial functions, and a Euler backward scheme is employed for time discretization. The discrete formulation of both the proposed schemes—semidiscrete and fully discrete (with time discretization) is discussed in detail, and the unique solvability of the resulted schemes is discussed. A priori error estimates for the proposed schemes (semidiscrete and fully discrete) in H1‐ and L2‐norms are derived under the assumption that the source term f is Lipschitz continuous. Some numerical experiments are conducted to illustrate the performance of the proposed scheme and to confirm the theoretical convergence rates.  相似文献
