Model Construction for the A–B–C Ring System of Lysergic Acid via Vilsmeier–Haack‐Type Cyclization of 1H‐Indole‐4‐propanoic Acid Derivatives

Vilsmeier–Haack‐type cyclization of 1H‐indole‐4‐propanoic acid derivatives was examined as model construction for the A–B–C ring system of lysergic acid ( 1 ). Smooth cyclization from the 4 position of 1H‐indole to the 3 position was achieved by Vilsmeier–Haack reaction in the presence of K₂CO₃ in MeCN, and the best substrate was found to be the N,N‐dimethylcarboxamide 9 (Table 1). The modified method can be successfully applied to an α‐amino acid derivative protected with an N‐acetyl function, i.e., to 27 (Table 2); however, loss of optical purity was observed in the cyclization when a chiral substrate (S)‐ 27 was used (Scheme 5). On the other hand, the intramolecular Pummerer reaction of the corresponding sulfoxide 20 afforded an S‐containing tricyclic system 22 , which was formed by a cyclization to the 5 position (Scheme 3).

Synthetic approach to natural products by Claisen rearrangement of glyceraldehyde derivatives

For the purpose of organic syntheses of some corynanthe-type indole alkaloids, sesquiterpenes, and steroids, optically active intermediates cyclopentanone 3-allylalcohol, α-methylene-γ-butylo lactones andtrans-hydrindanone-propionic acid, were synthesized from (R)- and (S)-isopropylideneglyceraldehyde derived fromD-mannitol andLascorbic acid, respectively, utilizing Claisen rearrangement as a key reaction. Actually total syntheses of natural alkaloids, (-)-antirhine, (+)-dihydroantirhine and (-) dihydrocorynantheol were accomplished.     

Two new C18-norditerpenoid alkaloids from Aconitum delavayi

Further phytochemical investigation of the unique C18-norditerpenoid alkaloids from the roots of Aconitum delavayi Franch ledto the isolation of two new norditerpenoid alkaloids,delavaconitine F 1 and delavaconitine G 2.Their structures were determinedfrom spectroscopic evidence.     

Palladium-catalyzed benzylic-like nucleophilic substitution of benzofuran-, benzothiophene- and indole-based substrates by dimethyl malonate anion

The palladium-catalyzed benzylic-like nucleophilic substitution of acetates derived from benzofuran, benzothiophene and indole was investigated. The asymmetric substitution on racemic 1-(2-benzofuryl)ethyl acetate gave disappointing results, but the substitution product was obtained in 98% ee from (S)-1-(2-benzofuryl)ethyl acetate with overall retention of configuration.     

Synthesis of marine guanidine alkaloids and their application as chemical/biological tools

Ptilomycalin A and crambescidins, novel marine guanidine alkaloids, have a unique pentacyclic guanidine structure, and exhibit a considerable array of biological activities. The first method developed for the synthesis of the pentacyclic guanidine core structure involved successive 1,3-dipolar cycloaddition reactions and resulted in the first total synthesis of crambescidin 359. The synthesis of other pentacyclic guanidine derivatives has been based on this methodology and applied as tools for studying biological activities, and as chemical reaction catalysts.     

Synthesis of 2,5-bis{(diethyl-3′-indolyl)methyl}furan and its N,N′-dimetallated complexes (lithium, sodium and potassium): X-ray crystal structures of 2,5-bis{(diethyl-3′-indolyl)methyl}furan and the polymeric tetrahydrofuran adduct of the dilithiated complex

The synthesis of 2,5-bis{(diethyl-3′-indolyl)methyl}furan by the acid catalysed condensation of 2,5-bis(diethylhydroxymethyl)furan with indole is presented. Dilithium, disodium and dipotassium derivatives are prepared by the reaction of the bis(indole) with n-BuLi, NaH and K, respectively, in the presence of various Lewis bases. The X-ray structures of 2,5-bis{(diethyl-3′-indolyl)methyl}furan and the dilithiated derivative (as a polymeric tetrahydrofuran adduct) are reported.     

Two new tryptamine-derived alkaloids from Chimonanthus praecox f. concolor

A new pyrrolidinoindoline-type alkaloid, CPC-1, and a new tryptamine-derived dimeric alkaloid, CPC-2, were isolated from the seeds and rinds of Chimonanthus praecox f. concolor. The structure including the absolute configuration of CPC-1 was determined by chiral total synthesis. CPC-2 has a unique hexahydropyrroloquinoline skeleton.
